Abstract The physicochemical and bacteriological quality of groundwater was assessed to show the impact of the agriculture and human activities in the Triffa Plain located in North-East of Morocco. The current levels of contamination of the groundwater were estimated by analysing electrical conductivity, nitrate, nitrite, ammonia, orthophosphate, and indicators of faecal pollution content. Water samples from 55 locations were collected during two period of time, the wet and the dry season of the year 2016. Result obtained indicated that most samples are highly contaminated. The electrical conductivity varied from 800 to 9 100 μS∙cm−1. Nitrate levels ranged from 25 to 216 mg·dm−3, with 78% of samples exceeding the critical level value set at 50 mg·dm−3. Nitrate concentrations are slightly higher during the wet period in 73% of studied cases. Nitrite rarely exceeded the normal rate fixed by World Health Organization and reached 0.90 mg·dm−3. Ammonia and orthophosphate contents do not exceed these norms. The study revealed a wide contamination of groundwater by microbial agents such as, total coliforms, faecal coliforms and faecal streptococci, with content ranged from 0 to 14 000, 0 to 5 000 and 0 to 5 000 CFU·(100 cm3)−1 respectively, confirming the impact of septic tanks, wastewater discharge into rivers without treatment, and the use of animal waste on the ground water vulnerability. Samplings and measurements were carried out according to the international standard ISO 13395, ISO 11732 and ISO 15681-2 for chemical compounds and ISO 9308-1 and ISO 7899-2 for microbiological numerations.

Abstract Aims Asses the added value of quantitative evaluation of tricuspid regurgitation (TR), the proper cut-off value for severe TR and ‘torrential TR’ based on outcome data. The added value of quantitative evaluation of TR, and the cut-off values associated with increased mortality are unknown. Methods and results In patients with all-cause TR assessed both qualitatively and quantitatively by proximal iso-velocity surface area method, long-term and 1-year outcome analysis was conducted. Thresholds for excess mortality were assessed using spline curves, receiver-operating characteristic curves, and minimum P-value analysis. The study involved 676 patients with all-cause TR (age 73.9 ± 14 years, male 45%, ejection fraction 52.9 ± 14%). Effective regurgitant orifice (ERO) was strongly associated with decreased survival in unadjusted [hazard ratio (HR) 2.38 (1.79–3.01), P < 0.0001 per 0.1 cm2 increment] and adjusted [2.6 (1.25–5.0), P = 0.01] analyses. Quantitative grading was superior to qualitative grading in prediction of outcome (P < 0.01). The optimal cut-off value for the best separation in survival between groups of patients with severe vs. lesser degree of TR was 0.35 cm2 [P < 0.0001, HR =2.0 (1.5–2.7)]. ERO negatively impacted survival, even when including only the subgroup of patients with severe TR [HR 1.5 (1.01–2.3); P = 0.04]. The optimal threshold corresponding for the best separation for survival between groups of patients with severe vs. ‘torrential’ TR was 0.7 cm2 [P = 0.005, HR =2.6 (1.2–5.1)]. Conclusion TR can be severe and even ‘torrential’ and is associated with excess mortality. Quantitative assessment of TR by ERO measurement is a powerful independent predictor of outcome, superior to standard qualitative assessment. The optimal cut-off above which mortality is increased is 0.35 cm2, similar albeit slightly lower than suggested in recent guidelines. Torrential TR >0.7 cm2 is associated with poorer survival compared to patients with severe TR (ERO > 0.4 cm2 and <0.7 cm2).

The presence of Salmonella in food products and emergence of antibiotic resistance are the major challenges facing public health policies. A total of 2749 crocodile meat samples obtained from the Central Veterinary Laboratories in Zimbabwe were screened for Salmonella specieswere collected from three Zimbabwean commercial farms between the year 2012 and 2019 for a retrospective observational study to determine the prevalence and magnitude of antibiotics resistant Salmonella species in crocodile meat. The isolation of Salmonella was in accordance with the ISO 6579:2002 and the antibiotic susceptibility testing was carried out based on Clinical and Laboratory Standard Institute’s recommendations by means of the Kirby-Bauer disk diffusion method. SILAB Database was used to determine the prevalence of Salmonella species. Prevalence was stratified by year and farms. Twenty Salmonella isolates were identified using biochemical tests, and 15 were confirmed by polymerase chain reaction (PCR). Antimicrobial susceptibility profiles of the confirmed Salmonella isolates were examined using 14 antibiotics. The overall prevalence of Salmonella species in crocodile meat samples was 0.5%. The prevalence of Salmonella species ranged from 0.04% to 0.44% in the crocodile meat samples and annual prevalence ranged from 0.01% to 1%. The highest prevalence of Salmonella (4.4%) was recorded in the year 2012. Salmonella isolates from one of the three tested farms were resistant to Erythromycin (73.33%), Ampicillin (80%), and Penicillin G (100%). Generally, Salmonella isolates displayed lower resistance to Cefepime, Ceftriaxone, Amikacin, Tetracycline, Ertapenem, Florfenicol, and Erythromycin (0-53.33%) whereas all Salmonella isolates showed susceptibility to Cefepime, Ceftriaxone, Ertapenem, and Florfenicol. Although the study indicates low prevalence of Salmonella species in crocodile meat, there is a need for strict implementation of Hazard Analysis Critical Control Point (HACCP) to reduce contamination rates in meat and its products

Facilitating the uptake of established methodologies for risk-based decision-making in product conformity assessment taking into account measurement uncertainty by providing dedicated software is the aim of the European project EMPIR CASoft(2018–2020), involving the National Measurement Institutes from France, Sweden and the UK, and industrial partner Trescal (FR) as primary supporter. The freely available software helps end-users perform the required risk calculations in accordance with current practice and regulations and extends that current practice to include bivariate cases. The software is also aimed at supporting testing and calibration laboratories in the application of the latest version of the ISO/IEC 17025:2017 standard, which requires that“…the laboratory shall document the decision rule employed, taking into account the level of risk […] associated with the decision rule and apply the decision rule.” Initial experiences following launch of the new software in Spring 2020 are reported.

To guarantee of the security of ceramic medical products the identification and quantification of the current degradation products is required by the ISO 10993-14 standard. Two methods - the gravimetrical registration of the mass loss and the quantitative determination of the leached ions by spectrometric methods - are planned. When checking the application of the ISO standard to calcium phosphate ceramics that can be used as implant materials a considerable difference in the results obtained with both methods was observed. It seems to be that precipitation reactions are responsible for that situation. To clear up the influence of precipitation reactions on the results the working procedure of the ISO standard was slightly modified. A first interpretation of the results is tried.

SUMMARY Objective To evaluate polymerization shrinkage and depth of cure of two bulk fill flowable composites, one nanohybrid composite modified to a flowable consistency, and one standard flowable composite, comparing the scraping method to the Knoop hardness test. Methods Two bulk fill flowable composites, SureFil SDR flow (SSF) (Dentsply) and Venus Bulk Fill (VBF) (Heraeus Kulzer), one standard flowable, Filtek Supreme Ultra Flowable (FSUF) (3M/ESPE) (control), and one regular bulk composite that can be made flowable, SonicFill (SF) (Kerr), were used in this study. For polymerization shrinkage (PS), ten 2-mm samples were made for each composite and cured for 20 seconds and shrinkage was measured with a Kaman linometer. For hardness, ten specimens of each composite were made in a 10 × 10-mm mold and cured for 20 seconds; the bottom surface was scraped according to ISO 4049 specification, and the remaining thickness was measured with a micrometer. Hardness samples were prepared at 2-, 3-, 4-, and 5-mm thick ×14-mm diameter, cured for 20 seconds, and polished. After 24 hours of dry storage, a Knoop indenter was applied at 100 g load for 11 seconds. Three readings were made on the top and bottom of each specimen and averaged for each surface to calculate a Knoop hardness value and a bottom/top hardness ratio. One-way analysis of variance and Tukey tests were used to determine significant differences between thicknesses and between test methods for each material. Results PS values were 3.43 ± 0.51%, 3.57 ± 0.63%, 4.4 ± 0.79%, and 1.76 ± 0.53% for FSUF, SSF, VBF, and SF, respectively. VBF showed significantly greater shrinkage (4.4 ± 0.79%), followed by FSUF (3.43 ± 0.51%) and SSF (3.57 ± 0.63%), which were similar, and SF (1.76 ± 0.53%), which had significantly less shrinkage (p<0.05). Values for the scraping method for depth of cure were significantly greater for SSF and VBF (>5.0 mm), followed by SF (3.46 ± 0.16 mm) and FSU (2.98 ± 0.22 mm). Knoop top hardness values (KHN) were: VBF 21.55 ± 2.39, FSUF 44.62 ± 1.93, SSF 29.17 ± 0.76, and SF 72.56 ± 2.4 at 2 mm and were not significantly different at 3-, 4-, and 5-mm thick within each material. Ratios for bottom/top values (depth of cure) for 2, 3, 4, and 5 mm were: VBF 0.80 ± 0.1, 0.78 ± 0.03, 0.67 ± 0.10, and 0.59 ± 0.07, respectively; SSF 0.74 ± 0.08, 0.72 ± 0.08, 0.69 ± 0.18, and 0.62 ± 0.08, respectively; SF 0.82 ± 0.05, 0.68 ± 0.05, 0.47 ± 0.04, and 0.21 ± 0.02, respectively; and FSUF 0.56 ± 0.08 at 2 mm and 0.40 ± 0.08 at 3 mm. The bottom/top ratio was .80 or less at all depths and decreased below 0.70 at 4-mm depth for VBF and SSF, at 3 mm for SF and at 2 mm for FSUF.

Clove (Syzygium aromaticum) is an aromatic bud has antioxidant, antifungal, antiviral, antimicrobial, antidiabetic, anti-inflammatory, anti-thrombotic, anesthetic, analgesic and insect repellent effects. Propolis is a sticky resinous material with same properties like clove. Therefore, the aim of this study was to produce probiotic yoghurts added propolis (0.03%) and clove in different concentrations (0.1%, 0.3% and 1.0%). Five different probiotic yoghurts were produced, and analyzed for their microbiological, chemical and sensory properties. Microbiological analyzes of probiotic yoghurts were carried out according to IDF and ISO standards. Their chemical and sensory analysis were performed according to Turkish Standards Data were analyzed by SPSS. Propolis showed antibacterial effect except S. thermophilus while clove improve the development of bacteria especially Bifidobacterium. The titratable acidity of all groups was increased while their pH value was decreased. Propolis showed antibacterial effect except S. thermophilus while clove improved the development of bacteria especially Bifidobacterium. Clove and propolis together changed the microbiological, chemical and sensory properties of probiotic yoghurts while probiotic yoghurt with propolis got the lowest score according to sensory properties.

The information system of X Government Institution would be assessed based on 14 clauses contained in ISO/IEC 27001: 2013. The method used is qualitative method, data collection and data validation with triangulation technique (interview, observation, and documentation). Data analysis used gap analysis and to measure the maturity level of this research used CMMI (Capability Maturity Model for Integration). The result of the research showed that information security which had been applied by X Government Institution was at level 1 (Initial) which meant there was evidence that the institution was aware of problems that needed to be overcome, unstandardized process, and tended to handle the problem individually or by case.

Background: Muscle mass is an important determinant of metabolic health and physical function. It has previously been demonstrated that the postprandial rise in circulating essential amino acids (EAA) acts as the main stimulus for muscle protein synthesis (MPS). This study investigated postprandial plasma amino acid (AA) responses of 2 different forms of whey protein isolate (WPI) with iso-caloric and iso-nitrogenous profiles to investigate plasma concentrations of EAA. Methods: In all, 12 healthy men (n = 12) between 19 and 32 years of age were recruited for a randomized, cross-over design, which involved consumption of protein supplements on 2 testing days separated by a 6-day washout period between conditions. On each testing day, subjects consumed either 29.6 g of WPI or WPI + io (whey protein isolate plus Ingredient Optimized Protein®) mixed with 236 mL of water. Plasma EAA and branch chain amino acid (BCAA) concentrations were assessed from whole body donated by subjects at pre-consumption and 30, 60, 90, 120, and 180 minutes post consumption. Results: Plasma levels of total EAA concentration was significantly greater in WPI + io at 30, 60, 90, and 120 minutes post consumption ( P < .01, P < .001, P < .01, and P < .01, respectively). Plasma levels of total BCAA concentration was significantly greater in WPI + io at 30, 60, 90, and 120 minutes post consumption ( P < .01, P < .001, P < .01, and P < .05, respectively) compared with WPI. For leucine, only WPI + io had elevated levels compared with pre-test at 90 minutes post consumption ( P < .001). Discussion: Both conditions significantly elevated EAA, BCAA, and leucine from basal levels. However, we conclude that the consumption of the treated WPI significantly raises plasma EAA, BCAA, and leucine to a greater extent compared with WPI with no treatment. Thus, supplementation with WPI that has undergone Ingredient Optimized® technology may be highly beneficial for those who partake in regular exercise, elderly individuals, or those affected by a reduced sensitivity to amino acids.

Abstract The situation on the market of laboratory services in Poland is growing rapidly. In order to stay in this difficult market, one shall convince potential customers to his/her competences and quality of services. Testing or calibration laboratories, wishing to assert its position in the market, introduce the management system based on the PN-EN ISO/IEC 17025:2005 standard, the so-called ‘General requirements for the competence of testing and calibration laboratories’, and then apply for the certificate of accreditation in the Polish Centre for Accreditation (PCA) [14]. In order to maintain accreditation, as well as to maintain a competitive advantage, laboratories must prove that the management system is effective, which is the matter of this article. The first part presents the analysis of concepts on the effectiveness of the quality management systems and their improvement, in particular those set out in the standards of ISO 9000 series, as well as in the ISO/IEC 17025 standard (in the past and present). In the following part one identified and described the PN-EN ISO/IEC 17025:2005 standard on the effectiveness of the management system in the accredited laboratory (including the involvement of top management and quality policy, internal audits, corrective and preventive actions, evaluation of the effectiveness of training, quality control tests). Then one showed how to assess the effectiveness of the management system, which shall or may be used by the accredited laboratory (i.e. internal audit, self-assessment, management review, surveys and financial appraisal).

The paper presents the characteristics of innovative technology of micro-jet cooling in MIG welding (according to PN-EN ISO 4063:2009). Presented results received at different jet hole geometry and various coolant pressure. This technology allows get, with appropriately selected parameters of cooling increased ferrite AF content in weld metal, up to 14% for standard MIG technology. This structure proves toughness, especially at temperature -40°C.

Purpose: This article characterizes widefield fundus autofluorescence (WF-FAF) patterns in retinoschisis (RS), retinal detachment (RD), and combined retinoschisis-detachment (RS/RD), and to correlate them with spectral-domain optical coherence tomography (SD-OCT) findings. Methods: A retrospective case series of 13 eyes with senile RS, RD, or RS/RD is presented. One eye underwent imaging of 2 areas within the retina, resulting in 14 data points. Independent, masked graders classified pathology on SD-OCT as RS, RD, or RS/RD and graded WF-FAF images for either hypoautofluorescent areas or mixed autofluorescence (AF) (hyper-AF, iso-AF, hyper-AF with hypo-AF, hyper-AF with iso-AF, or hypo-AF with iso-AF). Results: There was no statistically significant correlation between the autofluorescence pattern and the type of retinal abnormality ( P = .74). Conclusions: High variability was found in the characterization of WF-FAF in patients with RS and RD. SD-OCT remains the criterion-standard imaging modality in distinguishing RS from RD in clinically ambiguous cases.

Purpose. Tosubstantiate scientifically and improve the technology of producing table caviar from ovulated sterlet eggs, which ensures the preservation of natural biologically active compounds and the stability of quality parameters. To investigate table caviar for microbiological, toxicological and toxico-biological safety. To determine the prospect of using the data in the practice of caviar sturgeon breeding. Methodology. Primary materials were obtained in the conditions of «Oasis Bisan» JSC. The results obtained were processed by organoleptic, biochemical, microbiological, toxicological, toxic-biological methods, as well as by the method of variation statistics using MS Excel with an emphasis on standard errors. Findings. Application of the heat treatment mode (78°C) of ovulated sterlet eggs while reducing the total time of the technological process to 1.0 minute contributes to the preservation of the integrity of food product components. Sterlet eggs from recirculated aquaculture system differed from a similar product from fish from natural waters by a lower mineral content (by 1.2%), an increased content of dry matter (by 8.1%), protein (by 7.5%) and lipids (by 3.7%). This fact indicates a decrease in the water content of the product and the acquisition of optimal gastronomic qualities. Sterlet caviar proteins were found to contain a full set of essential amino acids, the chemical score of which exceeded 100%; no limiting amino acids were found. Comparative analysis of sterlet caviar from recirculated aquaculture systems and from natural habitats showed the absence of significant differences in the amino acid composition, which demonstrated the biological value of the product. The use of the mode of short-term high-temperature heat treatment allows ensuring sufficient safety of the food product. The absence of E. coli and yeast in caviar was found. Pb, Cd, As and Hg contents in sterlet caviar were significantly lower than the maximum permissible levels. The total value of hexachloran and isomers was 0,0027mg/kg; DDT and metabolites – 0,016 mg/kg, polychlorinated biphenyls – 0,017 mg/kg and corresponded to permissible limits. The absence of altered forms, growth inhibition or death of tetrachimens confirms the toxicological safety of the finished product. The shelf life of the finished product without preservative is 6 months at a storage temperature of minus 2°C – minus 4°C, which parametrically meets the requirements of TU U 10.2-37758242-002: 2018. Originality. The technology of processing ovulated sterlet caviar has been scientifically substantiated and experimentally tested. New data on the comparison of the results of classical pasteurization and short-term high-temperature heat treatment of caviar are presented. For the first time, the amino acid composition of sterlet caviar was studied under conditions of recirculated aquaculture systems, and the analysis of grainy caviar for hexachloran and isomers, DDT and metabolites, and polychlorinated biphenyls was carried out. Information on the chemical composition of caviar in the process of technological processing was clarified and supplemented. Practical value. The advantage of the improved technology of processing raw caviar over the classical method of pasteurization was determined. The expediency of using an alternative technology in order to save energy and preserve traditional organoleptic properties, nutritional value and safety of the finished product were experimentally confirmed. On the basis of the results of the studies carried out, the technical documentation of TU «Technological instruction for the production of granular sturgeon caviar» and «Working instruction for the selection of caviar» were developed and agreed in accordance with the established procedure with the bodies and institutions of the State Sanitary and Epidemiological Surveillance and the State Standard. The developed technology was tested in the development of pilot batches of food sterlet caviar, which made it possible at the «Oasis Bisan» enterprise to introduce a food safety management system and conduct a certification audit by the International certification body TUV SUD for compliance with the International standard ISO 22000: 2005. Commission, the «Oasis Bisan» enterprise received an EU registration number: a-UA-14-20-121-VIII-PP, in connection with which it has the right to export products (sturgeon caviar) to the countries of the European Union. Androgen hormones and the androgen receptor (AR) pathway are the main targets of anti-hormonal therapies for prostate cancer. However, resistance inevitably develops to treatments aimed at the AR pathway resulting in androgen-independent or hormone-refractory prostate cancer (HRPC). Therefore, there is a significant unmet need for new, non-androgen anti-hormonal strategies for the management of prostate cancer. We demonstrate that a relaxin hormone receptor antagonist, AT-001, an analog of human H2 relaxin, represents a first-in-class anti-hormonal candidate treatment designed to significantly curtail the growth of androgen-independent human prostate tumor xenografts. Chemically synthesized AT-001, administered subcutaneously, suppressed PC3 xenograft growth by up to 60%. AT-001 also synergized with docetaxel, standard first-line chemotherapy for HRPC, to suppress tumor growth by more than 98% in PC3 xenografts via a mechanism involving the downregulation of hypoxia-inducible factor 1 alpha and the hypoxia-induced response. Our data support developing AT-001 for clinical use as an anti-relaxin hormonal therapy for advanced prostate cancer.

Glycaemic response to dietary carbohydrates might have an impact on cognitive performance. The present study investigated the effects of growing-up milks (GUM) with isomaltulose and extra minerals and vitamins or lower protein content on cognitive parameters in children aged 5–6 years. In a blinded, partly randomised, controlled, cross-over study, four GUM were provided, each taken over 14 d (2 × 200 ml/d): standard (Std) GUM; Std GUM+5 g isomaltulose (Iso-5 GUM); Iso-5 GUM with 26 % less protein (Iso-5 LP GUM); Std GUM with 2·5 g isomaltulose and extra Mg, Zn, Se, D3, B1, B2, B12, folic acid and choline (Iso-2·5 GUM). At test days, when GUM replaced breakfast, repeated (0, 60, 120 and 180 min post-dose) cognitive tasks were performed (picture presentation, simple reaction time, digit vigilance, choice reaction time, spatial and numeric working memory and picture recognition). Task performance of all subjects (n 50) worsened over the morning. Best performance was seen on isomaltulose GUM, most notably at 180 min. Iso-2·5 GUM showed best performance on several parameters of attention and memory, Iso-5 GUM performed best on parameters of memory and Iso-5 LP GUM was positively associated with parameters of attention but less with memory. Std GUM showed only a benefit on one attention and one memory task. Thus, isomaltulose-enriched GUM positively affected parameters of attention and memory at 180 min post-dose when compared with Std GUM. Extra minerals and vitamins seem beneficial, whereas lowering protein content might improve attention in particular.

World trends in this area are already directed to the production and promotion of so-called "healthy food" which, designated in several ways, is becoming more and more present on the tables of developed countries. Yugoslavia, beside its current economic difficulties, has great potential and a elastic chance to follow world trends and completely satisfy the EU standards EVRO-EMA and world ecological standards ISO 14 000 by the quality implementation of well planned ecologically and economically rational programs. n order to undertake appropriate measures for "sustainable development", it s previously necessary to assess the problem and objectively establish the status.

Background: Cyclocreatine phosphate (CCrP) is a potent bioenergetic cardioprotective compound known to preserve high levels of cellular adenosine triphosphate during ischemia. Using the standard Isoproterenol (ISO) rat model of heart failure (HF), we recently demonstrated that the administration of CCrP prevented the development of HF by markedly reducing cardiac remodeling (fibrosis and collagen deposition) and maintaining normal ejection fraction and heart weight, as well as physical activity. The novel inflammatory mediator, Nourin is a 3-KDa formyl peptide rapidly released by ischemic myocardium and is associated with post-ischemic cardiac inflammation. We reported that the Nourin-associated miR-137 (marker of cell damage) and miR-106b-5p (marker of inflammation) are significantly upregulated in unstable angina patients and patients with acute myocardial infarction, but not in healthy subjects. Objectives: To test the hypothesis that Nourin-associated miR-137 and miR-106b-5p are upregulated in ISO-induced “HF rats” and that the administration of CCrP prevents myocardial injury (MI) and reduces Nourin gene expression in “non-HF rats”. Methods: 25 male Wistar rats (180–220 g) were used: ISO/saline (n = 6), ISO/CCrP (0.8 g/kg/day) (n = 5), control/saline (n = 5), and control/CCrP (0.8 g/kg/day) (n = 4). In a limited study, CCrP at a lower dose of 0.4 g/kg/day (n = 3) and a higher dose of 1.2 g/kg/day (n = 2) were also tested. The Rats were injected SC with ISO for two consecutive days at doses of 85 and 170 mg/kg/day, respectively, then allowed to survive for an additional two weeks. CCrP and saline were injected IP (1 mL) 24 h and 1 h before first ISO administration, then daily for two weeks. Serum CK-MB (U/L) was measured 24 h after the second ISO injection to confirm myocardial injury. After 14 days, gene expression levels of miR-137 and miR-106b-5p were measured in serum samples using quantitative real-time PCR (qPCR). Results: While high levels of CK-MB were detected after 24 h in the ISO/saline rats indicative of MI, the ISO/CCrP rats showed normal CK-MB levels, supporting prevention of MI by CCrP. After 14 days, gene expression profiles showed significant upregulation of miR-137 and miR-106b-5p by 8.6-fold and 8.7-fold increase, respectively, in the ISO/saline rats, “HF rats,” compared to the control/saline group. On the contrary, CCrP treatment at 0.8 g/kg/day markedly reduced gene expression of miR-137 by 75% and of miR-106b-5p by 44% in the ISO/CCrP rats, “non-HF rats,” compared to the ISO/Saline rats, “HF rats.” Additionally, healthy rats treated with CCrP for 14 days showed no toxicity in heart, liver, and renal function. Conclusions: Results suggest a role of Nourin-associated miR-137 and miR-106b-5p in the pathogenesis of HF and that CCrP treatment prevented ischemic injury in “non-HF rats” and significantly reduced Nourin gene expression levels in a dose–response manner. The Nourin gene-based mRNAs may, therefore, potentially be used as monitoring markers of drug therapy response in HF, and CCrP—as a novel preventive therapy of HF due to ischemia.

AbstractThe effects of adrenergic challenge (in order to evaluate the adipose tissue lipolytic potential) and time of challenge relative to feeding on the response curve of plasma non-esterified fatty acids (NEFA) in four underfed or overfed non-lactating non-pregnant cows were studied. Basal NEFA and NEFA response to isoproterenol (ISO; 4 nmol/kg body weight) were higher when challenged before than after feeding and higher in underfed than overfed cows. Interaction between feeding level and time of challenge was significant (P < 0·001) for the rising part of NEFA response. Pooled NEFA response curves from several trials (no. = 63 challenges) were analysed in order to obtain a simplified procedure for the prediction of the NEFA response. High correlations were found between the response area or maximal NEFA response and NEFA response at 15 min (r = 0·95 and 0·98, respectively). This simplified procedure was applied on pooled data from 84 challenges in order to evaluate the effects of energy balance, physiological status, body condition score (BCS) and time of challenge relative to feeding. Energy balance had a significant effect on basal plasma NEFA and NEFA response at 15 min after ISO challenge (–7·6 and –14·2 μmol/l when the daily energy balance increased by 1 MJ, respectively). Time of ISO injection relative to feeding had a greater effect on stimulated NEFA than on basal NEFA (308 v. 239 μmol/l). There was a significant effect of BCS (41 μmol/l per unit of BCS) on the basal plasma NEFA level. The NEFA response at 15 min after ISO challenge was lower (387 μmol/l) in lactating cows than in dry cows. The NEFA response to ISO at 15 min could provide an efficient method of studying the adipose tissue lipolytic potential of cattle in vivo.

Abstract. Historically, as far as wet-snow loads were concerned, overhead line design was often based on experience or long-term applications with positive results. New standard like CENELEC EN 50341-1 (2012) take into account characteristic loads, i.e. 50-year return-period loads, for the overhead line design. This article proposes a method to estimate characteristic wet-snow loads based on meteorological data recorded at weather stations. The model used to calculate those loads is mainly inspired by a recent article written by Nygaard et al. (2013a) in which a new parameterization is proposed for the classical cylindrical wet-snow accretion model, as described in ISO 12494 standard (2001), annex C. After a complete description of the model and its parameterization adapted to French wet-snow events, the statistical issues are examined. Then, the model is used with the meteorological data of 87 weather stations in order to calculate wet-snow loads whose relevance has been positively tested according to real damages recorded in a complete wet-snow event database. At last, the characteristic loads of those 87 stations have been determined according to all the loads generated by the model and processed by a peak-over-threshold (POT) method. A practical method to determine the 90% confidence intervals of those characteristic values is given. As it is demonstrated that there is only one value of the ice class (IC) masses proposed by ISO 12494 in each confidence interval, characteristic loads can be easily expressed in terms of ICs. That method shows that ICs ranging from R1 (0.5 kg m−1) to R5 (5 kg m−1) could be used for overhead line design in France.

Abstract Platelets (PLTs) are essential for effectively treating severe hemorrhage. While widely considered standard-of-care, storage at room temperature (RT) is limited to 5 days due to bacterial growth risk, loss of function, and increased metabolism. Cold storage (4oC, 4C) slows bacterial growth and better preserves PLT function but also causes increased PLT activation. Storing PLTs in the cold in 65% PLT additive solution (PAS)/35% plasma has been shown to reduce activation and preserve PLT function (Getz, et al. 2016). In the US, only 2 apheresis (AP) and PAS systems are FDA-approved: Trima and Isoplate-PAS (ISO-PAS; Terumo) and Amicus and Intersol-PAS (INT-PAS; Fenwal). Differences in collection processes and PAS composition warrant distinct regulation. The goal of this study was to perform a comprehensive analysis of long-term 4C-stored PLTs collected via FDA-approved AP/PAS methods. AP PLTs were collected (n=4-5) in 65% ISO-PAS using a Trima (ISO) or in 65% INT-PAS using an Amicus system (INT). All bags were stored for 15 days at 4C without agitation and assayed on Day 1 (baseline, BL), 5, 10, and 15 of storage. PLT count and mean platelet volume (MPV) were obtained using a Coulter counter. Lactate and pH were obtained with i-STAT. All PLT samples were adjusted to a count of 3x105/μl using pooled fresh frozen plasma prior to functional testing. Chrono-Log aggregation was performed with ADP (5 μM), collagen (2.5 μg/ml), TRAP (20 μM), and epinephrine (EPI; 2 μM). Clot strength was evaluated using rotational thromboelastometry (ROTEM). Flow cytometry was used to assess PLT marker abundance (% positive) for CD62P (P-selectin), lactadherin (surface phosphatidylserine [PS]), and CD42b (GPIb). An Amnis ImageStream cytometer was used to visualize PLT mitochondrial depolarization and CD41a expression. Data were reported as means±SEM and analyzed with 1-way ANOVA and student's t-tests for multiple comparisons; significance at p<0.05. pH (Fig. 1A) was only higher in INT on Day 1 compared to ISO. More lactate production was seen in INT compared to ISO (Fig. 1B). PLT count (Fig. 1C) was significantly higher at all timepoints compared to ISO, no significant differences were observed after 15 days. ISO showed a slight advantage in terms of functional preservation (Fig. 1 D-G). Aggregation response to collagen (Fig. 1E) was preserved in ISO until Day 10, whereas INT showed a significant decrease as early as Day 5 (Day 5: 45±7% vs Day 1: 98±4%; p<.001). Though aggregation response to ADP was similar in both ISO and INT samples (Fig. 1D), increased spontaneous aggregation (Fig. 1F) was exhibited in INT by Day 10 compared to both BL-INT and Day 10-ISO (Day 10-INT: 13±2% vs BL-INT: 0%; p<.001; Day 10-INT vs Day 10-ISO: 4±%; p=0.02). Responses to TRAP and EPI were comparable for ISO and INT. While a significant change in MCF was observed at Day 15 in ISO compared to BL (Day 15: 45.5±1.6, BL: 55±1, p=.002), INT showed a significant decrease in function by Day 10 (Day 10: 45.1±1.3 vs BL: 57.1±0.9; p<.001). PS expression (Fig. 1H) increased at Day 10 in both ISO and INT compared to their respective BL levels. Producing a hemostatically effective PLT product with an extended shelf-life is of utmost importance due to recent epidemics of viruses such as Chikungunya and Zika and the concomitant increased risk of transfusion-transmitted disease. Storing PLTs at 4C in PAS offers preservation of PLT function for up to 15 days, potentially tripling the current shelf-life of PLT products. We have shown that PLTs collected with Trima in ISO offer better in vitro functional preservation over storage than PLTs collected with Amicus in INT. Implementation of 4C-PAS PLTs may allow for shipment of PLT products to epidemic disease areas and may enhance blood product support for trauma by increasing local inventories and PLT availability in remote locations. Further study and refinement of 4C-stored ISO and INT collection and storage techniques may result in additional improvements in PLT shelf-life and function. The development of a new pavement roughness index, which is compatible to the current International Standards Organization (ISO) standard on “exposure to whole-body vibration” is described. The index was intended to be the independent variable in the future development of relationships between user cost (i.e., vehicle depreciation, repairs, discomfort and so on) and pavement roughness; hence it was named RIDE (Roughness Index for Driving Expenditure). RIDE is based on the sprung mass acceleration response of a reference vehicle to the pavement profile. It is calculated in the frequency domain by multiplying the power spectral density (PSD) of the pavement profile by the square of the transfer function of the sprung mass acceleration of the reference vehicle. The resulting sprung mass acceleration PSD is integrated over frequency to yield the root-mean-square of the sprung mass acceleration per unit length of pavement traveled. The sprung mass acceleration is shown to be the main contributor of dynamic axle loads in heavy trucks, which relate to vehicle and cargo damage and also to pavement damage.

For the two kinds of commonly used codes in low frequency read-only RFID tag: Manchester encoding and Differential Bi-Phase encoding, we designed the corresponding encoding circuit, as shown in Fig. 5 and Fig. 10. It was fabricated by SMIC 0.35um CMOS process through full custom design method and has been successfully applied to a read-only RFID tag based on ISO/IEC 11784/5 standard. The chip testing result shows that the designed circuit can achieve ideal encoding effect, as shown in Fig. 14 and Fig. 15, which possesses the properties of high accuracy and strong anti-disturbance.

<p style="text-align: justify;">El estudio se basó en determinar el grado de limpieza de un fluido, por medio de un innovador dispositivo on-line de tipo óptico utilizado en la ingeniería mecánica y automotriz para el contaje y clasificación de partículas contaminantes en aceites lubricantes, este dispositivo cumple con la norma de limpieza ISO 4406 determinando códigos que involucran el tamaño (&lt;4, &lt;6, &lt;14) um y cantidad de partículas contaminantes por ml. en la muestra analizada. El ensayo estaba sólo recomendado para “sistemas hidráulicos u otros sistemas limpios como turbinas”, ya que los niveles de desgaste eran bajos, el sensor on-line demostró ser una herramienta de diagnóstico eficiente en donde el código ISO 4406 mostrará un valor alto en sus componentes numéricos si el contenido de partículas es elevado y será muy bajo si el fluido está limpio, para garantizar estos resultados se ha calibrado el equipo basándose en la norma ISO 11171 que recomienda un fluido de calibración con requerimientos SRM 2806 (Standard Reference Material) certificado por NIST (Instituto Nacional de Estándares y Tecnología). Se concluye que, monitorizar el tamaño y cantidad de partículas contaminantes en aceites lubricantes de un sistema, permite la detección de estados iniciales de fallos.</p>

Abstract. A primary mercury gas standard was developed at Van Swinden Laboratory (VSL) to establish an International System of Units (SI)-traceable reference point for mercury concentrations at emission and background levels in the atmosphere. The majority of mercury concentration measurements are currently made traceable to the empirically determined vapour pressure of mercury. The primary mercury gas standard can be used for the accurate and precise calibration of analytical systems used for measuring mercury concentrations in air. It has been especially developed to support measurements related to ambient air monitoring (1–2 ng m−3), indoor and workplace-related mercury concentration levels according to health standards (from 50 ng m−3 upwards) as well as stationary source emissions (from 1 µg m−3 upwards). The primary mercury gas standard is based on diffusion according to ISO 6154-8. Calibration gas mixtures are obtained by combining calibrated mass flows of nitrogen and air through a generator holding diffusion cells containing elemental mercury. In this paper, we present the results of comparisons between the primary gas standard and mercury calibration methods maintained by NPL (National Physical Laboratory in the United Kingdom), a National Metrology Institute (NMI), and the Jozef Stefan Institute (JSI), a Designated Institute (DI). The calibration methods currently used at NPL and JSI are based on the bell-jar calibration apparatus in combination with the Dumarey equation or a NIST (National Institute of Standards and Technology in the United States) reference material. For the comparisons, mercury was sampled on sorbent traps to obtain transfer standards with levels between 2 and 1000 ng with an expanded uncertainty not exceeding 3 % (k=2). The comparisons performed show that the results for the primary gas standard and the NIST reference material are comparable, whereas a difference of −8 % exists between results traceable to the primary gas standard and the Dumarey equation.

Abstract Legipid®Legionella Fast Detection is a test based on combined magnetic immunocapture and enzyme-immunoassay (CEIA) for the detection of Legionella in water. The test is based on the use of anti-Legionella antibodies immobilized on magnetic microspheres. Target microorganism is preconcentrated by filtration. Immunomagnetic analysis is applied on these preconcentrated water samples in a final test portion of 9 mL. The test kit was certified by the AOAC Research Institute as Performance Tested MethodSM (PTM) No. 111101 in a PTM validation which certifies the performance claims of the test method in comparison to the ISO reference method 11731-1998 and the revision 11731-2004 “Water Quality: Detection and Enumeration of Legionella pneumophila” in potable water, industrial water, and waste water. The modification of this test kit has been approved. The modification includes increasing the target analyte from L. pneumophila to Legionella species and adding an optical reader to the test method. In this study, 71 strains of Legionella spp. other than L. pneumophila were tested to determine its reactivity with the kit based on CEIA. All the strains of Legionella spp. tested by the CEIA test were confirmed positive by reference standard method ISO 11731. This test (PTM 111101) has been modified to include a final optical reading. A methods comparison study was conducted to demonstrate the equivalence of this modification to the reference culture method. Two water matrixes were analyzed. Results show no statistically detectable difference between the test method and the reference culture method for the enumeration of Legionella spp. The relative level of detection was 93 CFU/volume examined (LOD50). For optical reading, the LOD was 40 CFU/volume examined and the LOQ was 60 CFU/volume examined. Results showed that the test Legipid Legionella Fast Detection is equivalent to the reference culture method for the enumeration of Legionella spp.

Abstract Hairy cell leukemia variant (HCLv) presents with high disease burden, lack of typical antigens like CD25, and poor response to standard treatments like cladribine. Occasionally, patients with classic HCL respond poorly. Clinical and molecular features of HCL and HCLv has not been compared. Rearrangements expressing immunoglobulin VH chain were sequenced, including 22 from 20 patients with HCLv and 63 from 62 patients with classic HCL. Most patients were seeking relapsed/refractory trials, representing a poor-prognosis population. VH4-34, a gene commonly used in autoimmune disorders, was observed in 8 (40%) HCLv and 6 (10%) classic (P = .004) HCL patients. Compared with 71 VH4-34− rearrangements, 14 VH4-34+ rearrangements were more frequently (P < .001) unmutated, defined as greater than 98% homologous to germline sequence. VH4-34+ patients had greater white blood cell counts at diagnosis (P = .002), lower response rate (P < .001) and progression-free survival (P = .007) after initial cladribine, and shorter overall survival from diagnosis (P < .001). Response and survival were more closely related to VH4-34 status than to whether or not patients had HCLv. VH4-34+ HCL is an important disorder that only partly overlaps with the previously described HCLv. Response to initial single-agent cladribine therapy is suboptimal; these patients should be considered for alternative approaches, including antibody-related therapy.

Safety end points: missed acute coronary syndrome (ACS) and cumulative radiation exposure during the index visit and follow-up period.Results: 659 (66%) patients self-identified as Caucasian and 281 (28%) self-identified as AA. AA were younger and more often female compared with Caucasians, had a higher prevalence of hypertension (64% vs 49%, P<.001) and diabetes (23% vs 14%, P<.001) and a lower prevalence of hyperlipidemia (28% vs 51%, P<.001). ACS was more frequent among Caucasians (10% vs 2%, P<.001). Randomization to CCTA resulted in a reduction in median LOS for Caucasians (7.4 vs 24.7 hours, P<.001) and AA (8.9 vs. 26.3, P<.001; P-interac­tion=.88). The possibility of implementing a quality management system in the hotel and restaurant complex of Ukraine based on the William Edwards Deming’s principles of quality management to meet the requirements of ISO 9000 quality standards and Hazard Analysis and Critical Control Points (HACCP) is substantiated in the article. The purpose of the study is to adapt the William Edwards Deming’s principles of quality management to their implementation in the hotel and restaurant business to ensure the stability of services quality in the modern market, the orientation of managers on creation of conditions for the enterprise’s continuous improvement. Standard DSTU ISO 9000-2015 "Quality management systems. Fundamentals and vocabulary" provides seven general principles of quality management, specific to all activities: customer focus, leadership, employee engagement, process approach, improvement, decision-making based on evidence, relationship management. Ensuring these principles makes it possible to manage the quality of enterprises, including the hotel and restaurant industry. The quality management of hotel and restaurant services is considered as a set of interconnected subjects, objects, principles, methods and functions of management, focused on the development and satisfaction of quality requirements and reduction of its costs. Ukraine has a significant potential in the development of hotel and restaurant services. The expansion of the hotel and restaurant business infrastructure is required as well as the establishment of institutions for employees’ training and retraining. The priority is to develop the country’s own regulatory framework that would regulate the quality of services in this area. The quality management system of hotel and restaurant services must guarantee the customer’s requests satisfaction while being served in the hotel and restaurant at all stages and at all levels. Therefore, the issue of development and practical application of the enterprises’ internal standards which determine the general components of the hotel and restaurant services quality is of great importance.

This paper is concerned with the analysis of geometric composites, whose reinforcement was made of plain weave fabric with different geometries. They were determined following the basic parameters of the textiles e.g. crimp length, crimp amplitude, thickness of the woven fabric, dimensions of the cross-sectional tow (tow width, tow height) and crimp angle. The number of fibers in the warp and tow strands and number of layers in the composites were also determined. These composites comprised of the same materials were subjected to a standard tensile test, according to DIN EN ISO 14 129. The mechanical properties of the composite as a whole were determined by tensile test.

It has been shown that only the life cycle assessment allows make an eco-friendly packing choice that is inflicting the minimum harm to environment.

Abstract Background: Inotuzumab ozogamicin (InO), a humanized anti-CD22 antibody-calicheamicin conjugate, produced a superior response compared with standard of care (SOC) for relapsed/refractory (R/R) acute lymphoblastic leukemia (ALL) in an intent-to-treat (ITT) analysis of the first 218 of 326 patients (pts) randomized (ITT218) in the INO-VATE trial (complete remission [CR], including CR with incomplete hematologic recovery [CRi], 80.7% [95% CI, 72-88] vs 29.4% [21-39]; 1-sided P<0.0001; Kantarjian NEJM 2016). In an ITT survival analysis of all 326 pts randomized (ITT326), progression free survival (PFS) was significantly longer with InO vs SOC (HR, 0.45 [97.5% CI, 0.34‒0.61]; 1-sided P<0.0001; median, 5.0 [95% CI, 3.7‒5.6] vs 1.8 [1.5‒2.2] months [mo]). Overall survival (OS) was longer (HR, 0.77 [97.5% CI, 0.58-1.03]; 1-sided P=0.0203; median, 7.7 [95% CI, 6.0‒9.2] vs 6.7 [4.9‒8.3] mo; 2-year OS rate, 23% [95% CI, 16‒30] vs 10% [5‒16]). Here, outcomes in the ITT326 population by salvage 1 (S1) vs salvage 2 (S2) status are presented. Methods: In the INO-VATE trial (NCT01564784), adults with CD22-positive ALL due to receive S1 or S2 treatment were randomized (1:1) to InO (starting dose 1.8 mg/m2/cycle [0.8 mg/m2 on day 1; 0.5 mg/m2 on days 8 and 15 of a 21-28 day cycle for ≤6 cycles]) or SOC (either fludarabine/cytarabine [ara-C]/granulocyte colony-stimulating factor [FLAG], ara-C plus mitoxantrone, or high-dose ara-C). Outcomes included investigator assessed CR/CRi, minimal residual disease (MRD)-negativity in responders (assessed by central flow cytometry [threshold <0.01%]), duration of remission (DoR), OS, and PFS. Data as of March 8, 2016 for the final analysis are presented (long-term safety follow-up is ongoing). Results: Baseline characteristics for the ITT326 were balanced for S1 vs S2 for InO (S1, n=108; S2, n=56) and SOC (S1, n=107; S2, n=55); except more S2 vs S1 pts were Philadelphia chromosome-positive (InO, 20% vs 10%; SOC, 24% vs 14%), had complex cytogenetics (InO, 30% vs 9%; SOC, 18% vs 11%), and had prestudy SCT (InO, 31% vs 11%; SOC, 28% vs 14%). For InO vs SOC, CR/CRi rates were significantly higher for both S1 and S2 pts (both 1-sided P≤0.0002; Table). For S1 vs S2, CR/CRi rates were numerically higher for the InO arm and were similar for the SOC arm; comparable MRD-negativity rates among responders were reported for the InO and SOC arm. None of these S1 vs S2 differences were significant. For S1 vs S2, DoR was longer for the InO and SOC arms; this difference was significant for the SOC arm. For InO vs SOC, PFS was significantly longer for both S1 and S2 pts. For this same comparison, OS was significantly longer for S1, but not S2. For S1 vs S2, PFS was numerically longer in both the InO and SOC arms; this difference was significant in the SOC arm. For S1 vs S2, OS was also numerically longer in the InO arm and was similar in the SOC arm. The most common Grade (Gr) ≥3 adverse events (AEs) in both S1 and S2 pts receiving InO or SOC were cytopenias; Gr ≥3 febrile neutropenia rates were similar for S1 vs S2 for the InO(24% vs 29%) and SOC (56% vs 51%) arms, as were Gr ≥3 infection rates (system-organ class: InO, 28% vs 28%; SOC; 54% vs 57%). For S2 vs S1,Gr ≥3 hepatobiliary AEs rates were higher for the InO (26% vs 12%) arm and were similar for the SOC (9% vs 6%) arm. During InO therapy or during follow-up without intervening SCT, more S2 vs S1 pts had veno-occlusive disease (VOD: 6% [n=3/51] vs 2% [n=2/111]).For InO vs SOC, poststudy SCT rates were significantly higher in both S1 and S2 pts (S1, 51% [56/111] vs 25% [24/95]; S2, 39% [20/51] vs 17% [8/47]; both P≤0.02); VOD rates in pts with poststudy SCT were higher for InO vs SOC (S1, 18% [10/56] vs 4% [1/24]; S2, 35% [7/20] vs 0% [0/8]). More S1 vs S2 pts receiving InO had poststudy SCT (51% [n=56/111] vs 39% [n=20/51]); the VOD rate among pts who receivedpoststudy SCT was greater for S2 (35% [n=7/20]) vs S1 (18% [n=10/56]). Seven of the VOD cases occurring in the InO arm were reversible.Five pts receiving InO had grade 5 VOD (all post-SCT). Conclusions: InO provides clinical benefit regardless of salvage status for pts with R/R ALL, while OS benefit appeared more evident in S1 pts. However, small sample sizes impose limits on interpretation of these results. InO safety profiles were similar across subgroups, except for the incidence of liver-toxicities, including VOD, which may be greater in heavily pretreated pts such as those in later lines of therapy. It is likely to contain different pollutants, including pathogenic microorganisms, which, due to environmental factors, are dispersed in the hospital environment, where they are responsible for causing diseases in vulnerable populations. In addition to being considered a risk to health, nosocomial diseases generate very high treatment costs, ranging from 13 to 15 million pesos per illness during treatment. Objective: To determine the microbiological quality of the air in bathrooms, corridors, reception and storage of one optical clinic in Colombia. Methods: An initial visit to the clinic was carried out for a microbiological sampling of air with the MASS 100 equipment, based on a sampling route designed according to the ISO 14644 technical standard, in addition to the relative humidity and velocity percen¬tage measurements of the wind for two weeks. Bacterial characterization was verified using a Vitek automated system. Results: It was identified that 64% of the sampling sections are above the limit established by the ISO 14644 standard of colony forming units (CFU) for clean areas. This study identified 14 bacterial genera, such as Staphylococcus (26.3%), Pantoea (10.5%), and Sphingomonas (5.3%). Conclusions: Although there is a high number of CFU in the clinic’s air in the present study, most of these genera are saprophytic and opportunistic bacterial agents.

Seven plants in the top rank were selected from the “MANOSROI III” database using the two Thai keywords which meant impotence and sexual tonic. Boesenbergia rotunda (L.) Mansf. extract [EDP1-001(1)] gave the highest PDE inhibition activity of 4.36-fold sildenafil, a standard anti-impotence drug. Plumbago indica Linn. extract [EDP2-001(1)] exhibited the highest NO release stimulation activity of 666.85% which was 1.50-fold acetylcholine, a standard drug. Most selected plant extracts were nontoxic to EA.hy926 cells at 1.0 mg/mL. EDP1-001(1) exhibited the LD50 value of acute oral toxicity in male ICR mice of over 5,000 mg/kg body weight. EDP1-001(1) also indicated the improvement of sexual behaviors in the paroxetine-induced sexual dysfunction male mice with the evaluation of number of courtships (NC), mount frequency (MF), intromission frequency (IF), and ejaculatory frequency (EF) at 87.67±6.17, 121.00±23.50, 36.00±3.21, and 13.67±2.96 which were 2.63-, 1.27-, 0.53-, and 0.62-fold sildenafil-treated mice at day 14 of the treatments, respectively. The present study has not only confirmed the traditional use of Thai plants for the treatment of ED but also indicated the potential and application of the “MANOSROI III” database for Thai plant selection to be developed as ED food supplements.

The issue of developing a uniform language for international communication in the feld of forensic science is considered. The matters of relative maturity of domestic forensic expert terminology and peculiarities of translated terms usage in forensic expertology and forensic practice are discussed. The concept of expertise subject formulated by modern leading scientists is analyzed, the conclusion is made about the subject of forensic examination as a process of establishing facts and circumstances, the purely applied nature of the concept, as well as the similarity to the defnition of expertise in the ISO international standard. Specifc examples are given. The need to translate some foreign standards into Russian and to adjust and implement several of their provisions in domestic legal system and expert practice to standardize forensic and expert activity is emphasized.

Abstract Background High-dose rifampicin may improve outcomes of tuberculous meningitis (TBM). Little safety or pharmacokinetic (PK) data exist on high-dose rifampicin in human immunodeficiency virus (HIV) coinfection, and no cerebrospinal fluid (CSF) PK data exist from Africa. We hypothesized that high-dose rifampicin would increase serum and CSF concentrations without excess toxicity. Methods In this phase II open-label trial, Ugandan adults with suspected TBM were randomized to standard-of-care control (PO-10, rifampicin 10 mg/kg/day), intravenous rifampicin (IV-20, 20 mg/kg/day), or high-dose oral rifampicin (PO-35, 35 mg/kg/day). We performed PK sampling on days 2 and 14. The primary outcomes were total exposure (AUC0–24), maximum concentration (Cmax), CSF concentration, and grade 3–5 adverse events. Results We enrolled 61 adults, 92% were living with HIV, median CD4 count was 50 cells/µL (interquartile range [IQR] 46–56). On day 2, geometric mean plasma AUC0–24hr was 42.9·h mg/L with standard-of-care 10 mg/kg dosing, 249·h mg/L for IV-20 and 327·h mg/L for PO-35 (P &lt; .001). In CSF, standard of care achieved undetectable rifampicin concentration in 56% of participants and geometric mean AUC0–24hr 0.27 mg/L, compared with 1.74 mg/L (95% confidence interval [CI] 1.2–2.5) for IV-20 and 2.17 mg/L (1.6–2.9) for PO-35 regimens (P &lt; .001). Achieving CSF concentrations above rifampicin minimal inhibitory concentration (MIC) occurred in 11% (2/18) of standard-of-care, 93% (14/15) of IV-20, and 95% (18/19) of PO-35 participants. Higher serum and CSF levels were sustained at day 14. Adverse events did not differ by dose (P = .34). Conclusions Current international guidelines result in sub-therapeutic CSF rifampicin concentration for 89% of Ugandan TBM patients. High-dose intravenous and oral rifampicin were safe and respectively resulted in exposures ~6- and ~8-fold higher than standard of care, and CSF levels above the MIC.

The aim of this paper is to evaluate the possibility of using the Impact-echo method for assessment of extremely long period of frost resistance of ceramic tiles. Sets of ceramic tiles of the Ia class to EN 14 411 B standard made by manufacture RACOs have been analyzed. The ceramic tiles under investigation have been subjected to 500 freeze-thaw-cycle based degradation in compliance with the relevant EN ISO 10545-12 standard. To verify the correctness of the Impact-echo method results, additional physical properties of the ceramic tiles under test have been measured. To analyze the specimen surface condition, we also used Olympus LEXT 3100 confocal scanning microscope. It has been proved that the acoustic method Impact-echo is a sensitive indicator of the structure condition and can be applied to the ceramic cladding element frost resistance and service life prediction assessment.

The article discusses the effect of the hybrid-welding process (laser–electric arc MAG Metal Active Gas) on the structure and properties of butt joints (having various thicknesses, i.e., 5 mm and 7 mm) made of steel S960QL. Welding tests were performed in the flat position (PA) and in the horizontal position (PC). Joints made of steel S960QL in the above-presented configuration are present in elements of crane structures (e.g., telescopic crane jibs). The welding tests involved the use of the G Mn4Ni1.5CrMo solid electrode wire and the Ar+18% CO2 shielding gas mixture (M21) (used in the MAG method). Non-destructive visual and radiographic tests did not reveal the presence of any welding imperfections in the joints. The welded joints obtained in the tests represented quality level B in accordance with the requirements of the ISO 12932 standard. Microscopic metallographic tests revealed that the heat-affected zone (HAZ) contained the coarse-grained martensitic structure resulting from the effect of the complex welding thermal cycle on the microstructure of the joints. Destructive tests revealed that the joints were characterised by tensile strength similar to that of the base material. The hybrid welding (laser–MAG) of steel S960QL enabled the obtainment of joints characterised by favourable plastic properties and impact energy exceeding 27 J. The tests revealed the possibility of making hybrid-welded joints satisfying the quality-related requirements specified in the ISO 15614-14 standard.

Objectives.To evaluate the reproducibility and workability of the in vivo test model of the European test standard EN 12791 regarding the effectiveness of surgical hand antiseptics and, as a secondary objective, to evaluate the power of the model to discriminate between the effectiveness of various formulations of surgical hand antiseptics.Design.Prospective, randomized, multicenter study with a Latin square design.Setting.Five laboratories at 2 universities, 2 disinfectant manufacturers, and 1 private testing institution.Participants.Twenty healthy adults in each laboratory.Intervention.Surgical hand antisepsis was performed by scrubbing with chlorhexidine gluconate 4% detergent (CHG) or by rubbing the hands with propan-2-OL (70% by volume; Iso 70) or ethanol 85% (E 85); rubbing the hands and forearms for 3 minutes with propan-1-OL (N 60) was used as the reference disinfection procedure. We deliberately chose to use these antiseptics at the given concentrations because they were intended to cover the range of typical antiseptics submitted for approval according to EN 12791.Methods.In once-weekly tests, the immediate effects of the 4 antiseptics were established according to the method laid down in EN 12791 by assessing the release of skin flora from the fingertips as viable bacteria counts per milliliter of sampling fluids before treatment and viable bacteria counts immediately after treatment, separately for both hands, such that after 4 weeks each volunteer had used every formulation once.Results.The mean log reduction factor (RF) for the release of bacterial skin flora (the log RF was calculated as the log count before treatment minus the log count after treatment) and corresponding standard deviations for the 4 hand antisepsis formulations were as follows: for CHG, 1.1 ± 0.3 colony-forming units (cfu) per milliliter of sampled fluid; for Iso 70, 1.7 ± 0.3 cfu/mL; for E 85, 2.1 ± 0.3 cfu/mL; and for N 60, 2.4 ± 0.4 cfu/mL. The differences between these values proved significant (P<.001) by analysis of variance and in Tukey's “honestly significantly different” (HSD) post hoc test. Although, with regard to their immediate antibacterial activity, the same ranking of these antiseptics was found at all laboratories, the levels of efficacy were significantly different across laboratories (P<.001); no statistical difference was found between left and right hands (P>.01). Relating the log RF values of the other 3 formulations to those of the reference formulation (N 60) abolished differences between laboratories (P = .16); in addition, the interclass correlation coefficient decreased from 9.1% to 4.5%. With 20 volunteers, a minimum difference of 0.47 log between the mean log RFs of the reference formulation and an inferior test formulation will be detected as significant at an α of .05 (1-sided) and a 1 — β value of .8.Conclusion.The test method described in EN 12791 yielded the same conclusion on the effectiveness of the tested formulations in every laboratory and proved, therefore, reproducible and workable.

Abstract Objective Intra-individual variability (IIV) has been associated with cognitive dysfunction and traumatic brain injury (TBI) severity. This study examined IIV in physical medicine and rehabilitation patients grouped by TBI severity and performance validity. Method Participants (N = 213) were 32% female and 90% white with average age and education of 42 (SD = 13) and 14 (SD = 2) years, respectively. Participants completed an outpatient evaluation using a flexible battery of up to 33 common neuropsychological measures. Participants were grouped by TBI severity and validity: mild (32%), moderate (14%), severe (32%), and questionable performance validity (QPV; 22%). QPV was determined on the basis of embedded and freestanding validity measures. Test battery scores were converted to T scores. Cases with four or more scores below 36 T were identified as impaired. Measures of IIV were calculated including overall test battery mean (OTBM), test battery standard deviation (SD), kurtosis, skew, range, and unbiased coefficient of variation (UCV). IIV measures were compared across groups. Results Groups were significantly different in OTBM (p &lt; .001), kurtosis (p &lt; .03), and UCV (p &lt; .001). In valid cases, TBI severity was associated with OTBM (rho = −.31), SD (.19), kurtosis (−.16), and UCV (.33). Impaired and unimpaired cases were significantly different in OTBM (p &lt; .001), SD (p &lt; .001), range (p &lt; .01), and UCV (p &lt; .001). QPV cases were similar to impaired cases on measures of IIV. Conclusions IIV measures have potential clinical utility in characterizing cognitive deficits in TBI and warrant further study.

An Aegilops ventricosa Tausch (2n = 28, DvDvNvNv) subtelocentric chromosome added to wheat (Triticum aestivum L.) in a disomic addition line was found to carry the genes for resistance Yr17, Lr37, Sr38, and Cre5 already transferred onto chromosome 2AS of the wheat line VPM1. Previous works demonstrated that this Ae. ventricosa chromosome is translocated with respect to the standard wheat genome. The present investigations showed that this chromosome pre-existed in Ae. ventricosa and contains only chromatin specific to the N genome. Using biochemical markers and suitable cytogenetic materials including the monoisosomic addition line for the translocated long arm (6NvL–2NvS), its structure was defined as being 6NvSdel.6NvL–2NvS. It consists of a segment of the short arm 2Nv, containing the resistance genes, attached to a group 6 chromosome lacking a distal part of its short arm. The 2 re arrangements could already be present in Aegilops uniaristata Vis. ABSTRACT Background Trainees are responsible for conducting advance care discussions but are often stressed by this role. Objective We developed an instrument to determine whether residents could identify a clinical scenario that necessitated an examination of a patient's goals and preferences as they pertain to clinical care, and subsequently measured their readiness to engage in such discussions. Methods Participants responded verbally to open-ended case presentations and completed survey items. We scored responses according to proximity to idealized answers. Results The sample consisted of 44 internal medicine residents, 12 students, 5 hospitalists, and 3 palliative care attendings, all of whom volunteered for the study and participated in standard interviews. Residents had widely varying scores (range 0–12, maximum score of 15) on the scored open response items. For eliciting values, mean score increased with training, and students, trainees, and attending physicians had mean scores of 3.7, 5.7, and 8.7, respectively (P = .01). For recommending care, mean scores were 3.0, 6.5, and 9.3, respectively (P &lt; .001). Scores were correlated closely with increasing clinical experience and inversely with self-reported stress when conducting a goals-of-care discussion. The Kuder-Richardson Formula 20 reliability for the instrument was 0.52. Interrater reliability for sections about eliciting and recommending care were 0.64 (P &lt; .001) and 0.50 (P &lt; .001), respectively. The 1-week test-retest reliability was 0.91 for open response items and 0.76 for Likert responses. Conclusions A verbally administered instrument can readily and rapidly characterize a trainee's readiness to participate in advance care planning with patients.

Purpose: To examine the intermethods agreement of dual-energy X-ray absorptiometry (DXA) and foot-to-foot bioelectrical impedance analysis (BIA) to assess the percentage of body fat (%BF) in young male athletes using air-displacement plethysmography (ADP) as the reference method. Methods: Standard measurement protocols were carried out in 104 athletes (40 swimmers, 37 footballers, and 27 cyclists, aged 12–14 y). Results: Age-adjusted %BF ADP and %BF BIA were significantly higher in swimmers than footballers. ADP correlates better with DXA than with BIA (r = .84 vs r = .60, P < .001). %BF was lower when measured by DXA and BIA than ADP (P < .001), and the bias was higher when comparing ADP versus BIA than ADP versus DXA. The intraclass correlation coefficients between DXA and ADP showed a good to excellent agreement (r = .67–.79), though it was poor when BIA was compared with ADP (r = .26–.49). The ranges of agreement were wider when comparing BIA with ADP than DXA with ADP. Conclusion: DXA and BIA seem to underestimate %BF in young male athletes compared with ADP. Furthermore, the bias significantly increases with %BF in the BIA measurements. At the individual level, BIA and DXA do not seem to predict %BF precisely compared with ADP in young athletic populations.
