Der volle Inhalt der QuelleThe environmental risk assessment of chronic exposure to ionizing has become a major concern. The aim of this PhD was to study the multigenerational effects of chronic gamma radiation in an integrated manner (to the life history traits from the subcellular mechanisms) in a model organism, the nematode Caenorhabditis elegans. First, studying the effects of chronic gamma radiation on the life history traits of C. elegans was performed. For that, three generations have been exposed to different dose rates and two generations have been placed in "control" environment after parental exposure. The second part of this thesis aimed to characterize the different subcellular mechanisms that could explain the observed effects on the life history traits after multigenerational exposure. The results showed that (i) the reproduction was the most sensitive endpoint to gamma radiation, (ii) an increase in radiosensitivity was observed over three exposed generations and (iii) the effects of the parental generation were transmitted to the non-exposed generations. An increase in apoptosis, a reduction in the stock of sperm, and to a lesser extent, a decrease in the number of mitotic cells, could explain the observed decrease in reproduction for the exposed generations. Only a decrease in sperm number was observed in parallel with a reduction in the cumulative number of larvae in the non-exposed generations. This research contributes to our knowledge on the multigenerational effects of gamma irradiation and shows the importance of an integrated approach to better understand the mechanisms of action related to the action of a pollutant and improve the environmental risk assessment

Der volle Inhalt der QuelleThis thesis explores the effects of pollutants from the industry in the English Channel, in particular radionuclides, on the early stages of development of the Japanese oyster Crassostrea gigas. Original experimental devices were designed to study in laboratory controlled conditions the growth of a large number of larvae and spats. Emphasis was given to a joint use of stress markers at integrated and molecular levels. First exposures to non-radioactive pollutants revealed a highly sensitive response of the growth of juveniles exposed to a metal (zinc). Thereafter, juveniles were exposed for several weeks to dose rates of ionizing radiation and radionuclide activities several orders of magnitude higher than the levels likely to be encountered in the marine environment. An eco-genotoxicological approach contributed to the understanding of the effects of ionizing radiations on DNA. The results strongly suggest that biochemical mechanisms act efficiently against radio-induced damages as early as the first days of life of the Japanese oyster. It still remains a lack of information about the long-term effects of DNA damages to higher level of biological organization

Der volle Inhalt der QuelleAfter the nuclear accidents at Chernobyl and Fukushima, radiation protection of the environment is a major concern. Studies show an increased sensitivity to ionizing radiation (IR) during development. However, there are few data on the molecular mechanisms leading to the effects of IR on embryogenesis. In this context, the objective of this thesis is to better understand the effects of exposure to IR on the development of zebrafish. Part of this work has focused on studying the effects of IR at an early stage of development. For this, a multi-omics approach with the study of the transcriptome and the methylome has been implemented. These results, highlight potential effects on the development of the central nervous system (CNS) and muscles. The other part of this work focused on assessing the effects of IR exposure at later developmental stages using a system biology approach. The results obtained at the different scales highlight effects on the retinoic acid pathway, the development of the CNS and muscles. This work has shown effects of IR on the development of the CNS and muscles from early development at the molecular level. These effects are confirmed at later stages of development. This study suggests that the molecular effects observed during early development are predictive of the effects observed at later developmental stages. In addition, this thesis work allows to propose a model of "Adverse Outcome Pathway" where the deregulation of the retinoic acid pathway by exposure to IR will lead to effects on the development of the CNS and muscles

Der volle Inhalt der QuelleThe population of uranium miners is a reference population for studying the health effects of chronic exposure to various sources of ionising radiation (IR): radon, gamma rays and uranium dust. However, two statistical problems arise in these studies: 1) the miners' exposures measures are error-prone; 2) the exposures to the three sources of IR are highly correlated. In radiation epidemiology, measurement errors in exposures are often ignored and health risks are estimated source by source, ignoring the synergistic or antagonistic effects of simultaneous exposures. The aim of this work, which is divided into two parts, is to promote the use of hierarchical Bayesian models to address the two problems raised above. All the statistical methods proposed in this work are applied to estimate a health risk from survival data in the French cohort of uranium miners.A model is proposed for estimating a health risk while considering complex measurement errors on radon exposures. These measurement errors depend on the miner's workplace and its work habits which change little over time. These errors are therefore spatially and temporally correlated. They are also heteroscedastic: their variances decrease over time as methods for assessing radon exposure improve. The proposed models are used account for measurement errors in radon exposure when estimating the risk of death by lung cancer, kidney cancer, brain and central nervous system cancer and leukaemia. The correction of the measurement errors and the estimation of the health risk are carried out simultaneously so that the estimation of the risk coefficient account for the uncertainty in the exposures. An MCMC algorithm was implemented in Python 3.8 to infer the model within a Bayesian framework. A simulations study is then carried out to estimate the impact of model misspecification on risk estimates.The three exposures to IRs are considered simultaneously when assessing a health risk by using profil regressions mixture (PRM) models. These models are used to create groups of miners with similar exposure profiles and similar health risks. As before, the inference of groups and the estimation of health risk are carried out simultaneously so that the uncertainty in the grouping is accounted in the estimation of risk. The number of groups in the model is infinite, but only a finite number of groups are non-empty. This assumption, which implies that the number of model parameters is infinite, introduces a difficulty in inferring the model. In addition, the output of the inference algorithm cannot be interpreted directly: post-processing must be carried out in order to form the different groups of individuals. While the choice of post-processing used has an impact on the grouping of individuals, there are only few guidelines on this in the scientific literature. This work proposes a Python implementation of a time-efficient MCMC algorithm for inferring PRM models. This algorithm is used to estimate the risk of death from lung cancer in the French cohort of uranium miners associated with simultaneous exposure to radon, gamma rays and uranium dust. Finally, a simulation study is carried out to compare different post-treatment procedures and provide guidelines on their use

Der volle Inhalt der QuelleThe aim of this work was to assess whether direct intramarrow injection of an adenoviral vector expressing human Il-1a gene stimulates hematopoiesis in healthy non-irradiated and gamma irradiated cynomolgus macaques. In the first hand, we have evaluated the feasability of this gene therapy strategy in two healthy non-irradiated macaques. In this work, we have observed an increase of neutrophil, monocyte and platelets in the two animals treated with the therapeutic construct. This effect was associated with no abnormal clinical side effect. On the other hand, we have evaluated this strategy in non-human primate exposed to a sublethal gamma irradiation. Two of three animals treated by the therapeutic construct reduced significantly the neutropenia, thrombocytopenia and anemia radio-induced. In conclusion, this gene therapy strategy gave a similar clinical benefit comparatively to systemic administration of huIL-1a but without severe side effect
