Artikkeli liittyy kulttuurikasvatuksen toteuttamiseen varhaiskasvatuksessa. Tarkastelemme, kuinka sivistyskasvatus heijastuu varhaiskasvatuksen opettajien erilaisissa tavoissa soveltaa erästä suomalaisiin luontouskomuksiin ja myytteihin liittyvää pedagogista materiaalia. Sivistyskasvatuksella viittaamme J. A. Hollon (1895–1967) ajatuksiin kasvatuksesta. Hollon sivistyskasvatusajattelun ytimessä ovat muun muassa luovuus, mielikuvittelu ja ilo, jotka kiteytyvät älyllisen, eettisen, esteettisen ja toiminnallisen kasvatuksen toteuttamisessa. Kulttuuri, sivistys ja taiteet olivat Hollolle tärkeitä kasvatusta ohjaavia seikkoja; myös tarinoiden merkitys ja voima korostuvat Hollon ajattelussa. Nämä näyttäytyvät tutkimuksessamme tärkeinä varhaiskasvatukseen liittyvän kulttuurikasvatuksen ja pedagogisen kehittämisen taustoittajina. The adaptation of pedagogical material Whisper of the Spirit in early childhood education as a reading of J. A. Hollo's educational thinking Abstract This article is related to implementing cultural education in early childhood education. We consider how early childhood education teachers' various ways of adapting a pedagogical material about Finnish nature beliefs and myths are reflecting the idea of Bildung. We are especially focusing on Juho August Hollo's (1895–1967) educational thinking. Hollo emphasized creativity, imagination and joy, together with intellectual, ethical, aesthetic and experiential education. In his educational thinking, culture, education and arts were influential, as well as the meaning and power of stories. In our research, these are important bases of cultural education and pedagogical development in early childhood education. Kuluneena syksynä tuli kuluneeksi 60 vuotta siitä, kun Urpo Harva nimitettiin maamme ensimmäiseksi kansansivistysopin professoriksi. Eurooppalaisittain Suomi oli edelläkävijä. Sittemmin Yhteiskunnallisesta korkeakoulusta tuli Tampereen yliopisto ja kansansivistysoppi muuttui aikuiskasvatustieteeksi. Tuon viran haltijana oli sittemmin myös Annikki Järvinen 1997–2005. Hän oli samalla ensimmäinen aikuiskasvatuksen professoriksi nimitetty nainen Suomessa. Virka sai kuitenkin alamääritteeksi ammatillisen aikuiskoulutuksen. Julkaisemme professori emerita Järvisen muistumia sekä hänen ”tiedepoliittisen testamenttinsa”. Teksti perustuu hänen jäähyväispuheeseensa Tampereen yliopistossa.

Abstract In this article the etymology of the Modern German word Kobold ‘house spirit’ and its cognates is revised. It is argued that the Germanic root *kub- meaning ‘hut, small chamber’ which consitutes the first element of Modern German Kobold, is a loan from the Latin/Romance group of words deriving from Lat. cubīle, cubīculum. This Romance element may have replaced an earlier PGm. *gub- meaning ‘fire’, attested in Old Norse gufa ‘vapour, steam’, which goes back to the PIE root *ghu̯obh-. This theory is supported by French gobelin where the initial *g- is easily explained from Germanic *g-. The second element of the compound should be identified with the source of Finnish haltija ‘house spirit’ which derives from Gm. *haldija-. The compound was therefore Gm. *gub-haldija- and referred to the house spirit as the keeper of the fire, a concept well-known from Northern European folklore.

Nisotra chrysomeloides Jacoby, 1885, N. dohertyi (Maulik, 1926), N. gemella (Erichson, 1834), and Nisotra nigripes Jacoby, 1894 are redescribed with illustrations of aedeagi, antennae, gonocoxae, abdominal ventrite VIII, and spermathecae. Nisotra nigripes is recorded for the first time from Taiwan. The immature stages and life history of N. gemella were studied in the laboratory using a novel rearing design. Four synonyms previously proposed are confirmed: Sphaeroderma javana de Motschulsky, 1866, S. orbiculata de Motschulsky, 1866, Nisotra bowringi Baly, 1876, and Podagrica hibisci Bryant, 1941 with N. gemella (Erichson, 1834). Lectotypes are designated for Haltica gemella Erichson, 1834, N. chrysomeloides Jacoby, 1885, N. bowringi Baly, 1876, and Podagrica hibisci Bryant, 1941.

There are four lysosomal diseases of which the neuronal ceroid lipofuscinosis is the rarest. The clinical presentation and their characteric abnormal ultrastructure subdivide them into four types. These are known as the Infantile form (Santavuori-Haltia), Late infantile form (Jansky-Bielschowsky), Juvenile form (Batten-Spielmeyer-Voght) and the Adult form (Kuph's).An 8 year old Omani girl presented wth myclonic jerks since the age of 4 years, with progressive encephalopathy, mental retardation, ataxia and loss of vision. An ophthalmoscopy was performed followed by rectal suction biopsies (fig. 1). A previous sibling had died of an undiagnosed neurological disorder with a similar clinical picture.

Pajak memiliki peranan penting bagi perekonomian Negara, namun di sisi lain, pajak dianggap sebagai biaya yang akan mengurangi keuntungan perusahaan dan memperkecil jumlah laba bersih. Oleh karena itu, perusahaan melakukan berbagai kegiatan untuk dapat meminimalkan beban pajaknya. Agresivitas pajak merupakan upaya yang perusahaan gunakan dalam mengurangi beban pajaknya dengan kegiatan mengurangi pemasukan kena pajak melalui dua cara, yaitu Tax Avoidance & Tax Evasion. Meskipun dalam beberapa haltidak bertentangan dengan peraturan yang ada, tetapi semakin besar celah penghematan pajak sebuah industri maka industri tersebut semakin agresif atas pajak. Penelitian ini bertujuan untuk mengetahui determinan faktor dari agresivitas pajak. Penelitian ini dilakukan terhadap perusahaan manufaktur sub sektor makanan dan minuman yang terdapat di Bursa Efek Indonesia periode 2018-2021, dengan metode pemilihan diperoleh sampel penelitian sebanyak 24 perusahaan. Data yang digunakan adalah laporan keuangan yang dipublikasikan oleh Bursa Efek Indonesia. Alat analisis yang digunakan adalah analisis regresi linear berganda untuk mengetahui pengaruh ukuran perusahaan, capital intensity, pendanaan aset, dan profitabilitas terhadap agresivitas pajak. Pengolahan data dalam penelitian ini menggunakan program Statistical Package for the Social Science (SPSS) versi 25. Dari hasil penelitian diketahui bahwa ukuran perusahaan dan capital intensity berpengaruh positif terhadap agresivitas pajak, pendanaan aset dan profitabilitas berpengaruh negatif terhadap agresivitas pajak.

The substitution of an active-site aspartic acid residue by asparagine in the lysosomal protease cathepsin D (CTSD) results in a loss of enzyme activity and severe cerebrocortical atrophy in a novel form of neuronal ceroid lipofuscinosis in sheep [Tyynelä, Sohar, Sleat, Gin, Donnelly, Baumann, Haltia and Lobel (2000) EMBO J. 19, 2786—2792]. In the present study we have introduced the corresponding mutation by replacing aspartic acid residue 293 with asparagine (D293N) into the mouse CTSD cDNA to analyse its effect on synthesis, transport and stability in transfected HEK-293 cells. The complete inactivation of mutant D293N mouse CTSD was confirmed by a newly developed fluorimetric quantification system. Moreover, in the heterologous overexpression systems used, mutant D293N mouse CTSD was apparently unstable and proteolytically modified during early steps of the secretory pathway, resulting in a loss of mass by about 1kDa. In the affected sheep, the endogenous mutant enzyme was stable but also showed the shift in its molecular mass. In HEK-293 cells, the transport of the mutant D293N mouse CTSD to the lysosome was delayed and associated with a low secretion rate compared with wild-type CTSD. These data suggest that the mutation may result in a conformational change which affects stability, processing and transport of the enzyme.

AbstractThe first aim of the current study was to examine the latent structure of attachment states of mind as assessed by the Adult Attachment Interview (AAI) among three groups of parents of children at risk for insecure attachments: parents who adopted internationally (N= 147), foster parents (N= 300), and parents living in poverty and involved with Child Protective Services (CPS;N= 284). Confirmatory factor analysis indicated the state of mind rating scales loaded on two factors reflecting adults’ preoccupied and dismissing states of mind. Taxometric analyses indicated the variation in adults’ preoccupied states of mind was more consistent with a dimensional than a categorical model, whereas results for dismissing states of mind were indeterminate. The second aim was to examine the degree to which the attachment states of mind of internationally adoptive and foster parents differ from those of poverty/CPS-referred parents and low-risk parents. After controlling for parental age, sex, ethnicity, and socioeconomic status, (a) internationally adoptive parents had lower scores on the dismissing dimension than the sample of community parents described by Haltigan, Leerkes, Supple, and Calkins (2014); (b) foster parents did not differ from community parents on either the dismissing or the preoccupied AAI dimension; and (c) both internationally adoptive and foster parents had lower scores on the preoccupied dimension than poverty/CPS-referred parents. Analyses using the traditional AAI categories provided convergent evidence that (a) internationally adoptive parents were more likely to be classified as having an autonomous state of mind than low-risk North American mothers based on Bakermans-Kranenburg and van IJzendoorn's (2009) meta-analytic estimates, (b) the rates of autonomous states of mind did not differ between foster and low-risk parents, and (c) both internationally adoptive and foster parents were less likely to be classified as having a preoccupied state of mind than poverty/CPS-referred parents.

Abstract Introduction Neoadjuvant chemotherapy (NACT) has emerged as a well-established treatment modality for advanced-stage patients with high-grade serous carcinoma (HGSC). However, relapse and platinum resistance remain significant challenges, resulting in unfavorable long-term outcomes. The interplay between NACT and the tumor microenvironment (TME) is crucial for treatment response, yet its mechanisms are poorly understood. Methods We prospectively recruited >300 newly-diagnosed patients to the Oncosys-OVA study. We performed BRCA1/2 mutational profiling and genomic scarring (ovaHRDscar) to annotate the HGSCs to distinct genomic groups. Detailed surgical and clinical data were collected to assess NACT responses, while histological examination using H&E staining annotated the morphological response patterns of HGSC. Tissue cyclic immunofluorescence (tCycIF) analysis was conducted on FFPE samples collected from the same cohort. We built and validated a tCycIF panel with antibodies targeting cell cycle-regulators and checkpoint inhibitors (Cyclin A, Cyclin D, p21, p27), proliferation (Ki-67, PCNA), and DNA damage markers (RAD51, γH2AX, p53), alongside other cell-type markers for tumor, stromal, lymphoid and myeloid populations. We used GeoMX spatial transcriptomics to capture spatially resolved and cell-population deconvoluted gene expression profiles within regions of interest from tCycIF spatial analyses. Results We employed tCycIF utilizing a validated custom-made antibody panel targeting up to 50 markers to characterize the cell populations in the TME, including their functional states, cell cycle profiles and DNA damage patterns. Via advanced image analysis, we revealed the spatial topographies of tumor-immune and stromal cells, cellular communities, and cell-cell interactions within the TME. Through the utilization of computational methods, we reconstructed the cell cycle dynamics and assessed the expression of markers in cells in pseudotime. Furthermore, by conducting a multivariate assessment of the proliferation markers, we were able to evaluate the spatial distribution of cells with distinct proliferative characteristics. To explore the transcriptome of these regions, we developed a pipeline that integrates tCycIF and GeoMx data, enabling the precise targeting of cell populations in adjacent slides. By examining the gene-expression signatures and enriched pathways within these regions, we uncovered molecular signatures associated with distinct morphological response patterns to NACT. These comprehensive analyses were integrated with the matching clinical, pathological, and genomic profiling information, leading to the identification of recurring cellular neighborhoods with potential therapeutic implications. Conclusions By integrating multi-omics and spatio-temporal analyses with clinicopathological features, we identified response-related patterns that could inform patient stratification and treatment selection. This study can potentially drive advancements in immuno-oncology and personalized therapeutic approaches for patients with HGSC. Abstract Introduction: Single agent immunotherapies have shown poor responses in high-grade serous ovarian cancer (HGSC), highlighting the importance of combinatorial approaches. However, identifying patients who will benefit from different combinatorial approaches requires representative ex vivo models containing the native immune microenvironment. Here, we established HGSC patient-derived immunocompetent cultures (iPDCs) for high-throughput testing of targeted agents and immunotherapy combinations at single cell level. Material and methods: Freshly resected HGSC patient tumor tissue samples were collected under the approval of ethical board of the Women’s Hospital, Helsinki University Hospital. Tumors were enzymatically dissociated and cultured in a patient-derived matrix with customized growth medium. Immune cell abundance and composition in the source tumor and matched cultures were evaluated using flow cytometry. The genetic landscapes of the iPDCs were profiled using whole-exome sequencing. iPDC responses to combination of DNA damaging, and immunotherapy agents were evaluated using single cell fluorescent imaging, and multiparameter flow cytometry. Results: We show that our novel patient-derived matrix significantly improved the growth of iPDCs compared to the conventional matrix. Importantly, we validate that the iPDCs morphologically and genetically match the source tumor and contain functional immune cells of the native tumor, including T cells, NK cells, and a subset of myeloid cells relevant for response to immunotherapies. Importantly, iPDCs with homologous recombination DNA repair deficiency responded to poly ADP-ribose polymerase inhibitor (PARPi), and to anti-PD1 antibody treatment measured by decreased tumor cell count. Further, the iPDC responses to PARPi or cisplatin corresponded to the clinical responses in PARPi and/or chemo-resistant patients. Interestingly, while iPDCs from the BRCA1 mutant and PARPi resistant patient showed sensitivity to the WEE1 and Chk1/2 inhibitors, iPDCs from another PARPi resistant patient with BRCA1 reversion mutation did not respond to these agents, suggesting distinct mechanisms driving drug sensitivities following PARPi resistance. Importantly, single-cell RNA sequencing revealed distinct epithelial and immune cell pathway activity in PARPi and chemo-resistant patients correlating with drug responses from iPDC. Furthermore, we identify patient-specific responses to novel immunotherapy and DNA damaging agents including drugs targeting the cyclic GMP–AMP synthase (cGAS) stimulator of interferon gene (STING), and Lysophosphatidic acid (LPA) pathways. In summary, these results highlight the utility of iPDCs for the investigation of patient-specific drug sensitivity and resistance in HGSC. Conclusion: Our novel iPDC culture system harboring physiologically relevant patient-derived matrix combined with patient-specific tumor infiltrating immune cells provide a platform for high- throughput testing of immuno-oncology agents and combination treatments in HGSC. The purpose of this study is to investigate whether father involvement can influence children's problematic behavior. This study used the systematic literature review (SLR) method by referring to 10 valid articles published in the last 10 years with the publication years between 2013 - 2023. The finding of the literature shows that there is a significant impact of father involvement on the children's problematic behavior. The more the father is involved in the children's development, the lower the level of children's problematic behavior is. The image of a father as a mentor and motivator for early childhood can still be explored in depth. Because of the limitations of this study, this study suggests that future research can further discuss the impact of the father’s involvement in the children's problematic behavior in a specific cultural aspect by considering cross-cultural factors. This is intended to conclude the relationship between the father’s interactive situations with the impact of the father’s involvement in the children's problematic behavior. West Indian genera of the flea beetle subtribe Oedionychina Chapuis, 1875 are reviewed. Two new genera and one new species are described: Argusonychis Konstantinov, Van Roie, & Furth gen. nov. with Argusonychis clarki Konstantinov, Van Roie & Furth sp. nov. as the type species (Bahamas); and Ciguapanychis Konstantinov, Van Roie & Furth gen. nov. with Alagoasa bicolor (Linnaeus, 1767) as the type species (Puerto Rico). An illustrated key to the genera found in the West Indies is provided. Each genus is illustrated with images of external and internal morphology, including thoracic structures, metendosternite, and female and male genitalia. Asphaera subfasciata Clark, 1860 is designated as the type species for Asphaera Duponchel & Chevrolat in d’Orbigny, 1842. The genus Ptena Chevrolat, 1836 is considered a long-overlooked senior synonym of Asphaera; the use of Asphaera is maintained. The lectotypes are designated for: Chrysomela albicollis Fabricius, 1787, Oedionychis crucipennis Suffrian, 1868, and Haltica decipiens Klug, 1829. Pleurasphaera Bechyné, 1958 is recognized as an objective junior synonym of Litosonycha Chevrolat, 1836 since these two generic names share the same type species, Haltica decipiens Klug, 1829. Oedionychis punctipennis Blake, 1971 is synonymized with Alagoasa sharpi (Jacoby, 1897). The following 17 new combinations are proposed: Alagoasa lunata (Fabricius, 1801), Alagoasa sharpi (Jacoby, 1897), Alagoasa smithi (Jacoby, 1897), Argusonychis megalopia (Blake, 1939), Argusonychis orientensis (Blake, 1959), Argusonychis zayasi (Blake, 1959), Asphaera amplilimbata (Blake, 1959), Asphaera cristalensis (Blake, 1959), Ciguapanychis bicolor (Linnaeus, 1767), Ciguapanychis dimidiatus (Olivier, 1808), Ciguapanychis fasciatus (Fabricius, 1798), Ciguapanychis jamaicensis (Jacoby, 1894), Ciguapanychis pseudofasciatus (Blake, 1940), Ciguapanychis viridipennis (Blake, 1971), Kuschelina pseudothoracica (Blake, 1945), Omophoita turquino (Zayas, 1988), and Walterianella gurneyi (Blake, 1968).
