Der volle Inhalt der QuelleIn this dissertation we treat in three parts the valuation of assets subject to default risk. In the first part we identify the conditions leading to the unicity of prices, for both structural and reduced-form approaches. For the reduced-form modeling, we emphasize the non-unicity of the intensity process, such as it is defined in the credit risk literature. We plead for the methods using the enlargement of filtrations, which permit a more precise characterization of the default time. Finally, we propose a evaluation formula to apply when the prices of default-free assets may jump at the default time. The second part characterizes the optimal leverage and payout policies, when shareholders act to maximize an indicator of the expected performance of shares, which depends on the expected dividends, the level of the default risk and a fixed minimum return level. In this framework, we also define the upper reorganization triggers and give a measure of the agency costs induced by the dividends policy. The third part treats the evaluation when agents face imperfect information. We give a structural explanation of the default event by a state variable, and suppose that a noise continuously obscure the perception of the state variable by the investors. We prove that the (H) hypothesis holds and the default time is totally inaccessible. Lastly, we compute the default probabilities and propose evaluation formulas in presence of incomplete information

Der volle Inhalt der QuelleThis dissertation uses the evaluation of asset managers by professional investors as a case for the study of the ways in which economic agent deal with the necessity of acting and to give account of one's actions while facing uncertainty.This dissertation follows a durkheimian approach as it combines a morphological analysis of the asset management field with an analysis of evaluation practices of asset managers by professional investors.Empirical materials of various kinds have been gathered, in order to record the numerous ways in which a total social fact manifests itself. Some of the materials derive from an ethnographic research in the field of asset management: these are semi-directive interviews, direct observations of social interactions, and artefacts produced and used as part of the practices that the research investigates. The other materials are statistics or institutional data that have been collected during the field research. Interpretive analysis has been applied to them.The morphological analysis of asset management as a social field highlights the divisions, articulations and proportions of the organizations where financial asset management practices are carried out. The autonomization of asset management as a professional field goes along with a social division of labour that is structured around the asset managers. It makes them become speculators, that is economic agents who focus solely on price variations. By contrast, other actors ensure that the speculative movement is followed by a fully carried out trade. The income structure of asset management firms feeds a conflict between asset managers and salespersons about the distribution of profit. The resolution of this conflict leads to a polarization of asset management firms between firms following a model designed after the manufacturing industry, and firms following a model leaning towards craftmanship.The evaluation of asset managers by professional investors makes sense in a social frame where it stands as an appraisal of the efficiency of speculative actions undertaken by agents who try and reduce the financial uncertainty that they face. Professional investors classify their evaluation practices among a "quantitative" analysis and a "qualitative" analysis of asset managers. Although this form of classification may suggest that the two types of analysis cover two kinds of knowledge that are imperfect yet complementary, it is not quantification or calculation per se that differentiates between them. They are functionally related to each other: assessment of the form taken by financial uncertainty on the one hand, and search for the social conditions that caused uncertainty to take such form on the other hand. The form of classification "quantitative/qualitative" meets imperfectly the phenomena to which it is applied. It matches the social distribution of resources that professional investors gained during their socialization trajectory, and that they mobilized in the frame of a specific kind of social division of labour, where analytic practices are also distributed among differentiated positions within organizations.This dissertation complexifies and enriches the understanding of financial practices and of uncertainty reduction practices by showing that such understanding cannot forget their social foundations. It circumscribes the domain of validity of a specific form of classification and it provides interested actors with tools with which they can discuss anew the efficiency or the relevance of their analytic processes

Der volle Inhalt der QuelleOur research investigates the methods of evaluation of public policy concerning the rehabilitation of social welfare housing in France. The analysis is based on 103 evaluations of welfare housing rehabilitation projects on record and an evaluation in Woippy-Saint-Eloy (57) which we assessed. This study permitted us to bring to light the difficulties inherent to an evaluation: choice of an appraiser, the spatio-temporal dimension of resulting effects of an action and the interpretation of those effects. It also clearly points out the political nature of evaluations and demonstrates that the process of evaluation concerning welfare housing rehabilitation does not seem ti achieve it's principal goal: to analyze the consequences of an action and then follow up with the appropriate re-adjustments. The nonexploitation of information gathered during an evaluation excludes the positive evolution of existing programs. Moreover, the knowledge and know-generated by local evaluations rarely attains it's destination in the upper echelons. The evaluation therefore fails to achieve it's purpose, that of a "transitional tool", employed to transmit pertinant information between local communities - responsible for rehabilitation projects - and State Officials - "architects" of public policy

However, the assessment of an action’s impact on flood-related damages is characterized by high complexity. Few research works include the society’s capacity to take action in their models of vulnerability assessment.The objective of this research is to provide an assessment of vulnerability to flooding that takes in account this capacity of action. It relies on an experimental design that aims at assessing the effectiveness of actions through expert judgments. The panel of experts gathers varied actors involved in flood risk management. The method is implemented on Actions Programs for Flood Prevention, as they are a key component of the french flood risk management public policy

Der volle Inhalt der QuelleThe first aim of this study focuses on the apoptotic effect of Berberis Libanotica on human erythroleukemia cell lines HEL, K562, and K562(COX-2+) and it is effect on the expression of COX-2. In light of the reported chemopreventive and chemosensitive effects of natural products on various tumor cells and animal models, we postulated that our Bl extract may mediate their effects through apoptosis induction with suppression of cell survival pathways. We showed that this extract induces apoptosis in eryrhtroleukemia cells by activation of the late markers of caspase-3 activation, PARP cleavage and DNA fragmentation. In the other hand, we showed that Bl extract reduced significantly expression of COX-2 by a dose-dependent manner. In regard to our results, it is clear that the simultaneous inhibition of Akt and NF-κB signalling can significantly contribute to the anticancer effects of Bl extract in human erythroleukemia cells.The second objective of this report is to elucidate wich molecule present in our extract can exert this effects. We found that berberine, the majoritory compound, can induce an apoptotic effect and inhibits the survival pathway of NF-ĸB and p-AKT similarly to the extract.Key words: erythroleukemia, apoptosis, Berberis libanotica ,berberine, COX-2, NF-ĸB, p-AKT

Meanwhile, the emergence of the concept of corporate social responsibility encourages companies to contribute to sustainable development goals. Thus, corporate social responsibility (CSR) approaches are implemented by a growing number of companies, including retailers, to integrate these issues into their business model.But how to evaluate the relevance of the actions against retailers' CSR issues? How to assess their contribution to companies' CSR goals? How to assess the multiple potential positive and negative effects of actions that are initially proposed to reduce one specific environmental impact? How to account for the multitude of stakeholders and issues, sometimes difficult to quantify, subject to uncertainty and changing over time? Despite the numerous existing methods, they do not adequately enable the identification and the assessment of these issues.Conducted within the Environment Department of a multinational retailer, this work tries to overcome these limitations by providing a methodological approach for a prior assessment of social and environmental impacts of actions both qualitatively and quantitatively on the basis of issues listed in the ISO 26000 standard. Der volle Inhalt der QuelleTransition risk, inherent in the shift to a low-carbon economy, presents significant challenges and opportunities for financial markets. This thesis aims to quantify and analyze the influence of this risk on stock returns, taking into account regulatory, technological, and consumer and investor preference developments. Taking part initially in the debate on the most appropriate measure to approximate a company’s contribution to transition risk, in Chapter 1, we propose the use of verified carbon emissions to assess the impact of transition risk on companies participating in the EU ETS. Our results show that the emissions trading system alters the risk-return profile of stocks, which can provide a financial incentive to consider emissions in investment decisions. Next, recognizing the pivotal role of central banks in the transition to a low-carbon economy, Chapter 2 provides an evaluation of the environmental footprint of U.S. monetary policy concerning transition risk. The main conclusion of non-neutrality in U.S. monetary policy, favoring polluting companies, leads to the third chapter. This chapter, also focused on the United States, explores how transition risk is taken into account in market valuation and how climate regulation can influence this integration. Our results suggest that the long-term impact of U.S. laws on the relationship between carbon emissions and stock returns needs strengthening to ensure its sustainability. Currently, legislative measures have a more pronounced effect in the medium term, but their sustainability remains uncertain. In conclusion, these chapters aim to enable a better integration of transition risk into stock evaluation by investors, which would empower regulatory authorities and financial market participants to develop more suitable policies and preventive measures against this risk

Der volle Inhalt der QuelleThe design of the territory project within urban plans comes from a public action instrumentation. The construction of a metropolitan master plan is the source of conflicting interests. In order to succeed in the project, seeking consensus is the rule, to the detriment of its operational aspects. In this context of interpretation, we have for hypothesis that the landscape policies in the urban plan’s aren’t enough defined for an efficient implementation to reach their objectives. Adopting a landscape policy assessment as a tool for the public action analysis, we propose to analyse the SYSDAU’s intervention in his 2001 urban plan. We have experienced a 4-step approach: a historical analysis, a reconstitution of the action theories, the compatibility analysis between the master plan and three different land use plans, and finally, the analysis of the way the population perceive the landscape. The results from this approach help in identifying the efficiency of the landscape scheme, but also its limits. The assessment aims at, not only explaining the impacts of landscape public policy in 2001, but also understanding their mechanisms, that lead to success or failure. The results show how important the landscape is in building a territory program, and it reveals the strength of a landscaped approach in any urban planning. So, we can now offer to the different actors an evaluation of their land policy following an “ex-post” approach, and thus, identify the leverages or the possible ways to improve the wording of the landscape policy of the SCOT to come

The aim of the project was to characterize the range of nucleic acid binding proteins in the context of viral infection in flies. We identified a wide repertoire of proteins, which recognize viral nucleic acids in five species (human, mouse, chicken, fruit fly and roundworm). Among these proteins, there are ones, which are conserved in insects and humans, and therefore their function can be easily studied in the fruit fly model. Afterwards, we have performed a large screen in flies to study more precisely the function of 100 proteins in infection with 5 different viruses. We have found eight promising candidates as a result of this screen. We identified two Drosophila proteins CG5641 and Zn72D, which are also present in humans, as proviral factors. We also identified a protein Tao, which is conserved in humans, and is antiviral against several types of viruses

Der volle Inhalt der QuelleIt is believed that small subpopulation of cells within the tumor, with powerful self-renewal capacity, are involved in tumor progression, aggressiveness and resistance to both chemo- and radio-therapy. These cells, named cancer stem cells (CSCs), are known to express the stemness factors Oct4 and Nanog, when they are highly pluripotent. The aim of my thesis was therefore to analyze the effects of small pharmacological molecules which are able to target in CSCs these self-renewal regulators, in order to bring new effective therapies for cancer. More specifically, this thesis was aimed to study the selective anticancer activity of a synthetic flavagline, namely FL3,on a poorly differentiated and highly malignant CSC model (i.e. the teratocarcinomal stem-like cell) that expresses the stemness factors. Here in we also used a model of very restricted normal stem cell (NSC) (i.e. the fibroblasticstem-like cell), to evaluate the selective effect of this drug. We found that, unlike in NSCS, FL3 was able to trigger a mitochondrial pro-apoptotic process in CSCS, via the activation of p38 MAPK and caspase3, followed by a downregulation of Oct4 and Nanog. We newt investigated the molecular mechanism involved in the protection ofNSCS against the cytotoxic effects of the drug. We found that FL3 selectively activated the prosurvival proteins Akt and Bad in NSCS. Indeed, forced inhibition of the expression of these proteins triggered a caspase-3 proapoptotic process in FL3-treated NSCS. In a next step, we showed that the drug, at low concentration, was able to induce the differentiation of CSCS, by downregulating the expression of Oct4 and Nanog at both transcriptionand translation levels. This effect coincided with an upregulation of the expression of several neural markers. Taken as a whole, the results reported in my thesis clearly demonstrate that the synthetic flavagline FL3 is a powerful anticancer compound, since it acts as a selective proapoptotic and pro-differentiating agent on cancer stem-like cells, without having any effect on normal stem-like cells

This scientific method has been championed as the most rigorous toassess the impact of public intervention. It has promoted the use ofscientific evidence by policy makers through the evidence-based policymovement. Therefore, this method is to be understood as a socialinstitution whose aim is to organize a joint learning between policy actorsand scientists. The study of this interaction is the core of this dissertation,which analyzes how stakeholders’ memberships to their respective strategicaction fields drive this method's use in France. Through the execution ofthis method, we observe a range of different institutional sites. They aretestimonies of the opposing practices, interests and learning patterns of theactors involved. This dissertation analyses 15 case studies of this method'simplementation in France. It reveals the inherent tensions at work in its useon new social programs, and challenges this method's ability to produce ajoint learning between policy actors and scientists

Der volle Inhalt der QuelleCancer stem cells (CSCs) are a small subpopulation of cells in the tumor which are able to self-renew. These cells are involved in the initiation and the metastatic growth of tumors, as well as in the resistance to conventional treatments. In their highly undifferentiated state, CSCs are known to express the stemness factors Oct4 and Nanog. The aim of this work was therefore to analyze the effects of phytochemicals on Oct-4 expressing teratocarcinomal stem-like cells. In a first study, we investigated the selective anti-carcinogenic activity of rooperol, the aglycone of the plant-derived compound hypoxoside. We observed that the rooperol-induced apoptosis in CSCs was associated with an oxidative stress, dependent of the activation of p53 and cleaved caspase 3 expression. These modifications were accompanied by reduced expression of the stemness factors. Such effect was however not observed in normal stem cells (NSCs). We investigated therefore the mechanism involved in the resistance of NSCs to the drug. In contrast to CSCs, rooperol does not cause ROS (reactive oxygen species) formation in NSCs and selectively activates the pro-survival proteins Akt and Bad. In a second study, we examined the selective anti-cancer potential of several acacic acid-type saponins, from different species of African Albizzia, on epidermoid carcinoma cells; in a next step, we investigated the molecular targets of the anti-cancer compound which showed the greatest proapoptotic activity. The study was performed on a model of aggressive metastatic melanoma cell known to express Oct4. We observed that the pharmacological agent had a selective pro-apoptotic effect, via the activation of p38 MAPK and caspase 3, followed by a decrease on the expression of Oct4. Such effect could not be detected in normal cells. These results highlight new and powerful anticancer compounds able to induce selectively apoptosis of CSCs and their descendants, while sparing NSCs and their descendants

Der volle Inhalt der QuelleThe evaluation of the cytotoxic effect of different fruit juices, naturally rich in polyphenols, on four different colorectal cancer cell lines proved that cranberry juice was the most active. Indeed, cranberry polyphenols induce apoptosis associated with the overexpression of two important proapoptotic transcription factors, p73 and FOXO3a on one hand. Furthermore, it has been also correlated with a decrease in the expression of SIRT1, the deacetylase of several non-histone proteins such as p73, KU70 and FOXO. Other early events such as ROS production and DNA damage, which are known to regulate the expression of SIRT1 were confirmed. The second study aims at validating the potential anticancer effects in an in vivo model of colorectal cancer in BALB/c mice injected subcutaneously of murine colon adenocarcinoma cells. Accordingly, we chose the black chokeberry juice, which showed an interesting cytotoxic profile in vitro. The analysis of tumors demonstrated that the administration of chokeberry juice leads to a reduction in tumor cell proliferation. Finally, the significant increase in the mobility of LC3 suggests the activation of autophagic cell death. To validate the clinical use of polyphenols, we evaluated the cytotoxic effects of blueberry polyphenols on lymphocytes of CLL patients. Our results show that the polyphenolic extract induces an oxidative stress-dependent apoptosis that involve various pro-apoptotic proteins in cells of patients with CLL but not in healthy subjects

This development, present in the regulation of April 24, 1996, for the health domain, and the law of January 2nd, 2002, for the medico-social domain, implies that the professional domain be able to formulate innovative solutions, especially with regard to the evaluation of professional actions, and to the on-going improvement of the quality of services. In a context of a global crisis concerning the ways of managing resources, organizational innovations are conceived according to an approach centering on work autonomy and follow-up. Problems of communication and organization overhaul, of professional discourses and stylistics, and other mini-cultural issues are analyzed through a method of participant-observation in order to understand professional mutations taking place since the law of January 2nd, 2002, and of the regulation of April 24th, 1996

Der volle Inhalt der QuelleThe objective of this thesis was to evaluate Espace de Transition (ET), an innovative art-based program aimed at fostering the rehabilitation of adolescents and young adults with recently stabilized psychiatric disorders and diminishing the stigmatization experienced by individuals with mental health problems. This program was designed by a team of clinicians from Ste-Justine Hospital’s psychiatry unit, in collaboration with professional artists. It is delivered since the spring of 2009. The evaluation of program ET was conducted in an independent yet collaborative manner, using a naturalistic approach and mixed methods. Data were collected before, during, and after the program from all participants who took part in two distinct sessions (N = 24), as well as from the two artist instructors, the psychoeducator who provided clinical support during the workshops, and the clinicians who referred participants (N=11). Individual and group semi-structured interviews, direct observations, and questionnaires served to collect the necessary data. Quantitative analyses of variance and qualitative thematic analyses were conducted and their results were interpreted in an integrated manner. The first empirical chapter of the thesis reports on the evaluation of the implementation of program ET. This evaluation showed that the program reached participants closely corresponding to its targeted population and that these youths globally presented elevated levels of assiduity, engagement in, and appreciation of the program. This evaluation also allowed to conclude that most essential program components were delivered as intended by designers and were largely appreciated and judged pertinent by participants and other respondents. The second empirical chapter presents an article detailing the evaluation of the effects of the program ET. This evaluation focused on three of its intermediate objectives: the improvement of 1) global functioning, 2) social comfort and competence, and 3) self-perception in program participants suffering from mental disorders. Results of this study revealed significant and positive changes in the global functioning of participants following the program, as assessed by their referring clinicians and partially corroborated by participants themselves. Improvements in social comfort and competence and self-perception were also noted in a substantial proportion of participants, although they did not translate in significant differences detected by group-level quantitative analyses. The third and last empirical chapter of the thesis reports on a study which sought to explore the mechanisms underlying the effects of program ET and to inductively elaborate a theory of action of the program. This qualitative investigation revealed four categories of mechanisms: 1) social experience and gratification, 2) desensitization by gradual exposure, 3) success and valorization, and 4) normalization. This study also allowed to identify program characteristics and components that seemed to favor the onset or impact of these mechanisms, such as the non-clinical setting and conducting of the program, the heterogeneous composition of the groups, the use of performing arts, and the pursuit of a collectively achieved artistic product and its presentation to an external public. Globally, the studies presented in the thesis support the relevance and promising character of program ET, while suggesting potential improvements for future versions. By extension, the thesis supports the potential efficacy of rehabilitation strategies which include components analogous to those of ET in fostering the recovery of youths with mental disorders.

Der volle Inhalt der QuelleIn 2005, in the wake of the adoption of the Youth Criminal Justice Act (YCJA) and its attendant restrictions on incarceration, the Centre jeunesse de Montréal - Institut universitaire (CJM-IU) [Montreal youth centre - University institute] launched a community-oriented intensive differential case management program (IDCMP). The clinical internship reported here had the following objectives: allow participation in the cross-sectional implementation of the program, improve the program’s robustness, support program specialists, and develop an assessment tool that would improve this program’s referral process. The results of this internship indicated the necessity of reviewing the IDCMP’s implementation process, specifically through constructive, pedagogical assessment of the congruence of the plan’s theoretical model and day-to-day application, as well of the obstacles encountered. A qualitative methodology appeared appropriate for these purposes. Two techniques were used: field participant observation, and content analysis of written documents (committee minutes). Chen's Program Action Model and the Texas Christian University (TCU) Program Change Model provided the conceptual framework for the internship report. The former’s six components were the basis for an assessment checklist and oriented the global overview of the implementation of the IDCMP, while the latter provided a model of the main stages of change and the individual, organizational, and program-specific factors that favour or hinder innovation. The results indicate that overcoming obstacles to the application of the IDCMP would not require any significant changes to the program’s underlying theoretical basis. Modifications of the program’s implementation process are presented. In conclusion, we consider that the successful implementation of the IDCMP is dependent on certain conditions, and will always be subject to constraints.
