In 2004, Ts. Turbat made a discovery of a significant site of engraved rock art, dating back to the Upper Paleolithic period. Situated on the south bank of the Baga Oigor River, this remarkable site lies within the protective zone of the World Heritage site of the Mongolian Altai rock art complex. The Baga Oigor-VI complex boasts an impressive array of 41 depictions featuring animals such as horses, ibex, argali, deer, cattle, and snakes from the Upper Paleolithic period, complemented by an additional 16 images originating from the Bronze Age, constituting a total of 57 images. Noteworthy is the superior preservation status of this monument, eclipsing that of comparable sites and presenting unparalleled clarity in visibility. The scarcity of open Paleolithic rock art sites across the globe that bear resemblance to this exceptional find underscores its singular nature. The geographically nearest analogous site of significance is the Gobustan monument in the South Caucasus followed by largest concentration of similar monuments in Iberian peninsula. Beyond the renowned ochre paintings of the Khoid Tsenkher Cave, an extensive exploration of carved rock art open air monuments is underway, elevating the Mongolian Altai region to the status of a pivotal hub in the realm of ancient art. The sheer abundance and widespread distribution of these monuments contribute to its characterization as a veritable “cradle of prehistoric art”.

Even given a state-of-the-art masking scheme, masked software implementation of some cryptography functionality can pose significant challenges stemming, e.g., from simultaneous requirements for efficiency and security. In this paper we design an Instruction Set Extension (ISE) to address a specific element of said challenge, namely the elimination of leakage stemming from architectural and microarchitectural overwriting. Conceptually, the ISE allows a leakage-focused behavioural hint to be communicated from software to the micro-architecture: using it informs how computation is realised when applied to masking-specific data, which then offers an opportunity to eliminate associated leakage. We develop prototype, latencyand area-optimised implementations of the ISE design based on the RISC-V Ibex core. Using them, we demonstrate that use of the ISE can close the gap between assumptions about and actual behaviour of a device and thereby deliver an improved security guarantee.

AbstractIn the summer of 1991 excavation was made of the largest barrow of a complex at Kuturguntas in the Bertek hollow of the Altai Highlands. The fairly well-preserved log-roofed construction consisted of an outer burial chamber of blackened roughhewn logs, an inner chamber of worked timber, a sarcophagus made of a single large log, and a storeroom. The coffin and storeroom had been ransacked in antiquity, but surviving grave goods-which include fragments of fur clothing, felt applique, and metal and wooden objects-parallel those found in other Pazyryk graves. Two carved wooden ibex heads had however not been attested among Pazyryk grave goods elsewhere. The wealth of the people buried in such medium-sized barrows lay in the thoroughbred horses buried with them. The burial here of 10 horses had not been disturbed, although only the wooden bridle decorations of the principal 'master's horse' survive well. These again show the popularity of the griffin motif in Pazyryk art. Fragments of three wooden bridle decorations displaying anthropomorphic horned faces probably had a protective function and are linked by the author to the mongoloid Hsiunnu, near neighbours of the Pazyryk people, and to the eclectic receptivity of the Pazyryk people to diverse iconography borrowings from the Middle and Far East.

Background: Bacterial meningitis is a severe infectious disease that affects the central nervous system and can result in high mortality rates and neurological complications. In neonates, meningitis is mainly caused by neonatal meningitis Escherichia coli (NMEC), which invades the intestine, leading to hematogenous spreadand potential bacterial meningitis through blood-brain barrier. Previous studies indicated that the ability of NMEC to adhere and invade is associated with biofilm formation and a deficiency in ibeA may result in decreased virulence and biofilm-forming ability of avian pathogenic Escherichia coli (APEC). Furthermore, the ibeA gene is positioned downstream of ibeR. Our previous studies have shown that ibeR and ibeA genes can both regulate the virulence and relevant phenotypes in E. coli K1. However, the effects of ibeR and ibeA on the biofilm formation and invasion of K1 strain remain to be further studied. Recent studies suggest that indole may act as a signaling molecule to regulate the expression of adhesion and biofilm-promoting factors in E. coli. However, the mechanism by which indole affects biofilms is still unclear. This study aimed to investigate the mechanism of ibeA inhibition of indole-induced NMEC invasion and biofilm formation. Methods: The viability and growth of the wild strain E44 and ibeA-deletion mutant strain ZD1 were assessed by MTT cell proliferation assay kit (Coolaber). The absorbance values were measured at 490 nm and 540 nm by ELISA. The invasion activities and the biofilm formation ability of E44 and ZD1 were detected in vitro studies. We conducted a cell invasion test using the Human Brain Microvascular Endothelial Cells (HBMEC) model. The ability to form a biofilm was measured at 600 nm using a specific biofilm formation index. Biofilm phenotypic was observed by an confocal laser-scanning microscope(Olympus FV-1000, Japan). Then, we used indole as a regulatory agency to compare its biofilm inhibitory efficacy between E44 and ibeR-deletion mutant strain BR2. The experimental data were analyzed using the SPSS 19.0 statistical package. A statistically significant difference was indicated by P<0.05. Results: No difference was found in bacterial growth between the E. coli strains E44 and ZD1. The biofilm formation and HBMECs invasion of ZD1 was significantly lower than that of E44. After treatment with 0.5mM indole, the biofilm formation ability and the ibeA protein expressions of E44 were significantly inhibited. However, indole at different concentrations had no obvious inhibition on BR2 biofilm formation and the ibeA protein expressions. Conclusion: In summary, the study found that ibeA had a significant regulatory effect on HBMECs invasion and biofilm formation of NMEC, which may inhibit by indole via regulating ibeA through ibeR. Background: Myeloid cells are critical determinants of the sustained inflammation in Crohn’s Disease (CD). Targeting such cells may be an effective therapeutic approach for refractory CD patients. Bromodomain and extra-terminal domain protein inhibitors (iBET) are potent anti-inflammatory agents; however, they also possess wide-ranging toxicities. In the current study, we make use of a BET inhibitor containing an esterase sensitive motif (ESM-iBET), which is cleaved by carboxylesterase-1 (CES1), a highly expressed esterase in mononuclear myeloid cells. Methods: We profiled CES1 protein expression in the intestinal biopsies, peripheral blood, and CD fistula tract (fCD) cells of CD patients using mass cytometry. The anti-inflammatory effect of ESM-iBET or its control (iBET) were evaluated in healthy donor CD14+ monocytes and fCD cells, using cytometric beads assay or RNA-sequencing. Results: CES1 was specifically expressed in monocyte, macrophage, and dendritic cell populations in the intestinal tissue, peripheral blood, and fCD cells of CD patients. ESM-iBET inhibited IL1β, IL6, and TNFα secretion from healthy donor CD14+ monocytes and fCD immune cells, with 10- to 26-fold more potency over iBET in isolated CD14+ monocytes. Transcriptomic analysis revealed that ESM-iBET inhibited multiple inflammatory pathways, including TNF, JAK-STAT, NF-kB, NOD2, and AKT signaling, with superior potency over iBET. Conclusions: We demonstrate specific CES1 expression in mononuclear myeloid cell subsets in peripheral blood and inflamed tissues of CD patients. We report that low dose ESM-iBET accumulates in CES1-expressing cells and exerts robust anti-inflammatory effects, which could be beneficial in refractory CD patients.

The massive amounts of data collected by Internet of things (IoT) devices can be stored in clouds to solve the problem of the low storage capacity of IoT terminals. However, the privacy and security of outsourced IoT data may be compromised on the cloud side. Traditional cryptographic technologies can protect data privacy but require the user to retrieve the data for decryption and further processing, which would bring vast amounts of bandwidth and computation burden to users. This paper proposes a dual-server identity-based encryption scheme supporting authorized ciphertext equality test (DS-IBE-AET), where two noncolluding servers with authorizations from users can collaboratively carry out an equality test on outsourced IoT ciphertexts without decrypting the data. DS-IBE-AET can resist offline keyword guessing attacks confronted by existing encryption schemes with equality test in the single server model. Security analysis demonstrates that the proposed DS-IBE-AET scheme offers unforgeability for private keys of users and servers and confidentiality protection for outsourced IoT data and authentication tokens. The performance analysis indicates the practicality of our DS-IBE-AET construction for securing outsourced IoT data in clouds.

Abstract In 2020, Trapholt (a Danish museum of modern art, craft and design) and textile designer Iben Høj launched a grand collaborative art project involving almost 800 embroiderers. The project, named Stitches Beyond Borders, was part of the centenary celebratrions of Denmark’s reunification with Southern Jutland, and participants were asked to embroider their personal vision of borders. By using a mixed method approach we firstly analyse how Stitches Beyond Borders, as a collaborative art project, created a strong sense of community and cultivated creative agency. Secondly, we focus on the discursive nature of the female public created by the art project. Taking into account the rich and complex history of embroidery as an underestimated female activity tied to repressive power mechanisms, we discuss whether the project ends up merely (re)creating an innocuous female public by favouring a personal take on the border theme.

Convergent evolution provides powerful opportunities to investigate the genetic basis of complex traits. The Tibetan antelope (Pantholops hodgsonii) and Siberian ibex (Capra sibirica) belong to different subfamilies in Bovidae, but both have evolved similar superfine cashmere characteristics to meet the cold temperature in plateau environments. The cashmere traits of cashmere goats underwent strong artificial selection, and some traces of domestication also remained in the genome. Hence, we investigated the convergent genomic signatures of cashmere traits between natural and artificial selection. We compared the patterns of convergent molecular evolution between Tibetan antelope and Siberian ibex by testing positively selected genes, rapidly evolving genes and convergent amino acid substitutions. In addition, we analyzed the selected genomic features of cashmere goats under artificial selection using whole-genome resequencing data, and skin transcriptome data of cashmere goats were also used to focus on the genes involved in regulating cashmere traits. We found that molecular convergent events were very rare, but natural and artificial selection genes were convergent enriched in similar functional pathways (e.g., ECM-receptor interaction, focal adhesion, PI3K-Akt signaling pathway) in a variety of gene sets. Type IV collagen family genes (COL4A2, COL4A4, COL4A5, COL6A5, COL6A6) and integrin family genes (ITGA2, ITGA4, ITGA9, ITGB8) may be important candidate genes for cashmere formation and development. Our results provide a comprehensive approach and perspective for exploring cashmere traits and offer a valuable reference for subsequent in-depth research on the molecular mechanisms regulating cashmere development and fineness.

This work adds to the recent debate in corporate social responsibility (CSR) and its effects on performance and firm value. By analysing Spanish companies participating in the IBEX-35 stock-exchange index, this paper empirically tests whether there is a significant price reaction to environmental friendly announcements. Using event studies methodology, the distinction among sectors allows for a better understanding of investors reaction. Results show first, that investors do act in response to this kind of practices and second, that the sign of their reaction depends crucially on the business of the firm and the sector where it operates. In this sense, results may help in reconciling the opposite views regarding the effects of CSR policies.

Treatment of quiescent MG-63 cells with 12-O-tetradecanoylphorbol-13-acetate (TPA) or platelet-derived growth factor (PDGF) stimulates the rapid accumulation of c-myc RNA. We have now determined that a similar effect can be induced by cAMP. Treatment with forskolin (an activator of adenylate cyclase), IBMX (a phosphodiesterase inhibitor), PGE1, and isoproterenol stimulated accumulation of both cAMP and c-myc RNA, but no increase in either cAMP or c-myc RNA was seen with the inactive forskolin analog 1,9-dideoxyforskolin. Forskolin and IBMX acted synergistically in stimulating accumulation of both cAMP and c-myc RNA. However, three lines of evidence indicated that PDGF action is not mediated by cAMP. First, PDGF treatment caused no elevation of cAMP within 1 h, even in the presence of IBMX. Second, the kinetics of c-myc RNA elevation after treatment with PDGF or forskolin were similar, ruling out delayed onset of cAMP stimulation. Finally, simultaneous treatment with forskolin and the calcium ionophore A23187 enhanced the elevation of c-myc RNA levels; no such effect was seen with PDGF. We had previously shown that PDGF action is not affected by prior treatment of MG-63 cells with TPA, a treatment which desensitizes the c-myc response to TPA. Similarly, TPA pretreatment had minimal effect on forskolin or IBMX-induced c-myc expression. These data suggest that cAMP, phorbol esters, and PDGF act independently to stimulate c-myc RNA expression in MG-63 cells. However, nuclear runoff experiments and RNA half-life measurements demonstrated that PDGF, phorbol ester, and cAMP all act to increase the transcription of the MYC gene.

AbstractBone accumulation in Palaeolithic archaeological sites is often the result of activities carried out by hunter-gatherer groups. Cultural choices may have influenced prey representation in archaeological assemblages, distorting their palaeoecological meaning. We present a comparison between large mammal and small mammal assemblages from the Upper Palaeolithic sequence of Grotta Paglicci (Apulia, southern Italy) that extends from the Marginally Backed Bladelet Aurignacian (about 39,000 cal yr BP) to the Final Epigravettian (about 13,000 cal yr BP). At Paglicci, the high frequency of horse and ibex remains indicates open and dry environments for most of the Upper Palaeolithic. This is confirmed by the predominance of the common vole among small mammals. The alternation between horse and ibex, which takes place during the Upper Palaeolithic, however, looks to be more related to variations in hunting territories. Taxon frequencies change abruptly at 17,955–16,696 cal yr BP, with an increase in woodland-related ungulates together with micromammals, indicating a climatic evolution towards milder and more humid conditions. Results demonstrate that when the association of ungulate taxa is considered as a whole, it has a good palaeoecological signal, whilst considering taxa separately can help to better understand cultural choices of past hunter-gatherer communities.

Epoxyeicosatrienoic acids (EETs) are produced from arachidonic acid via the cytochrome P-450 epoxygenase pathway. EETs are able to modulate smooth muscle tone by increasing K+ conductance, hence generating hyperpolarization of the tissues. However, the molecular mechanisms by which EETs induce smooth muscle relaxation are not fully understood. In the present study, the effects of EETs on airway smooth muscle (ASM) were investigated using three electrophysiological techniques. 8,9-EET and 14,15-EET induced concentration-dependent relaxations of the ASM precontracted with a muscarinc agonist (carbamylcholine chloride), and these relaxations were partly inhibited by 10 nM iberiotoxin (IbTX), a specific large-conductance Ca2+-activated K+ (BKCa) channel blocker. Moreover, 3 μM 8,9- or 14,15-EET induced hyperpolarizations of −12 ± 3.5 and −16 ± 3 mV, with EC50 values of 0.13 and 0.14 μM, respectively, which were either reversed or blocked on addition of 10 nM IbTX. These results indicate that BKCa channels are involved in hyperpolarization and participate in the relaxation of ASM. In addition, complementary experiments demonstrated that 8,9- and 14,15-EET activate reconstituted BKCa channels at low free Ca2+ concentrations without affecting their unitary conductance. These increases in channel activity were IbTX sensitive and correlated well with the IbTX-sensitive hyperpolarization and relaxation of ASM. Together these results support the view that, in ASM, the EETs act through an epithelium-derived hyperpolarizing factorlike effect.

The Inuit Bachelor of Education (IBED) program in Labrador is a partnership between the Nunatsiavut Government (NG) and Memorial University of Newfoundland. It is preparing teachers to be key participants in NG’s education system. The IBED students and Sylvia Moore, the lead faculty member in the program, have based this paper on a collaborative presentation. The writers explore the tensions between the current provincial curriculum offered in the regional schools and a curriculum that is founded on Inuit history, culture, and worldview, restores the central role of the Inuit language, and is community-based as recommended in the 2011 National Strategy on Inuit Education. The students discuss four key threads of culturally relevant education: land, language, resources, and local knowledge. Moore reflects on how the IBED program incorporates these same elements to support Inuit identity and the developing pedagogy of the pre-service teachers.

Summaryvon Willebrand factor (vWf) is released from endothelial cell storage granules after stimulation with thrombin, histamine and several other agents that induce an increase in cytosolic free calcium ([Ca2+]i). In vivo, epinephrine and the vasopressin analog DDAVP increase vWf plasma levels, although they are thought not to induce vWf release from endothelial cells in vitro. Since these agents act via a cAMP-dependent pathway in responsive cells, we examined the role of cAMP in vWf secretion from cultured human umbilical vein endothelial cells. vWf release increased by 50% in response to forskolin, which activates adenylate cyclase. The response to forskolin was much stronger when cAMP degradation was blocked with IBMX, an inhibitor of phosphodiesterases (+200%), whereas IBMX alone had no effect. vWf release could also be induced by the cAMP analogs dibutyryl-cAMP (+40%) and 8-bromo-cAMP (+25%); although their effect was weak, they clearly potentiated the response to thrombin. Epinephrine (together with IBMX) caused a small, dose-dependent increase in vWf release, maximal at 10-6 M (+50%), and also potentiated the response to thrombin. This effect is mediated by adenylate cyclase-coupled β-adrenergic receptors, since it is inhibited by propranolol and mimicked by isoproterenol. In contrast to thrombin, neither forskolin nor epinephrine caused an increase in [Ca2+]j as measured by fura-2 fluorescence. In addition, the effects of forskolin and thrombin were additive, suggesting that they act through distinct signaling pathways. We found a close correlation between cellular cAMP content and vWf release after stimulation with epinephrine and forskolin. These results demonstrate that cAMP-dependent signaling events are involved in the control of exocytosis from endothelial cells (an effect not mediated by an increase in [Ca2+]i) and provide an explanation for epinephrine-induced vWf release.

Previous studies have demonstrated enhanced phosphorylation of phospholipase C-tau (PLC-tau), a key regulatory enzyme in phosphoinositide metabolism, in cells treated with platelet-derived growth factor (PDGF) and epidermal growth factor, both of which act via specific receptor tyrosine kinases. Our studies on BALB/c-3T3 cells show that agents that promote cellular cyclic AMP accumulation also increase the phosphorylation, specifically the serine phosphorylation, of this enzyme. Increased phosphorylation of PLC-t (2-3-fold) was evident within 5-10 min of addition of isobutylmethylxanthine (IBMX) and either cholera toxin or forskolin to cells, and persisted for at least 3 h. Treatment of cells with cyclic AMP agonists also enhanced, with similar kinetics, the phosphorylation of a 76 kDa protein co-precipitated by anti-PLC-tau monoclonal antibodies. Brief exposure of cells to cholera toxin/IBMX or forskolin/IBMX decreased inositol phosphate formation induced by the GTP-binding protein (G-protein) activator aluminium fluoride by approx. 50%, but was without effect on PDGF-stimulated inositol phosphate formation. These findings suggest that PLC-tau, and perhaps the 76 kDa co-precipitated protein, are substrates of cyclic AMP-dependent protein kinase in BALB/c-3T3 cells: however, the lack of effect of cyclic AMP elevation on PDGF-stimulated inositol phosphate formation indicates that the intrinsic activity of PLC-tau is unaltered by cyclic AMP-mediated phosphorylation.

A potential natural melanogenesis inhibitor was discovered in the form of a sesquiterpene isolated from the flowers of Inula britannica, specifically 6-O-isobutyrylbritannilactone (IBL). We evaluated the antimelanogenesis effects of IBL on B16F10 melanocytes and zebrafish embryos. As a result, we found that 3-isobutyl-1-methylxanthine (IBMX)-induced melanin production was reduced in a dose-dependent manner in B16F10 cells by IBL. We also analyzed B16F10 cells that were and were not treated with IBMX, investigating the melanin concentration, tyrosinase activity, mRNA levels. We also studied the protein expressions of microphthalmia-associated transcription factor (MITF), tyrosinase, and tyrosinase-related proteins (TRP1, and TRP2). Furthermore, we found that melanin synthesis and tyrosinase expression were also inhibited by IBL through the modulation of the following signaling pathways: ERK, phosphoinositide 3-kinase (PI3K)/AKT, and CREB. In addition, we studied antimelanogenic activity using zebrafish embryos and found that the embryos had significantly reduced pigmentation in the IBL-treated specimens compared to the untreated controls.

Klithi is a rockshelter in the lower reaches of the Voidomatis gorge, near the village of Klithonia in Epirus. Excavations in progress since 1983 have revealed evidence of a late Upper Palaeolithic occupation dated between 16,000 BP and 10,000 BP, with rich microlithic stone tool industries and faunal assemblages dominated by chamois and ibex. The excavations have been accompanied by wider investigations of the local and regional palaeoenvironment and reexamination of the Middle and Upper Palaeolithic sites excavated by Eric Higgs in the 1960s, notably Kokkinopilos, Asprochaliko, and Kastritsa. This paper presents some of the detailed results of the Klithi excavations and sets the results within the wider context of the global issues which inform the study of Palaeolithic archaeology, the Palaeolithic of Greece as a whole, and the regional picture of Palaeolithic settlement in Epirus.

The concept of earnings quality has been widely analyzed after several cases of companies reporting false earnings and experiencing dramatic collapses. Hence, the need for stakeholders to be knowledgeable about the current situation and future prospects of the companies they are involved with. To aid the system, this paper aims to find company-specific financial and corporate governance factors that can act as determinants of the quality of earnings. The researchers took a sample of the companies listed in the Spanish stock market under IBEX 35 over the period 2017–2021. To test the hypotheses, panel, and cross-sectional regressions were run on Stata with the different quality of earnings measured as the dependent variables. The results showed that a company’s earnings age and earnings growth positively impact earnings quality, while its performance and liquidity have a negative impact. Company size can have positive or negative effects based on the chosen quality of earnings measure. Moreover, the different measures of earnings quality reacted differently to independent variables.

The effects of cAMP-elevating compounds IBMX (3-isobutyl-1-methyl­xanthine) and isoproterenol, and that of rutin (an effective superoxide scavenger) were studied on orthovanadate- (a putative protein-phosphotyrosine phosphatase inhibitor) induced nitric oxide (NO) production in J774A.1 mouse macrophage cells. As we previously reported (Koncz and Horváth, 2000), rutin and sodium orthovanadate act synergistically to induce production of high amount of NO in J774A.1 cells. IBMX, an agent that can elevate cAMP level in the cells, can reduce the production of both the LPS- and rutin + orthovanadate-induced NO in macrophages. In contrast, isoproterenol, a non-selective ß-adrenergic receptor agonist, that reduced the LPS-induced NO production in macrophage cells, was unable to reduce the rutin + orthovanadate-induced NO production without negatively affecting cell viability. Moreover, isoproterenol dramatically enhanced the orthovanadate-induced NO synthesis in J774A.1 cells. Our previous study clarified that rutin and orthovanadate, in a specific concentration ratio of both, were able to produce hydrogen peroxide (H2O2). Using 2',7'-dichlorofluorescein-diacetate as a marker for H2O2, isoproterenol alone induced its oxidation but the rutin plus orthovanadate-induced H2O2 production was reduced by isoproterenol. These observations have revealed that, in some cases, H2O2 and superoxide (O2-) scavengers can act in a reverse mode on macrophage cells depending on the presence or absence of orthovanadate.

Across the globe, wireless devices with Internet facilities such as smartphones and tablets have become essential assets for communication and entertainment alike for everyday life for millions of people, which increases the network traffic and the demand for low-latency communication networks. The fourth-generation (4G)/long-term evolution (LTE)/ fifth-generation (5G) communication technology offers higher bandwidth and low latency services, but resource utilization and resiliency cannot be achieved, as transmission control protocol (TCP) is the most common choice for most of the state-of-art applications for the transport layer. An extension of TCP—multipath TCP (MPTCP)—offers higher bandwidth, resiliency, and stable connectivity by offering bandwidth aggregation and smooth handover among multiple paths. However, MPTCP uses multiple disjointed paths for communication to offer multiple benefits. A breach in the security of one of the paths may have a negative effect on the overall performance, fault-tolerance, robustness, and quality of service (QoS). In this paper, the research focuses on how MPTCP options such as MP_CAPABLE, ADD_ADDR, etc., can be used to exploit the vulnerabilities to launch various attacks such as session hijacking, traffic diversion, etc., to compromise the availability, confidentiality, and integrity of the data and network. The probable security solutions for securing MPTCP connections are analyzed, and the secure key exchange model for MPTCP (SKEXMTCP) based on identity-based encryption (IBE) is proposed and implemented. The parameters exchanged during the initial handshake are encrypted using IBE to prevent off-path attacks by removing the requirement for key exchange before communication establishment by allowing the use of arbitrary strings as a public key for encryption. The experiments were performed with IBE and an elliptic curve cryptosystem (ECC), which show that IBE performs better, as it does not need to generate keys while applying encryption. The experimental evaluation of SKEXMTCP in terms of security and performance is carried out and compared with existing solutions.

ABSTRACT The adenylate cyclase stimulator forskolin increases intracellular cyclic AMP (cAMP) in rat FRTL-5 cells within minutes and, after a lag phase of 20–24 h, an increase of cells in metaphase is seen. The dose– response relationships were similar in both systems, with significant increases in the number of metaphases observed at ∼0·1 μmol/l and a doubling of cAMP levels at 1 μmol/l, whilst doses of 0·1 mmol/l and above proved cytotoxic. An involvement of intracellular cAMP as a positive intermediate in cell division was further suggested by the finding that a low dose of forskolin (0·1 μmol/l) potentiated TSH stimulation of mitosis. Isobutyl methyl xanthine (IBMX), a phosphodiesterase inhibitor, also acted as a mitogen and potentiated TSH action. Moreover, the simultaneous inclusion of low doses of IBMX and forskolin additionally potentiated TSH stimulation of mitosis. An analogue of cAMP, dibutyryl cAMP, also stimulated mitosis and acted over a restricted dose range, with maximal stimulation at 1 mmol/l. Calycosin and formononetin are two structurally similar isoflavonoids that have been shown to induce vasodilation in aorta and conduit arteries, but study of their actions on endothelial functions is lacking. Here, we demonstrated that both isoflavonoids relaxed rat mesenteric resistance arteries in a concentration-dependent manner, which was reduced by endothelial disruption and nitric oxide synthase (NOS) inhibition, indicating the involvement of both endothelium and vascular smooth muscle. In addition, the endothelium-dependent vasodilation, but not the endothelium-independent vasodilation, was blocked by BKCa inhibitor iberiotoxin (IbTX). Using human umbilical vein endothelial cells (HUVECs) as a model, we showed calycosin and formononetin induced dose-dependent outwardly rectifying K+ currents using whole cell patch clamp. These currents were blocked by tetraethylammonium chloride (TEACl), charybdotoxin (ChTX), or IbTX, but not apamin. We further demonstrated that both isoflavonoids significantly increased nitric oxide (NO) production and upregulated the activities and expressions of endothelial NOS (eNOS) and neuronal NOS (nNOS). These results suggested that calycosin and formononetin act as endothelial BKCa activators for mediating endothelium-dependent vasodilation through enhancing endothelium hyperpolarization and NO production. Since activation of BKCa plays a role in improving behavioral and cognitive disorders, we suggested that these two isoflavonoids could provide beneficial effects to cognitive disorders through vascular regulation.

Network embedding has shown its effectiveness in many tasks, such as link prediction, node classification, and community detection. Most attributed network embedding methods consider topological features and attribute features to obtain a node embedding but ignore its implicit information behavior features, including information inquiry, interaction, and sharing. These can potentially lead to ineffective performance for downstream applications. In this article, we propose a novel network embedding framework, named information behavior extraction (IBE), that incorporates nodes’ topological features, attribute features, and information behavior features within a joint embedding framework. To design IBE, we use an existing embedding method (e.g., SDNE, CANE, or CENE) to extract a node’s topological features and attribute features into a basic vector. Then, we propose a topic-sensitive network embedding (TNE) model to extract a node’s information behavior features and eventually generate information behavior feature vectors. In our TNE model, we design an importance score rating algorithm (ISR), which considers both effects of the topic-based community of a node and its interaction with adjacent nodes to capture the node’s information behavior features. Eventually, we concatenate a node’s information behavior feature vector with its basic vector to get its ultimate joint embedding vector. Extensive experiments demonstrate that our method achieves significant and consistent improvements compared to several state-of-the-art embedding methods on link prediction.

Patterns of spatial genetic variation can be generated by a variety of ecological processes, including individual preferences based on habitat. These ecological processes act at multiple spatial and temporal scales, generating scale-dependent effects on gene flow. In this study, we focused on bobcats (Lynx rufus), a highly mobile, generalist felid that exhibits ecological and behavioral plasticity, high abundance, and broad connectivity across much of their range. However, bobcats also show genetic differentiation along habitat breaks, a pattern typically observed in cases of isolation-by-ecology (IBE). The IBE observed in bobcats is hypothesized to occur due to habitat-biased dispersal, but it is unknown if this occurs at other habitat breaks across their range or at what spatial scale IBE becomes most apparent. Thus, we used a multiscale approach to examine isolation by ecology (IBE) patterns in bobcats (Lynx rufus) at both fine and broad spatial scales in western Texas. We genotyped 102 individuals at nine microsatellite loci and used partial redundancy analysis (pRDA) to test if a suite of landscape variables influenced genetic variation in bobcats. Bobcats exhibited a latitudinal cline in population structure with a spatial signature of male-biased dispersal, and no clear barriers to gene flow. Our pRDA tests revealed high genetic similarity in similar habitats, and results differed by spatial scale. At the fine spatial scale, herbaceous rangeland was an important influence on gene flow whereas mixed rangeland and agriculture were significant at the broad spatial scale. Taken together, our results suggests that complex interactions between spatial-use behavior and landscape heterogeneity can create non-random gene flow in highly mobile species like bobcats. Furthermore, our results add to the growing body of data highlighting the importance of multiscale study designs when assessing spatial genetic structure.

To date, the underlying mechanisms by which cAMP modulators act during in vitro maturation to improve oocyte developmental competence are poorly understood. Here, we sought to fill this knowledge gap by evaluating the use of phosphodiesterase inhibitor 3-isobutyl-1-methylxanthine (IBMX) and adenylyl cyclase activator forskolin during a culture period of 2 h before in vitro maturation (pre-IVM) on the nuclear and cytoplasmic maturation features in essential organelles, cumulus cells activity, and in vitro developmental potential of sheep oocytes. Results showed that pre-IVM treatment significantly decreased (p < 0.05) the DNA damage of mature oocytes (pre-IVM = 2.08% ± 3.51% vs. control = 20.58% ± 3.51%) and increased (p ≤ 0.05) expanded blastocyst rates compared to the control (from the total of oocytes: pre-IVM = 23.89% ± 1.47% vs. control = 18.22% ± 1.47%, and from the cleaved embryos: pre-IVM = 45.16% ± 1.73% vs. control = 32.88% ± 1.73%). Considering that oocytes are highly vulnerable to the accumulation of DNA damage because of exposure to in vitro culture conditions, our results suggest that the modulation of intra-oocyte cAMP levels with forskolin and IBMX before IVM might afford oocytes a more effective DNA repair mechanism to overcome damage obstacles and ultimately improve developmental competence. This previously unappreciated action of cAMP modulators could help to develop improved methods for assisted reproduction technologies in animal and clinical research.

A digital twin is a computerized representation of the physical object or phenomenon, which acts as its real-time digital equivalent. Artificial intelligence (AI) is becoming increasingly widespread, and museums and art shows will need to change their old working and thinking processes to fully fulfill their potential. In an increasingly digital world, combining AI technologies and wearable devices into digital museums could assist in boosting involvement. It is necessary to investigate AI’s strategic function and application in affecting tourist experiences at art galleries and museums, as well as its potential to improve education. Also, digital twin (DT) paves the way to alleviate difficulties or might improve access to digital museums via digital experiences. The historical report recommends that digital museum items be represented in a way that poses the fewest threats to its legitimacy. Owing to its uniqueness and vulnerability to substantial environmental dangers, the safeguarding of art and cultural heritage in digital museums is a serious issue. There are many approaches for assessing the state of artistic heritage and protecting it from extreme acts. The current study offers a framework for digitized museum art design based on the utilization of digital twin (DT), artificial intelligence (AI), and fifth-generation (5G) technologies. Further to enhance the security of the system, we employ the improved blowfish encryption algorithm (IBEA). Also, the authentication is done using the Diffie-Hellman protocol. The proposed system is simulated using MATLAB, and its performance is analyzed and compared with conventional systems. From the result obtained, it was revealed that the suggested methodology express high range of security level (95%) than the existing mechanisms. This study is very important in two directions: (1) it presents an analytical structure for having to adapt digital information to complicated systems in museums and (2) it explains the artistic obstacles for heritage properties, such as accessibility, execution time, security level, and the efficiency of 3-dimensional designs.

Biofuels are receiving increased scientific attention, and recently different biofuels have been proposed for spark ignition engines. This paper presents the state of art of using biofuels in spark ignition engines (SIE). Different biofuels, mainly ethanol, methanol, i-butanol-n-butanol, and acetone, are blended together in single dual issues and evaluated as renewables for SIE. The biofuels were compared with each other as well as with the fossil fuel in SIE. Future biofuels for SIE are highlighted. A proposed method to reduce automobile emissions and reformulate the emissions into new fuels is presented and discussed. The benefits and weaknesses of using biofuels in SIE are summarized. The study established that ethanol has several benefits as a biofuel for SIE; it enhanced engine performance and decreased pollutant emissions significantly; however, ethanol showed some drawbacks, which cause problems in cold starting conditions and, additionally, the engine may suffer from a vapor lock situation. Methanol also showed improvements in engine emissions/performance similarly to ethanol, but it is poisonous biofuel and it has some sort of incompatibility with engine materials/systems; its being miscible with water is another disadvantage. The lowest engine performance was displayed by n-butanol and i-butanol biofuels, and they also showed the greatest amount of unburned hydrocarbons (UHC) and CO emissions, but the lowest greenhouse effect. Ethanol and methanol introduced the highest engine performance, but they also showed the greatest CO2 emissions. Acetone introduced a moderate engine performance and the best/lowest CO and UHC emissions. Single biofuel blends are also compared with dual ones, and the results showed the benefits of the dual ones. The study concluded that the next generation of biofuels is expected to be dual blended biofuels. Different dual biofuel blends are also compared with each other, and the results showed that the ethanol–methanol (EM) biofuel is superior in comparison with n-butanol–i-butanol (niB) and i-butanol–ethanol (iBE).

In this paper we evaluate the use of a laser scanner for future advanced driver assistance systems. We focus on the important task of predicting the target vehicle for longitudinal ego vehicle control. Our motivation is to decrease the reaction time of existing systems during cut-in maneuvers of other traffic participants. A state-of-the-art laser scanner, the Ibeo Scala B2 R , is presented, providing its sensing characteristics and the subsequent high level object data output. We evaluate the performance of the scanner towards object tracking with the help of a GPS real time kinematics system on a test track. Two designed scenarios show phases with constant distance and velocity as well as dynamic motion of the vehicles. We provide the results for the error in position and velocity of the scanner and furthermore, review our algorithm for target vehicle prediction. Finally we show the potential of the laser scanner with the estimated error, that leads to a decrease of up to 40% in reaction time with best conditions.

We infer that detached binary white dwarfs with orbital periods of a few hours exist because we observe both their progenitors and their descendents. The binary LB 3459 has an orbital period of 6.3 hr and contains a pair of hot subdwarfs that will eventually cool to become white dwarfs (Kilkenny, Hill, and Penfold 1981). L870-2 is a pair of white dwarfs and, given enough time, its 1.55 d orbital period will decay to shorter periods (Saffer, Liebert, and Olszewski 1988). GP Com, AM CVn, V803 Cen, and PG1346+082 are interacting binary white dwarfs with orbital periods between 1051 s for AM CVn and 46.5 min for GP Com (Nather, Robinson, and Stover 1981; Solheim et al. 1984; Wood et al. 1987; O’Donoghue and Kilkenny 1988). These ultrashort period systems must be descendents of detached pairs of white dwarfs. We also expect short-period binary white dwarfs to exist for theoretical reasons. All calculations of the evolution of binary stars show that main-sequence binaries can evolve to binary white dwarfs (e.g., Iben and Tutukov 1984). Among Population I stars, 1/2 to 2/3 of all main-sequence stars are binaries and about 20% of these binaries should become double white dwarfs with short orbital periods (Abt 1983, Iben and Tutukov 1986). Thus, about 1/10 of all white dwarfs could be close binaries (Paczynski 1985). Nevertheless, no detached binary white dwarfs with extremely short periods have yet been found.

The synthesis of endoglucanase by young induced mycelia of Trichoderma reesei QM 9414 incubated in the presence of 1 mM sophorose (a potent cellulase inducer) was stimulated or repressed by additions of dibutyryl cyclic AMP (dBcAMP), depending on the concentration. At low concentrations (10−6 and 10−5 M), dBcAMP stimulated the formation of endoglucanase; higher concentrations of dBcAMP (10−3 and 10−2 M) repressed the synthesis of endoglucanase. Addition of the phosphodiesterase inhibitor 3-isobutyl-1-methylxanthine (IBMX) at 1 and 10 μM concentrations to young induced mycelia caused an increase in intracellular cAMP and stimulated the production of endoglucanase. Neither exogenous dBcAMP nor IBMX was capable of inducing endoglucanase synthesis by itself, and neither was able to relieve catabolite repression of endoglucanase synthesis caused by glucose. All of the monosaccharides tested caused a more or less transient increase in intracellular cAMP. However, the effect of these treatments on endoglucanase synthesis was varied. The phosphorylable hexoses, both metabolizable and nonmetabolizable, increased the intracellular level of glucose-6-phosphate or its analogs and repressed endoglucanase synthesis. Nonphosphorylable sugars, such as 6-deoxyglucose, xylose, L-fucose, and (or) L-sorbose, did not influence the glucose-6-phosphate level and stimulated endoglucanase production to varying degrees. It is concluded that both cAMP and glucose-6-phosphate are involved in regulating cellulase synthesis in T. reesei.
