Around 1992 Navstar/GPS will become fully operational. Public access is then provided in the Standard Precision Service, SPS, at a reduced accuracy of 100 metres for 95 per cent of the fixes. The exclusively US military-operated system and the deliberately introduced degradation (SA, selective availability) of the attainable accuracy have some drawbacks for European navigation. Neither the probability density function of the error amplitude, nor its power spectral density function is known. So, using Navstar/GPS as a sole-means precise and reliable navigational aid for high-risk transports is not possible.

The purpose of this article is to clarify the scientific basis of physical fitness assessment in children and to review the European efforts to develop a EUROFIT fitness test battery for the youth in the countries of the Council of Europe. The development of EUROFIT is based on the efforts made in the United States in the 1950s and in Europe in the 1980s. Physical fitness measurement is not identical to physiological measurement: The EUROFIT tests are aimed at measuring abilities rather than skills. Correlations between physical fitness tests and physiological laboratory tests show varying results and, therefore, need to be continued. Reliability of fitness tests needs to be continually studied. Because of the multipurposes of physical fitness testing, EUROFIT norm- and criterion-referenced scales for EUROFIT have to be developed. Examples of scaling methods are given. Implementation of the EUROFIT fitness tests for educational purposes is urgently needed.

This paper, and the following six papers, were presented during the NAV 98 Conference held at Church House, Westminster, London on 9th and 10th December 1998. A full listing of the Conference, and how to obtain a copy of the proceedings, is shown on Page 300.The existing Loran-C and Chayka infrastructure can, with some minor changes, become a very powerful augmentation system for GNSS (GPS, GLONASS and the future Galileo). Delft University initially proposed the Eurofix concept in 1989. Although the necessary modification to the LF navigation systems are minimal, the GNSS user may get significant benefits from the Eurofix signals in terms of accuracy, integrity and availability. The reason is the high signal structure, signal propagation, and the operations dissimilarity of both systems. The broadcast correction and integrity data improves GNSS accuracy down to the metre level. In this way, the measured Loran-C and Chayka ranges are continuously updated. Thus, in the case of GNSS signal interruptions, highly calibrated Loran-C/Chayka may take over the navigation function. Tests carried out in Europe at the Loran-C station at Sylt (Germany) drew large international attention, leading to further tests in the USA by the US Coast Guard in 1998. Recently, a Dutch–Russian consortium implemented Eurofix on the Chayka transmitter at Bryansk (Russia) which is now successfully broadcasting DGPS as well as DGLONASS correction data. This paper highlights some on-air Eurofix DGPS performance experiments carried out in Europe and the USA. With all the European Loran-C and Chayka transmitters modified, Eurofix can be used all over the European continent. As multiple stations can normally be received simultaneously, the user may locally apply networked DGNSS, which may reduce spatial decorrelation effects significantly. Post- processed results of this Regional Area Augmentation System are presented.

Abstract Background HCV genotype 1a (HCV g1a) followed by HCV g1b, –g2, and –g3 are the most common etiologic agents in the ~3 million current HCV-infections in the US. To achieve effective therapy, antiviral drug resistance testing is often essential but not fully available. Knowledge of both the genotype and the presence of HCV mutations in the genes of the major drug targets (NS3, NS5A and NS5B) can assist in optimal treatment selection. Methods The HCV genotype of 1000 HCV positive clinical plasmas and sera was determined (HCVg Direct, GenMark). Ten independent Sanger sequencing assays detecting antiviral drug resistance mutations in the genes encoding NS3, NS5a, and NS5b were developed. Six of these assays address mutations in all three genes in the two most common genotypes (HCV g1a and g1b). In addition, four more assays address mutations in NS5a and NS5b of HCV g2 and g3. These mutations are resistance determinants against 11 anti-HCV drugs as shown in Figure 1. A streamlined workflow employs conventional reverse transcriptase PCR, gel electrophoresis, spectrophotometry, bi-directional Sanger sequencing and reporting. The assays were designed to cover hot spot regions and capturing all known resistance mutations in NS3, NS5a and NS5b. Results Consistent with previous US HCV incidence reports, g1a, g1b, g2, and g3 comprised 99% of 1000 sequentially tested HCV patient specimens (62%, 12%, 11%, and 14%, respectively). Testing of more than 20 clinical samples each for g1a, g1b, g2, and g3 resulted in successful detection of NS3, NS5a, and NS5b mutations that confer drug resistance. The design successfully permitted detection of relevant mutations known to date for all 11 drugs. The number of reportable mutations range from 20 ‒ 36, 9 ‒ 49, and 10 ‒ 29 for the NS3, NS5a, and NS5b inhibitors, respectively (Figure 1). Conclusion These assays provide the most comprehensive commercially available antiviral drug resistance information to date for mutations in HCV NS3, NS5A, and NS5B. This testing will assist physicians in deciding on the most appropriate treatment options for their patients. Disclosures J. Nutt, Viracor Eurofins Laboratories: Employee, Salary;

The purpose of this study was to examine the rest-retest reliability of the Eurofit motor fitness tests performed by university students. A total of 98 undergraduate students who were enrolled in physical education departments in Greece participated (29 men aged 19.5 ± 2.7 hr. and 66 women aged 19, 4 ± 2.7 yr.). ALL Eurofit motor fitness tests and anthropometric measurements were obtained twice with one week between the two measurements, Intraclass correlation coefficient indicated satisfactory coefficients above .70 for most tests. The only exception was the plate-tapping test which yielded a low value ( R = .57). Further, the majority of the Eurofit test battery fitted well within the 95% confidence interval, and only three Eurofit motor fitness test items (flamingo balance, plate tapping, and sit-ups) presented a confidence limit below the value of .70, These findings indicated that the Eurofit test battery yielded reliable data for undergraduate students. However, modifications should be considered to improve the reliability of certain test items, for application to undergraduates.

Abstract Background Congenital cytomegalovirus (cCMV) infection is a significant, but potentially under-recognized health threat. Approximately 1 of 150 neonates in the US is born with cCMV infection, with 20% exhibiting long-term health problems due to infection. Both targeted CMV testing of newborns with failed hearing screens and universal CMV screening of all newborns have been proposed as approaches to identify infected newborns early in life. Congenital CMV infection can be diagnosed by testing a newborn’s saliva, urine, or blood by CMV qPCR or culture. Dried blood spots for use in qPCR assays have been shown to be a minimally sensitive specimen. Of the three specimens that are recommended, only saliva is simple, noninvasive and easy to collect. Methods In this study, we have validated a real-time (TaqMan) PCR assay for use in testing saliva samples from neonates for the presence of CMV. In conjunction with compatible clinical findings, a CMV positive PCR result forms the basis for a clinical diagnosis. The assay was shown to be specific for CMV, with no cross-reactivity detected for other human herpesviruses or for other human viral pathogens. Since CMV shedding levels from cCMV cases are known to be above the analytical limit of detection for the assay, and samples are collected in a nonsterile environment in which incidental CMV shedding may be present from other neonatal or pediatric patients, the reporting cutoff for this assay was set at 1000 IU/mL. Following analytical validation of the assay, stored (-80°C) residual de-identified clinical saliva samples were tested. The comparator assay was CMV cell culture, and the clinical diagnosis was used to resolve discrepant results. Results A total of 9 saliva samples, collected at approx. 1 month of age (or earlier) were tested by both assays. Two samples were negative by both assays and 6 samples were positive by both assays. A single sample was positive by qPCR but negative by cell culture; the qPCR value for this sample was 15,100 IU/mL. This infant had two positive urine cultures, a positive saliva shell vial culture and was clinically confirmed to have cCMV infection. Conclusion This study suggests improved sensitivity of qPCR over CMV cell culture for identification of neonates congenitally infected with CMV. Disclosures M. Eurofix is an integrated navigation system, which combines Differential GNSS and Loran-C. The Loran-C system is used to provide differential corrections for the GNSS pseudo-range measurements and integrity information by additionally modulating the transmitted signals. This modulation, however, is not allowed to degrade normal Loran-C operations. As data transmission is hindered by the aggressive Loran-C signal environment, special modulation patterns and Forward Error Correcting codes are designed to fulfil the requirements. Compared to other DGNSS services, Eurofix has a number of advantages: It covers a large area, its implementation in an already existing infrastructure is basically low-cost, it offers enhanced datalink availability in urban and mountainous environment, it has wide-area capabilities and, finally, it provides a free backup navigation system in case either Loran-C or GPS fails.The paper describes the Eurofix system, focusing on the Loran-C data channel. A modified RTCM type-9 message format is used with special Forward Error Correction. At Delft University a real-life test set-up has been built to evaluate the Eurofix performance for different scenarios. The results on datalink and DGNSS performance are presented. It is shown that metre-level DGNSS performance is achievable with low bit rate data channels, using Loran-C stations at up to 1000 km distance.

Physical conditioning is crucial in building motion skills if the coach wants to ensure long-term athletic development. The essence is that physical conditioning needs to be developed before the development of technical skills of sports movements at a much higher intensity at each stage of the multi-annual training process. The adult competition system and training programs are being imposed on young athletes, so the essential motion skills are not being learned (Grāvītis @ Luika, 2015). The lack of physical conditioning in teenage years is very often reflected in the most inappropriate moments of an athlete's career, at the elite level. Insufficient physical conditioning is a reason for the instability of motion skills and injuries in extreme physical and psychological loads. The amount of physical activity can be measured as a step count. The aim of the research is to assess physical fitness rate for children practicing basketball, football or ice hockey with EUROFIT test battery. The study included boys, 100 basketball players, 100 soccer players and 100 ice hockey players from Latvia who were born in 2002 and started training in their sport during at the ages from 7 to 9, collected data of their conditioning for five years, divided into three age groups. According to EUROFIT tests, it was concluded that the ice hockey players have the highest physical conditioning rates according to EUROFIT standard from the athletes analyzed, and the basketball players have the lowest physical conditioning rates in EUROFIT tests from the athletes analyzed. We can conclude that the results of hockey players are higher than the scores of basketball and soccer players because ice hockey players practice more frequently, and dryland training is strictly organized, they develop all motor abilities that is needed for successive training in basic drills on ice.

Abstract Background Cell-free DNA (cfDNA) has emerged as an important clinical specimen to probe for pathogenic microbes, especially in organ transplant patients where the same data can be used to predict allograft rejection. Recent reports described viral, bacterial or the complete microbial diversity in plasma following cfDNA sequencing. The prevalence of certain viral families (anelloviridae) is associated with immunosuppressant dosage and the risk of antibody mediated rejection. While being informative, the cfDNA reads are inherently shorter in length (~160bp or 2x75bp) and predominated by the host DNA (~97-99%), causing challenges in their taxonomic annotation and lower specificity. Here we present a computational protocol which minimizes these challenges by merging the concept of “Reference-assisted Assembly” with K-mer profiles of NGS data, for highly sensitive and specific microbial detection. Methods We developed a pipeline in which non-host NGS data (reads not mapped to the human genome) undergo a reference-assisted assembly operation and then taxonomic annotation using KrakenUneq (a K-mer based classifier). We trained the KrakenUneq on an in-house and curated database of ~12,000 viral genomes. We used three different K-mer values (16, 21, 31) to train KrakenUneq, and final predictions are made by applying a majority-wins rule. Currently the default KrakenUneq database is used for bacterial & fungal metagenome analysis. We tested our method on 30 simulated and 124 clinical samples obtained from a biorepository. Results Our protocol currently screens for a targeted list of pathogens (15 viral species, 16 bacterial and 10 fungal genera). On a simulated set of viral sample mixes, our protocol had 100% accuracy. For 124 clinical samples, predictions were evaluated for specificity and sensitivity using qPCR assays for the following viral species: EBV, BKV, JCV, HSV1/2, HHV7, and CMV. Total 33/38 computational predictions (87%) were confirmed by qPCR. The prediction sensitivity in terms of cps/ml ranged from 6 - 106 copies/mL. Conclusion Our efforts to perform ‘Reference-assisted assembly’ followed by K-mer based taxonomic annotation of cfDNA data, led to development of a novel and accurate pathogen detection protocol.

The age of adolescence (11–17 years of age) is characterized by the most intensive development of all organism functions and systems. The greatest heterochronicity of physical development and different functional systems of an organism is noted in this period. Therefore, it is of top significance to explore the changes of learners‘ physical and functional fitness at this age and determine tentative referential scales of their indicators. The aim of the research is to investigate physical development of teenagers (boys) of different age groups, their physical and functional fitness, as well as to design referential scales of the obtained indicators. The object of the research is physical development as well as physical and functional fitness of 5–10th form teenage boys (11–17 year old). Physical development (height (cm), weight (kg)) and physical fitness of the boys (205 boys of 5–10th forms) were determined in terms of EUROFIT fitness tests (2002). The results of physical development research and their comparative analysis showed that a substantial growth in height and weight was observed in forms 7 and 9. The height of 5–10 form boys changed insignificantly throughout the decade and approximated the average: however, the average of the body mass considerably increased and was higher than provided in EUROFIT (2002) or designed height and mass charts of child growth (Tutkuvienė, 1995). Hence, it is assumed that such an increase in the aforesaid indicator results was determined by the processes incurred in the period of the most intensive development. The research findings in physical fitness demonstrated that the results of 10 × 5 meter Shuttle Run most significantly increased in form 8 and complied with the results achieved by boys, who were two years older. However, later the results were stabilized and changed insignificantly, whereas the dissemination of results was rather low. Explosive leg power (Standing Broad Jump) and the results of hand dynamometry increased in form 7. The results of both explosive and static hand power were similar or insignificantly higher in comparison with the ones of EUROFIT (2002). The results of the endurance of abdominal muscles (Sit-ups test) improved in forms 7 and 9; the results in all surveyed forms appeared to exceed or equal the ones of EUROFIT (2002). The boys’ balance (Flamingo Balance test) and flexibility (Sit- and-Reach test) decreased in form 7 and increased in form 9. However, the research results showed that the data on the balance and flexibility of the surveyed boys approximated or insignificantly exceeded the ones provided in EUROFIT (2002). The greatest increase in the test results of physical development and fitness were observed in forms 7 and 9. However, these results approximated or insignificantly surpassed the results of EUROFIT (2002), with an exception of the growth in body mass and improvement in balance and shuttle run. Presumably, such results were obtained due to the most intensive puberty processes and their conditioned discrepancies, such as acceleration and retardation outcomes, between biological and calendar age. The investigated physical development, physical and functional fitness, their changes in the period of adolescence, as well as the design referential scales enable teachers and sports couches to accurately assess physical and functional conditions, as well as improve the process of (self-) development in pursuance for greater results of physical education.

The purpose was to examine the reliability of physical fitness items from the Eurofit Test Battery for adolescent males with mild mental retardation (MMR; n = 63, mean IQ = 63.0 ±11.5, mean age = 15.5 ± 1.2) and those without (n = 22, mean age = 15.6 ± 0.6). Males with MMR scored significantly lower (p ≤ .005) than those without on all items except sum of skinfolds, height, and weight. Intraclass correlations (ICCs) ranged from .94 to .99 for males with MMR and .85 to .99 for those without. Percentage error of the mean for all items ranged from 0.5 to 47.5% for participants with MMR and 0.4 to 32.2% for those without. ICCs indicated that Eurofit physical fitness test items are reliable measures for males with and without MMR. However, the percentage error of the mean is quite large for sit-and-reach and 20-m shuttle test (20-MST) items.

ObjectiveTo develop sex-specific and age-specific normative values for the nine Eurofit tests in European children and adolescents aged 9–17 years.MethodsA systematic review was undertaken to identify papers that explicitly reported descriptive results for at least one of nine Eurofit tests (measuring balance, muscular strength, muscular endurance, muscular power, flexibility, speed, speed-agility and cardiorespiratory fitness (CRF)) on children and adolescents. Data were included on apparently healthy (free from known disease/injury) children and adolescents aged 9–17 years. Following harmonisation for methodological variation where appropriate, pseudodata were generated using Monte Carlo simulation, with population-weighted sex-specific and age-specific normative centiles generated using the Lambda Mu Sigma (LMS) method. Sex-specific and age-specific differences were expressed as standardised differences in means, with the percentage of children and adolescents with healthy CRF estimated at the sex-age level.ResultsNorms were displayed as tabulated centiles and as smoothed centile curves for the nine Eurofit tests. The final dataset included 2 779 165 results on children and adolescents from 30 European countries, extracted from 98 studies. On average, 78% of boys (95% CI 72% to 85%) and 83% of girls (95% CI 71% to 96%) met the standards for healthy CRF, with the percentage meeting the standards decreasing with age. Boys performed substantially (standardised differences >0.2) better than girls on muscular strength, muscular power, muscular endurance, speed-agility and CRF tests, but worse on the flexibility test. Physical fitness generally improved at a faster rate in boys than in girls, especially during the teenage years.ConclusionThis study provides the largest and most geographically representative sex-specific and age-specific European normative values for children and adolescents, which have utility for health and fitness screening, profiling, monitoring and surveillance.

Material and Method. This is a prospective, cross-sectional, and correlational study with a probabilistic sampling in which 150 teenagers from three different high schools from the city of Toluca, Mexico, aged 15–17, were assessed. Objective. To determine if weight, age, and gender have an influence on physical fitness evaluated with the EUROFIT and ALPHA-FITNESS batteries. Results. Women have a higher overweight and obesity rate than men (3 : 1). Adolescents who have normal weight have regular physical fitness (74.9%). When comparing genders we found that men have a higher mean than women in the tests, except for skinfold thickness and waist circumference. Age was only correlated with the plate tapping test (p=0.001). There are significant differences in the standing broad jump test and the Course-Navette of the EUROFIT and ALPHA-FITNESS batteries (p=0.000). Conclusions. It is likely that regular physical activity, and not normal weight, helps generate healthy physical fitness. Male subjects had a higher mean than women, reporting a better physical fitness and more frequent physical activity.

This article is a contribution to the experience of testing motor fitness and exploring the EUROFIT test in young children. In the age group 5–7 years, the motor fitness tests showed strong dependency on age and a small dependence on sex. Body weight and height did not appear to have any impact on the test variables for this age group. The reliability test showed significant difference between test and retest in the plate tapping test only. The reproducibility was low in bent arm hang and flamingo balance, with coefficients of variation of 67%. Modest validity of the flamingo balance test and the standing broad jump test was confirmed with correlations of 0.43 and 0.52, respectively, by laboratory testing on a force platform. Factor analyses extracted 3 components, which explained 62% of the total variance, but no single component could explain general motor fitness. The EUROFIT Motor Fitness Test appeared to be applicable also in young children, but the reproducibility of two test items was questionable. Modification of test items was suggested to fit this age group.

Purpose: To examine the effect of growth hormone (GH) treatment on physical performance in children with idiopathic short stature and normal GH secretion. Materials and Methods: A total of 24 children participated in the study (13 GH-treated, 11 non-treated, aged 8–13 y, 11 males and 13 females, Tanner stage 1–2). Participants performed a battery of motor skill performance tests (Eurofit), as well as the Wingate anaerobic test. Results: No statistically significant differences in any of the Eurofit physical fitness test results (eg, 20-m shuttle run 33.0 [15.1] vs 25.1 [21.0] laps in treated and nontreated participants, respectively, P = .25) or the Wingate anaerobic test were found between the groups (eg, peak power 5.0 [2.9] vs 3.9 [2.6] watts/kg in treated and nontreated participants, respectively, P = .2). Conclusions: Therapeutic usage of exogenous GH for pre and early pubertal children with idiopathic short stature and normal GH secretion was not associated with beneficial effects on physical performance indices. This suggests that the use of GH as a potential performance enhancing agent, in this age group, at least at commonly used doses, is not advantageous.

The 10 × 5-m shuttle run was replaced by a 5 × 10-m shuttle run. Intraclass correlations ranged from .63 to .91, indicating moderate-to-excellent reliability. However, systematic differences between test and retest were found for the sit-and-reach, bent-arm hang, and the modified 5 × 10-m shuttle run items. The results indicate that for most items, test-retest reliability was satisfactory, but that improvements need to be made to the test protocols of the sit-and-reach, bent-arm hang, and the 5 × 10-m shuttle run items to ensure test-retest reliability.
