BackgroundTransforaminal epidural steroid injection is widely used in clinical practice to effectively deliver injectate into the ventral epidural space. Complications associated with intravascular injection such as spinal cord infarction and paraplegia can occur during transforaminal epidural steroid injection. To improve the safety of the procedure, avoidance of intravascular injection is crucial, for which appropriate needle selection is important. The primary aim of this study was to compare intravascular injection rates during transforaminal epidural steroid injection between commonly used Quincke and Tuohy needles.MethodTwo hundred and four transforaminal epidural steroid injection cases were randomly assigned to one of two needle groups (22-gage Quincke needle or 22-gage Tuohy needle). Intravascular injection was evaluated using digital subtraction angiography. Spread of contrast medium to the ventral and medial epidural spaces was evaluated. Procedure time was compared between the two needle types.ResultsThe overall incidence of intravascular injection was 7.8%. The rate of intravascular injection was significantly lower in the Tuohy needle group than the Quincke needle group (2.9% vs 12.7%, p=0.009). The ventral and medial epidural spread rates of the Tuohy needle group were 92.2% and 95.1%, respectively, significantly higher than those of the Quincke needle group. The procedure time was shorter in the Tuohy needle group than in the Quincke needle group (97.4 (19.3) seconds vs 117.8 (31.9) s; mean difference −20.40 (95% CI −34.35 to −6.45), p=0.005).ConclusionsIn conclusion, Tuohy needles had a lower intravascular injection rate and higher medial and ventral epidural spreading rates than Quincke needles.Trial registration numberKCT0002095.

The accurate characterization of linear coefficient of thermal expansion (CTE) of thin films is vital for predicting the thermal stress, which often results in warpage and failure of a MEMS structure. In this paper, special emphasis is placed on the development of novel test method to extend an ISDG (Interferometric Strain/Displacement Gage) technique to the direct and accurate CTE measurement of MEMS materials, AlN and Au. The freestanding AlN and Au films are 1 μm thick and 5 mm wide. Strain is directly measured by a brand-new digital type ISDG with two Cr lines deposited on the specimen while heating a specimen in a furnace. The whole test system is verified first by measuring the CTE for the NIST’s SRM (Standard Reference Material) 736 (Cu) block. The measured CTE is 17.3 με/oC up to 167 oC, which agrees well with the NIST’s certified value. The CTE of Au is 25.4 ± 1.15 με/oC and that of AlN film is 3.77 ± 0.12 με/oC. The in-plane displacement resolution is about 5 nm at the best circumstances.

ABSTRACTMammalian prions are unconventional infectious agents composed primarily of the misfolded aggregated host prion protein PrP, termed PrPSc. Prions propagate by the recruitment and conformational conversion of cellular prion protein into abnormal prion aggregates on the cell surface or along the endocytic pathway. Cellular glycosaminoglycans have been implicated as the first attachment sites for prions and cofactors for cellular prion replication. Glycosaminoglycan mimetics and obstruction of glycosaminoglycan sulfation affect prion replication, but the inhibitory effects on different strains and different stages of the cell infection have not been thoroughly addressed. We examined the effects of a glycosaminoglycan mimetic and undersulfation on cellular prion protein metabolism, prion uptake, and the establishment of productive infections in L929 cells by two mouse-adapted prion strains. Surprisingly, both treatments reduced endogenous sulfated glycosaminoglycans but had divergent effects on cellular PrP levels. Chemical or genetic manipulation of glycosaminoglycans did not prevent PrPScuptake, arguing against their roles as essential prion attachment sites. However, both treatments effectively antagonizedde novoprion infection independently of the prion strain and reduced PrPScformation in chronically infected cells. Our results demonstrate that sulfated glycosaminoglycans are dispensable for prion internalization but play a pivotal role in persistently maintained PrPScformation independent of the prion strain.IMPORTANCERecently, glycosaminoglycans (GAGs) became the focus of neurodegenerative disease research as general attachment sites for cell invasion by pathogenic protein aggregates. GAGs influence amyloid formationin vitro. GAGs are also found in intra- and extracellular amyloid deposits. In light of the essential role GAGs play in proteinopathies, understanding the effects of GAGs on protein aggregation and aggregate dissemination is crucial for therapeutic intervention. Here, we show that GAGs are dispensable for prion uptake but play essential roles in downstream infection processes. GAG mimetics also affect cellular GAG levels and localization and thus might affect prion propagation by depleting intracellular cofactor pools.

This template explains and demonstrates how to design a measurement system based on the size of the linear structured light vision, the system could works at realized the high precision and fast measurement of the size of mechanical parts, and accurate calibration of the system. First of all, this paper set up the experimental platform based on linear structured light vision measurement. Secondly, this paper established a system of measurement model, and puts forward a new method of calibration of structured light sensor and set up the mathematical model of sensor calibration. This calibration method only need to use some gage blocks of high precision as the target, the target position need not have a strict requirements, and the solving process will be more convenient, much easier to field use and maintenance. Finally, measuring accuracy on the system by gage blocks with high precision is verified, the experiment shows that measurement accuracy within 0.050 mmin the depth of 0-80 - mm range. This system can satisfy the demands of precision testing of most industrial parts .with its simple calibration process and high precision, it is suitable for the structured light vision calibration.

The effect of glycosaminoglycans (GAGs) on the proliferation of smooth muscle cells (SMC) and fibroblasts was assessed by culturing cells with or without GAGs. Porcine heparan sulphate (HS) inhibited proliferation in a dose dependent manner. At 167 μg/ml of HS this reached 88% and 72% inhibition of SMC and fibroblast growth, respectively. Pig and beef mucosal heparins also blocked proliferation, but to a lesser extent. In contrast, beef lung heparin, chondroitin sulphate, and dermatan sulphate failed to block growth factor induced proliferation. Continuous presence of HS was not required, suggesting that the inhibitory effects resulted from a direct effect on the cell rather than an interaction of the GAG with growth factors. The mechanism by which GAGs inhibit proliferation will be addressed in future studies.

558 Background: Genomic platforms, such as Mammaprint (Agendia) (MP) and OncoType (Genomic Health) (OT), have been validated to determine the risk of relapse in therapeutic decision-making in early-stage hormone receptor positive (HR+), epidermal growth factor receptor 2 (HER2) negative breast cancer (BC). Discordances in risk allocation between these platforms affect up to 30% of patients. This study aims to develop the MamaPred test to improve the diagnostic performance of recurrence risk in HR+/HER2- early-stage BC. Methods: A total of 606 HR+/HER2- early-stage BC previously tested with OT [n = 287; Low Risk (LR) = 165, Intermediate Risk (IR) = 103 and High Risk (HR) = 19] and MP (n = 319; LR = 217 and HR = 102) were included. A retrospective independent series of 144 HR+/HER2- early-stage BC [median follow-up: 10.53 years (range: 3.1-23.1 yrs); age (median = 62.9 yrs (33-89 yrs); systemic relapse 10.5% (n = 15)] was used as validation set.The expression levels of 2560 cancer-related mRNAs were evaluated from one 5 μm thin-section of a FFPE block (15 mm2 tumor area) using the Oncology Biomarker Panel (OBP) and the HTG EdgeSeq System (HTG Molecular Diagnostics. Inc) and quantified by NGS on a NextSeq550 sequencer (Illumina). A predictive model was built from normalized and logarithmically transformed values (rescaled to [0, 1]) using as response a binary meta-variable constructed by taking the values -1 (for LR of MP and OT together the OT IR) and 1 (for HR MP and OT). Differential expression, GSEA and visualization were performed with DESeq2, gage and pathview packages respectively in R v4.0.1. Results: MamaPred consists of a logistic regression classifier with an elastic net penalty (mix of L1 and L2 priors as regularizer) where the mixing parameter is optimized along with regularization strength by selecting the ones that minimize the area under the precision and recall curve over a validation split for each training fold. Metrics of MamaPred were: balanced accuracy, 80.5%; Kappa, 0.562; specificity, 80.7%; and NPV, 91.4%. GSE analysis on differentially expressed genes (q < 0.1) showed four KEGG pathways overrepresented in HR (p < 0.05): adherens junction, tight junction, glutathione metabolism and focal adhesion; and two underrepresented: DNA replication (p = 0.0765) and pyrimidine metabolism (p = 0.086).The prognostic prediction of MamaPred was validated on the independent retrospective series, distant disease-free survival for HR and LR being 88.63% (95% IC: 78.72%-99.78%) and 98.1% (95% IC: 95.6%-100%) respectively (p = 0.00603). Correlation between the probabilities assigned to any given sample and its replicas was extremely high (r > 0.9 p < 1e-5). Conclusions: MamaPred identifies HR+/HER2- early-stage BC patients with high-risk of distant relapse improving the prognostic value of those studies that compare MP and OT, suggesting a more precise risk classification.

Black men have the highest age-adjusted death rate of any major race-gender group in America. Understanding their perceived barriers to accessing health care may benefit future interventions working to increase Black men’s health care engagement. Data collected from focus groups of Black men( N = 67), key informant interviews( N = 12), and interviews( N = 5) with participants who pilot tested an online health education system (called “Gabe”) were analyzed to explore their health care experiences and how computer-based health programs might better assist Black men. Concerns pertaining to health care systems’ failure to recognize the diversity among Black men, and physicians’ lack of sociocultural awareness about the challenges they regularly face, were most salient. Building trust with providers was cited as being central to engagement, with Gabe users perceiving the system to be both trustworthy and accessible. Participants reported an openness to technology assisting with health management and provided suggestions of how online systems can meet the needs of Black men.

High carbon steel stainless steel such as 440C martensitic stainless steel, are commonly used for automotive components, such as ball bearings, races, gage blocks and valve. In this study, 440C steel was coated with ZrO2 by electrolytic deposition in ZrO(NO3)2 aqueous solution. After annealing, the ZrO2 coated specimens were characterized by x-ray diffraction (XRD) and scanning electron microscope (SEM). Scanning electron micrograph showed that thickness of the coated sample was approximately 0.7µm. Besides that, secondary hardening effect occurred on the annealed SS440C substrate and it might be due to the presence of secondary carbide.

Glycosaminoglycans (GAGs) are linear polymers of amino sugar uronic acid disaccharides, and are generally attached to protein cores to form proteoglycans. GAGs interact with a large number of proteins and can participate in matrix organization, cell adhesion, differentiation, growth and apoptosis. Proteoglycans are expressed in tightly regulated spatio-temporal patterns during organ development, and changes in expression frequently correlate with developmental events. Here we review the evidence that GAGs play important roles in the development of mouse kidneys, which are organs that will undergo organotypic development in simple culture conditions and that are therefore highly accessible to experimentation. Depleting kidneys of GAGs, either biochemically or genetically, blocks the development of the urinary collecting-duct system, probably because critical signalling molecules require GAGs to form stable associations with their receptors. The in-sensitivity of GAG-deprived organ rudiments to physiological concentrations of growth factors can be used to screen candidate signalling molecules for morphoregulatory activity; candidate growth factors are applied at supraphysiological levels to GAG-deprived kidneys and assessed for their ability to rescue normal development. This approach has assisted the identification of four collecting-duct morphogens: hepatocyte growth factor, glial cell line-derived neurotrophic factor, nerturin and persephin.

The paper presents an application of resistive ladder sensors (or resistive crack gages) for health monitoring of an aircraft structure. An experiment was conducted during a Full Scale Fatigue Test (FSFT) of PZL 130 ORLIK TC II turbo-prop military trainer. The sensor can be successively used to detect and quantify fatigue cracks. It is a parametric transducer, similar to foil strain gages. Differences in shape of the measuring grid can be noticed. Principle of operation, sensor’s characteristics as well as block diagram of measurement system is presented in the paper. Also examples, both laboratory and FSFT data are delivered to prove surface crack detection capabilities. Simultaneously with new data collection, some investigation is carried out, connected with customize hardware elaboration, signal processing and structure-sensor integration method, what is briefly described.

ABSTRACTFiloviruses, including both Ebola virus (EBOV) and Marburg virus (MARV), can infect humans and other animals, causing hemorrhagic fever with a high mortality rate. Entry of these viruses into the host is mediated by a single filoviral glycoprotein (GP). GP is composed of two subunits: GP1, which is responsible for attachment and binding to receptor(s) on susceptible cells, and GP2, which mediates viral and cell membrane fusion. Although numerous host factors have been implicated in the entry process, the initial attachment receptor(s) has not been well defined. In this report, we demonstrate that exostosin 1 (EXT1), which is involved in biosynthesis of heparan sulfate (HS), plays a role in filovirus entry. Expression knockdown of EXT1 by small interfering RNAs (siRNAs) impairs GP-mediated pseudoviral entry and that of infectious EBOV and MARV in tissue cultured cells. Furthermore, HS, heparin, and other related glycosaminoglycans (GAGs), to different extents, can bind to and block GP-mediated viral entry and that of infectious filoviruses. These results strongly suggest that HS and other related GAGs are attachment receptors that are utilized by filoviruses for entry and infection. These GAGs may have therapeutic potential in treating EBOV- and MARV-infected patients.IMPORTANCEInfection by Ebola virus and Marburg virus can cause severe illness in humans, with a high mortality rate, and currently there is no FDA-approved vaccine or therapeutic treatment available. The ongoing 2014 outbreak in West Africa underscores a lack of our understanding in the infection and pathogenesis of these viruses and the urgency of drug discovery and development. In this study, we provide several pieces of evidence that demonstrate that heparan sulfate and other closely related glycosaminoglycans are the molecules that are used by filoviruses for initial attachment. Furthermore, we demonstrate that these glycosaminoglycans can block entry of and infection by filoviruses. Thus, this work provides mechanistic insights on the early step of filoviral infection and suggests a possible therapeutic option for diseases caused by filovirus infection.

Glycosaminoglycans (GAGs) play an important role in many biological processes in the extracellular matrix. In a theoretical approach, structures of monosaccharide building blocks of natural GAGs and their sulfated derivatives were optimized by a B3LYP6311ppdd//B3LYP/6-31+G(d) method. The dependence of the observed conformational properties on the applied methodology is described. NMR chemical shifts and proton-proton spin-spin coupling constants were calculated using the GIAO approach and analyzed in terms of the method's accuracy and sensitivity towards the influence of sulfation, O1-methylation, conformations of sugar ring, andωdihedral angle. The net sulfation of the monosaccharides was found to be correlated with the1H chemical shifts in the methyl group of the N-acetylated saccharides both theoretically and experimentally. Theωdihedral angle conformation populations of free monosaccharides and monosaccharide blocks within polymeric GAG molecules were calculated by a molecular dynamics approach using the GLYCAM06 force field and compared with the available NMR and quantum mechanical data. Qualitative trends for the impact of sulfation and ring conformation on the chemical shifts and proton-proton spin-spin coupling constants were obtained and discussed in terms of the potential and limitations of the computational methodology used to be complementary to NMR experiments and to assist in experimental data assignment.

Abstract Cancer-testis (CT) antigens are expressed in a variety of malignant tumors but in normal adult tissues solely in testicular germ cells. Based on this tumor-associated expression pattern, CT antigens are considered valuable targets for immunotherapy. While CT antigens have been studied in many solid tumors, much less is known about their presence in neoplasms of the hematopoetic system such as Non Hodgkin’s Lymphoma (NHL). Aims: To analyze the expression of 10 CT antigens in NHL by immunohistochemistry (IHC) using tissue microarray (TMA) technology, and to correlate its expression with histologic subtypes. Patients and Methods: Formalin-fixed paraffin-embedded tissues of 116 NHL cases dating from June 2003 to June 2007 were retrieved from the archives of the Department of Pathology, Federal University of São Paulo, Brazil. Two TMA blocks were generated comprising three cores/NHL case, as well as positive (testis) and negative (reactive lymph nodes) control tissue cores. The following monoclonal antibodies (mAbs) (to the following antigens) were used for IHC analysis: MA454 (MAGE-A1), M3H67 (MAGE-A3), 57B (MAGE-A4), CT7-33 (CT7/MAGE-C1), CT10#5 (CT10/MAGE-C2), E978 (NY-ESO-1), 219-510-23 (LAGE-1 and NY-ESO-1), #26 (GAGE) and SSX-2#7 (SSX-2). Two observers scored all slides independently and cases with loss of all three punches and/or missing tumor tissue were excluded from analysis. Results: The number of cases which could not be evaluated due to loss of tissue core and/or missing tumor differed, ranging between 8 for mAb #26 to 12 for mAbs MA454, CT10#5 and 219-510-23. 7/116 NHL cases were missing in all slides. 14/109 (12.8%) NHL cases expressed at least one CT antigen: diffuse large B-cell lymphoma (DLBCL) 9/58 (15.5%); anaplastic large cell lymphoma (ALCL) 1/3; follicular lymphoma (FL) 1/10 (10%); lymphoplasmacytic lymphoma (LPL) 1/3; peripheral T-cell lymphoma (PTCL) 1/9 and histiocytic lymphoma (HL) 1/1. No CT antigen expression was present in lymphoblastic lymphoma (5), mantle cell lymphoma (2), small lymphocytic lymphoma (6), marginal zone lymphoma (4), MALT lymphoma (5) and mycosis fungoides (3). The most frequently expressed CT antigens were: GAGE 6/108 (5.5%), NY-ESO-1 5/105 (4.7%), CT7 5/106 (4.7%), MAGE-A1 4/104 (3.8%) and MAGE-A3 4/107 (3.7%). According to cell origin, positivity of CT antigens was more prevalent in B-cell NHL (11/91; 12.1%) than in T-cell NHL (1/15; 6.7%). NY-ESO1 5/91(5.5%) was the most frequently expressed CT in B-cell NHL, followed by GAGE 4/91 (4.4%), MAGE-A1 3/91 (3.3%), MAGE-A3 3/91 (3.3%), CT7 3/91 (3.3%) and LAGE 3/91 (3.3%). The only positive T-cell lymphoma case expressed MAGE-A1 and GAGE. Conclusions: Expression of CT antigens in NHL is limited. In the present TMA-based IHC analysis, only 12% of NHLs express at least one CT antigen, with DLBCL showing the highest incidence and NY-ESO-1 being the most prevalent antigen. In spite of the low incidence, presence of CT antigens could offer the opportunity of vaccine-based immunotherapy in selected cases of NHL.

Abstract Determining how extracellular matrix (ECM) components influence cytokine-induced growth and differentiation of cells in malignancies and other diseases is critical for understanding disease pathophysiology and for developing novel treatment strategies. Bone morphogenetic proteins (BMPs) regulate the growth, differentiation and apoptosis of cells in the brain, bone, bone marrow and diverse tissues. ECM glycosaminoglycans (GAGs) such as heparan sulfate (HS) interact with and influence the biological activity of a number of proteins including BMPs. We examined if heparin, endogenous HS in malignant cells and the structurally abnormal HS accumulated in Hurler cells influence BMP signaling and activity. First we showed using real-time quantitative RT-PCR (qRT-PCR) that the BMP signaling pathway including BMPs 2–7, BMP and activin receptors and Smad-1 and -5 are expressed by SaOS-2 human osteosarcoma cells. Western immunoblotting showed that BMP-4 induced Smad-1 phosphorylation, activation and nuclear translocation. Optimal Smad-1 activation was achieved by 25 ng/ml BMP-4 at 30–60 min, and blocked by the extracellular BMP antagonist chordin. BMP-4 also induced a concentration-dependent increase in alkaline phosphatase activity, indicative of induction of osteogenic differentiation in these malignant cells. Soluble heparin directly inhibited BMP-4 induced Smad-1 phosphorylation, and also markedly augmented the inhibitory effect of chordin. Similar effects were seen with N-desulfated, N-re-acetylated heparin but to a lesser degree than with heparin, indicating that N-sulfation of glucosamine residues in heparin/HS contributes to the effect of GAGs on BMP signaling. Inhibition of sulfation of endogenous GAGs by sodium chlorate augmented BMP-4 mediated increase in alkaline phosphatase, suggesting that endogenous sulfated GAGs themselves block BMP-4 mediated malignant cell differentiation. Because BMPs play a critical role in neurogenesis and osteogenesis, we also examined if GAGs that accumulate in Hurler syndrome impair BMP-4 signaling. Neurological dysfunction and skeletal abnormalities are among the most devastating manifestations of Hurler syndrome, an inborn metabolic disorder due to lack of lysosomal GAG-degrading α-L-iduronidase (IDUA) enzyme that leads to HS and dermatan sulfate GAG accumulation. We recently showed that HS in Hurler syndrome cells are structurally and functionally abnormal, and have impaired capability to bind and mediate FGF-2 signaling (Pan C et al. Blood2005;106:1956–64). In the present study, using Affymetrix microarrays we found that expression of the BMP signaling cascade including BMPs 1–8, -10 and -15, BMP and activin receptors, Smads 1–8, chordin and inhibitors of DNA binding (IDs) 1–4 is equivalent in normal and Hurler bone marrow derived multipotent progenitor cells. In Hurler cells, BMP-4 did induce a concentration- and time-dependent activation and nuclear translocation of Smad-1 (confocal immunofluorescent microscopy). However, BMP-4 activity was significantly enhanced following clearance of the abnormally accumulated GAGs in Hurler cells by recombinant IDUA enzyme, indicating that GAGs in Hurler cells impair BMP-4 activity. Thus, both endogenous GAGs and exogenous (soluble) heparin, via N- and O-sulfated disaccharide residues, inhibit BMP-4 activity. These findings have implications for understanding the pathobiology of diverse diseases, and for developing novel therapeutic agents that may restore BMP signaling and activity.

Zusammenfassung Anamnese Eine 45-jährige Patientin wurde nach suizidaler Taxin-Intoxikation heimatnah stationär aufgenommen. Im Rahmen der Erstversorgung wurde sie intubiert und bei hämodynamisch relevanten bradykarden Rhythmusstörungen mehrfach reanimiert. Vor Verlegung in unser Zentrum erfolgten noch eine Gastroskopie, die Anlage eines passageren Schrittmachers, die Gabe von Natriumbikarbonat sowie die Gabe von Digitalis-Fab. Untersuchungen und Diagnose Bei Übernahme war die Patientin tief sediert, intubiert, kontrolliert beatmet und hoch Katecholamin-pflichtig. Der körperliche Untersuchungsbefund und eine Röntgen-Thorax-Untersuchung waren unauffällig. Im Aufnahme-EKG fanden sich breite Kammerkomplexe. Echokardiografisch hatte die Patientin normal große Herzhöhlen mit global hochgradig reduzierter Funktion ohne Nachweis höhergradiger Klappenvitien. Therapie und Verlauf Bei AV-Block Grad III und hochgradig eingeschränkter kardialer Funktion war die Therapie mit einer passageren Schrittmacheranlage allein nicht ausreichend, und es wurde zusätzlich mit Katecholaminen sowohl der Blutdruck stabilisiert wie auch positiv inotrop behandelt. Nach Verlegung kam es wiederholt zu ventrikulären Tachykardien, welche unter Amiodaron-Gaben sistierten. Nach 48-stündiger supportiver Therapie normalisierte sich die Herzfunktion. Die komplikationslose Extubation der Patientin war im Verlauf möglich und die Patientin konnte in eine psychiatrische Klinik zur weiteren Behandlung verlegt werden. Folgerung Taxin-Intoxikationen sind insbesondere kardiotoxisch. Die passagere Schrittmachertherapie trug zu einer Stabilisierung bei, eine Kreislaufunterstützung mit Katecholaminen war dennoch erforderlich. Wenn die genannten Maßnahmen nicht ausreichen, sollte eine Unterstützung durch eine extrakorporale Membranoxygenierung erwogen werden.

Natural sulfated glycosaminoglycans (GAGs) play a crucial role as components of the extracellular matrix (ECM). They participate in the regulation of important cellular functions including cell growth, differentiation and signalling. The generation of artificial ECM mimicking selected functions of the native ECM is a promising approach to improve the biological acceptance of materials which are in direct contact to living tissue. In this context we developed synthesis routes for polysaccharide and GAG derivatives bearing both bioactive sulfate and reactive (meth) acrylate functions of different degrees of substitution within the sugar repeating unit. In addition, we studied the photochemically initiated cross-linking of these biopolymer derivatives to form biodegradable hydrogels usable as coatings for biomaterials or scaffolds in tissue engineering.

Nests of Sceliphron caementarium (black and yellow mud dauber) were collected from Sant Gadge Baba Amravati University (SGBAU), Amravati campus and analysed for their physicochemical composition. The investigation gave results for the nest as - pH 6.57, porosity 0.87mg/g, bulk density1.02g/mL, moisture content 1.01%, total surface area 5.55g/mgI2 and total surface charge 1.11mmolH+eq/g. Values obtained for the ordinary soil from the campus were pH 7.82, porosity 0.75mg/g, bulk density1.48g/mL, moisture content 2.27%, total surface area 4.76g/mgI2 and total surface charge 0.98mmolH+eq/g. Higher content of calcium and magnesium were found in nest clay samples as compared to the normal soil, whereas the iodine adsorption number and phosphorus were lower than normal soil.

ABSTRACT Vaccinia virus has a wide host range and infects mammalian cells of many different species. This suggests that the cell surface receptors for vaccinia virus are ubiquitously expressed and highly conserved. Alternatively, different receptors are used for vaccinia virus infection of different cell types. Here we report that vaccinia virus binds to heparan sulfate, a glycosaminoglycan (GAG) side chain of cell surface proteoglycans, during virus infection. Soluble heparin specifically inhibits vaccinia virus binding to cells, whereas other GAGs such as condroitin sulfate or dermantan sulfate have no effect. Heparin also blocks infections by cowpox virus, rabbitpox virus, myxoma virus, and Shope fibroma virus, suggesting that cell surface heparan sulfate could be a general mediator of the entry of poxviruses. The biochemical nature of the heparin-blocking effect was investigated. Heparin analogs that have acetyl groups instead of sulfate groups also abolish the inhibitory effect, suggesting that the negative charges on GAGs are important for virus infection. Furthermore, BSC40 cells treated with sodium chlorate to produce undersulfated GAGs are more refractory to vaccinia virus infection. Taken together, the data support the notion that cell surface heparan sulfate is important for vaccinia virus infection. Using heparin-Sepharose beads, we showed that vaccinia virus virions bind to heparin in vitro. In addition, we demonstrated that the recombinant A27L gene product binds to the heparin beads in vitro. This recombinant protein was further shown to bind to cells, and such interaction could be specifically inhibited by soluble heparin. All the data together indicated that A27L protein could be an attachment protein that mediates vaccinia virus binding to cell surface heparan sulfate during viral infection.

In this context, glycosaminoglycans (GAGs), including heparin, have shown the ability to inhibit Pf-iE cytoadherence and growth. Nevertheless, the use of heparin was discontinued due to serious side effects, such as bleeding. Other GAG-based therapies were hampered due to the potential risk of contamination with prions and viruses, as some GAGs are isolated from mammals. In this context, we investigated the effects and mechanism of action of fucosylated chondroitin sulfate (FucCS), a unique and highly sulfated GAG isolated from the sea cucumber, with respect toP. falciparumcytoadhesion and development. FucCS was effective in inhibiting the cytoadherence of Pf-iEs to human lung endothelial cells and placenta cryosections under static and flow conditions. Removal of the sulfated fucose branches of the FucCS structure virtually abolished the inhibitory effects of FucCS. Importantly, FucCS rapidly disrupted rosettes at high levels, and it was also able to block parasite development by interfering with merozoite invasion. Collectively, these findings highlight the potential of FucCS as a candidate for adjunct therapy against severe malaria.

ABSTRACT We previously showed that vaccinia virus infection of BSC40 cells was blocked by soluble heparin, suggesting that cell surface heparan sulfate mediates vaccinia virus binding (C.-S. Chung, J.-C. Hsiao, Y.-S. Chang, and W. Chang, J. Virol. 72:1577–1585, 1998). In this study, we extended our previous work and demonstrated that soluble A27L protein bound to heparan sulfate on cells and interfered with vaccinia virus infection at a postbinding step. In addition, we investigated the structure of A27L protein that provides for its binding to heparan sulfate on cells. A mutant of A27L protein, named D-A27L, devoid of a cluster of 12 amino acids rich in basic residues, was constructed. In contrast to the soluble A27L protein, purified D-A27L protein was inactive in all of our assays, including binding to heparin in vitro, binding to heparan sulfate on cells, and the ability to block virus infection. These data demonstrated that the N-terminal region acts as a glycosaminoglycan (GAG)-binding domain critical for A27L protein binding to cells. Previously A27L protein was thought to be involved in fusion of virus-infected cells induced by acid treatment. When we investigated whether cell surface GAGs also participate in A27L-dependent fusion, our results indicated that soluble A27L protein blocked cell fusion, whereas D-A27L protein did not. Taken together, the results therefore demonstrated that A27L-mediated cell fusion is triggered by its interaction with cell surface GAGs through the N-terminal domain.

The paper presents the main results obtained from experimental and numerical investigations of the thermal and mechanical behaviors observed during curing of a massive concrete element made with type 20M cement. An experimental concrete block 920 mm in diameter and 1000 mm in thickness was instrumented with thermocouples and vibrating-wire strain gages in order to monitor the early age behavior of mass concrete. A numerical model was used to predict the temperature field that occurs in the concrete element upon hydration of cement. The modeling of the mechanical behavior was achieved using a law that accounts for autogenous shrinkage. The experimental results illustrate the importance of selecting the type of cement for use in mass concrete. The data provided by the numerical models are in good agreement with the experimental results. In order to evaluate the structural integrity of the breech system used for a thick-walled cylinder subjected to pulsating high internal pressure, fatigue life simulation of a breech system was performed. A stress analysis of the breech was performed to locate the critical region vulnerable to crack initiation. Low-cycle fatigue behavior of the breech material was investigated to obtain the fatigue crack initiation properties. Elastic-plastic finite element stress analysis resulted in a stress concentration at the breech ring groove root. Strains at the breech ring and block were experimentally measured using strain gages and resulted in similar values compared to the calculated strains. Local strain approach was employed to estimate the fatigue life of the breech system for crack initiation at the groove root of the breech ring. Fatigue tests using simulation specimens were performed and an averaged fatigue life was obtained, showing a very good agreement with the calculated fatigue life within a factor of two.

The purpose of this study was to measure the effect of bone–patellar tendon–bone autograft harvest for anterior cruciate ligament (ACL) reconstruction on the surface strain of the human patella. Through progressive removal of bone from the patella, three different defect shapes as well as the intact patella were tested in each of seven knees. Maximum principal strain and corresponding principal direction were determined from each of three gages around the defect for the four conditions (intact plus three defect shapes). There were no statistically significant differences in overall average surface strain between any of the defect shapes. Following graft harvest, overall average strain (all three defects combined) increased in the patella both medial (15 percent increase) and lateral (34 percent increase) to the defect, while decreasing in the region directly proximal (22 percent increase) to the harvest site compared to the intact patella. A statistically significant 7.5 deg shift of principal direction from longitudinal toward a more transverse (lateral-superior to medial-inferior) direction was observed in the medial region when a shallow-dome defect was made. We conclude that removal of a bone block from the anterior, inferior part of the patella induces a significant redistribution of the surface strain. This results in greater local strain adjacent to the upper border of the bone block, increasing the risk for patella fracture. This effect may be of importance in various complications known to occur after ACL reconstruction.

Selection of die components is an important activity for the design of a compound die. The traditional methods of carrying out this task are complex, time-consuming, and skill-intensive. This paper describes the research work involved in the development of an expert system for selection of components of compound die. The proposed system is constructed in the form of eight modules. For development of system modules, domain knowledge acquired from different sources of knowledge acquisition is refined and then framed in the form of "IF-Then" variety of production rules. System modules are coded in AutoLISP language and user interface is created using Visual Basic (VB). The system is capable of imparting expert advices for selection of type and size of all major die components including die block, die gages, stripper, stripper springs, punches, punch plate, die-set, fasteners, and knockout bar. The proposed system can be implemented on a PC having VB and AutoCAD softwares. Therefore, the implementation cost of proposed system is low and hence easily affordable for small scale stampingindustries.

Zusammenfassung Gegenstand und Ziel: Überprüfung der Sensitivität, Spezifität und Genauigkeit der transrektalen sonographischen Frühträchtigkeitsuntersuchung beim Schaf. Material und Methode: Die Untersuchung erfolgte an 126 Schafen der Rasse Pleven Black Head. Die in drei Untersuchungsgruppen (je n = 42) eingeteilten Tiere wurden mittels gestagenhaltiger Vaginalspangen und Gabe von 500 IE eCG zyklussynchronisiert. Es folgte eine laparoskopische Besamung mit aufgetautem Tiefgefriersperma 54–56 Stunden nach Entfernung der Vaginalspangen. Die transrektale sonographische Trächtigkeitsdiagnose (5 MHz) wurde 20 Tage (Gruppe 1), 25 Tage (Gruppe 2) bzw. 30 Tage (Gruppe 3) später durchgeführt. Die Resultate der sonographischen Trächtigkeitsdiagnostik wurden mit den Ablammergebnissen verglichen. Ergebnisse: Genauigkeit, Sensitivität und Spezifität ergaben sich wie folgt: Gruppe 1: 76,2%, 93,3% und 33,3%; Gruppe 2: 80,9%, 90,6% und 50%; Gruppe 3: 87,8%, 94,4% und 40%. Schlussfolgerung: Die transrektale Sonographie zur frühen Graviditätsdiagnose sollte beim Schaf am 30. Tag der Trächtigkeit durchgeführt werden.

ZusammenfassungEisen (Fe) ist ein essenzielles Spurenelement und nimmt in der tierärztlichen Praxis insbesondere durch seine Bedeutung bei der Eisenmangelanämie einen wichtigen Platz ein. Die Fe-Bereitstellung, z. B. für die Hämoglobinsynthese, stellt hohe Anforderungen an die Eisenhomöostase. Mit der Charakterisierung des Hepcidins als regulatives Protein des Eisenstoffwechsels wurde deutlich, dass Fe einer hormonähnlichen Regulation unterliegt. In der Leber gebildetes Hepcidin reguliert den membranständigen Fe-Transporter Ferroportin. Ein Anstieg der Hepcidinkonzentration führt zu einer Reduzierung der Abgabe von intrazellulär gespeichertem Fe in den Extrazellulärraum. Diese Internalisierung von Fe erfolgt vor allem im retikuloendothelialen System und in Monozyten/Makrophagen, aber auch die enterale Fe-Absorption wird durch Hepcidin vermindert. Auslöser für die hepatische Hepcidinsynthese sind nach derzeitiger Sicht einerseits hohe Fe- bzw. Transferrinkonzentrationen im Plasma. Aber auch Entzündungsmediatoren wie Zytokine führen zur gesteigerten Hepcidinsynthese. Hiermit wurde ein Erklärungsansatz für die oft beobachtete deutliche Abnahme der Fe-Konzentration im Plasma im Rahmen akuter Entzündungen entwickelt, der Mechanismus wird als sog. Zytokin-Hepcidin-Link bezeichnet. Da viele pathogene Mikroorganismen Fe für ihren eigenen Stoffwechsel benötigen, wird die über Hepcidin vermittelte Modulation der Fe-Verfügbarkeit mit Internalisierung von Fe in den Intrazellulärraum der unspezifischen Abwehr zugeordnet. Fe-Supplementationen durch den Tierarzt oder den Landwirt greifen in diese Regulation ein, doch gibt es bisher nur wenige Studien zu den klinischen und metabolischen Auswirkungen oraler oder parenteraler Fe-Supplementierung unter Einbeziehung des Hepcidins. Diese sollte zukünftig wissenschaftlich detaillierter beschrieben werden. Möglicherweise resultiert daraus eine Neubewertung der verschiedenen Formen der Fe-Gabe an Wiederkäuer, Schweine und/oder Pferde.

Recently, as well as hydraulic stability, structural strength of armor blocks has emerged as a major problem in breakwaters. This problem is studied by research organizations in many countries. In Nippon Tetrapod Co., Ltd., a series of hydraulic model tests was conducted to measure the surface strain on Tetrapods under wave action. In this paper, the results of the hydraulic model test are described. Prior to the hydraulic test, a similitude relation on impact stress and strain between the prototype and model was derived and a drop test was performed to investigate the scale effect on impact strain on Tetrapods. The hydraulic test was conducted to measure the surface strain on 50kg model Tetrapods using strain gages. In addition, impact strain on 50t Tetrapods under wave action was estimated applying the similitude relation. The strain on Tetrapods under wave action was clarified through this study.

While working around a basic plot-line of betrayal, Bruno Dumont’s L’il Quinquin references the codes and clichés both of comedy and television crime series by using the serial format to convey the work of a serial killer, and fully exploiting the possibilities for expanding characterization and reduplicating key actions and motifs. Comedy has always been present in Dumont’s work, of course, but only in small doses and only implicitly. If in Humanity the absurdity and burlesque effects were often just plain odd, in L’il Quinquin the laughter is frontal and explicit. It ranges from brutal black humor and caricature to social parody and satire of the police, the Church, science, and the media (long-standing themes in Dumont), and from physical gags and carnivalesque farce to vaudeville grotesquerie. L’il Quinquin swings constantly between the genres of light comedy, murder mystery, social drama, and the study of rural life.
