Der volle Inhalt der QuelleNowadays, bioelectronics becomes a discipline with prominent biomedical and pharmacological potential. Investigations reported in this manuscript are focused on the use of label-free electrochemical and optical biosensors for the study of bioaffinity and biocatalytic interactions of some small xenobiotics (odorants and steroid glycoalkaloids) with immobilized macromolecules, namely, G protein-coupled human olfactory receptor OR 17-40 and cholinesterases from human and equine serum. The butyryl cholinesterases immobilized on pH-sensitive field-effect transistors follow Michaelis kinetics of hydrolysis of their substrates. Glycoalkaloids α-solanine, α-chaconine and tomatine inhibit the equine enzyme reversibly and competitively while for the human cholinesterase a mixed mode of reversible inhibition is suggested. α-Chaconine is the most potent inhibitor of both enzymes. The affinity interactions of glycoalkaloids and equine butyryl cholinesterase have been probed with electrochemical impedance spectroscopy. The absence of enzymatic substrate can significantly improve label-free detection of weak and competitive inhibitors (α-solanine). Olfactory receptor OR 17-40 has been successfully employed as odorant-recognition part of impedimetric and surface plasmon resonance-based platforms. Possibility of direct monitoring molecular events trigeered by agonist-stimulated receptor is the first step towards the "biolectronic nose"

Der volle Inhalt der QuelleBacterial biofilms are in the background of many industrial, health and domestic adverse effects with economic losses leading to intensive research to design solutions to combat their formation and development. Two routes are commonly used to solve these issues, one aiming at preventing the adhesion of bacteria and the other at inhibiting and killing adhered microorganisms. Recent work has been done in that direction with the use of polymer-based antifouling or antibacterial surfaces acting either by contact effect or continuous release of bacterial substances. Besides, responsive surfaces to various stimuli (microenvironment, light, acoustic waves, etc.) have been developed to release biocides in a controlled way. In addition, the use of an electrical potential (bioelectric effect) has aroused interest to disrupt biofilms but remains underexploited. The present work focuses on the development of biocompatible electrostimulable surfaces based on either (i) self-assembled monolayers (SAMs) deposited on titanium or gold electrodes, or (ii) functionalized conductive polymers (CPs), to prevent adhesion and/or kill bacteria. Phosphonium-based SAMs have demonstrated interesting antibacterial properties, without release of biocidal agent, against Gram positive and negative strains (S. aureus, K. pneumoniae). In addition, thin films of homo- and copolymers of phosphonium-based pyrrole CPs were obtained by electropolymerization, and analyzed by modifying and applying a set of parameters and tested as antibacterial coatings. Lastly, hydrophilic phosphoniums have been synthesized and preliminary studies have highlighted their potential use as nanocarriers for drug delivery

Der volle Inhalt der QuelleUne brève description des milieux biologiques permet d'abord de souligner les difficultés liées à leur caractérisation électrique. Les différentes méthodes de caractérisation existantes sont présentées en mettant en valeur leurs particularités dans les domaines fréquentiels où elles s'appliquent. Les valeurs de permittivité et de conductivité données dans la littérature sont présentées pour montrer leurs spécificités et le manque de connaissance sur leur précision. Une méthode de caractérisation classique par mesure d'impédance à 4 électrodes est ensuite étudiée en détail entre 10Hz et 10MHz. Le système de mesure réalisé est testé sur des solutions ioniques pour en établir des modèles complets prenant en compte les éléments pouvant affecter la précision des résultats. Ces modèles sont utilisés pour estimer l'erreur de mesure et pour optimiser la méthode de caractérisation à 10Hz et 10MHz.
Les différentes méthodes numériques permettant la modélisation des problèmes de calcul de champs induits dans le corps humain sont ensuite présentées. Les difficultés de la modélisation de l'exposition de l'homme aux champs électromagnétiques sont explicitées. Elles sont dues aux propriétés électriques et géométriques particulières du corps et à la diversité des sources de rayonnement. Les différentes formulations et conditions aux frontières du domaine de calcul spécifiquement applicables au problème sont détaillées. En utilisant la méthode des éléments finis et en supposant les propriétés électriques du corps connues, différents modèles (maillage, formulation, conditions aux frontières) sont enfin étudiés. Leurs performances sont évaluées sur des problèmes canoniques 2D et 3D représentatifs. Les résultats permettent d'évaluer l'erreur sur calcul des phénomènes induits et de faciliter le choix d'une modélisation adaptée sur la gamme de fréquence 10Hz-1GHz.
Le travail proposé vise principalement à donner des arguments pour justifier de la pertinence des résultats des études de l'interaction champs électromagnétiques/corps humain. Il peut aussi trouver des applications directes dans la caractérisation précise des milieux biologiques et dans l'établissement de nouvelles normes sur l'exposition des hommes aux champs électromagnétiques.

Der volle Inhalt der QuelleThis work is part of a process aiming at evaluating and improving the performances of existing tools for the study of the interactions between the electromagnetic fields and the human body. The tools include characterisation methods to measure the macroscopic electrical properties of biological media, and numerical methods allowing to model and compute the fields induced by electromagnetic sources in the human body. The main properties of biological tissues are first described in order to explain the difficulties of their electrical characterisation. The existing characterisation methods are presented. Their specificities and operating frequency range are emphasized. Permittivity and conductivity values found in the literature are used to show the main parameters they depend on and the lack of knowledge on their accuracy. A usual characterisation method based on four-electrode impedance measurements is then studied between 10Hz and 10MHz. The measurement system is developed and tested on ionic solutions in order to build reliable and complete models. These models are used to estimate the accuracy of the measure and to optimize the characterisation method at 10Hz and 10MHz. Various numerical methods that allow modelling and computing induced electromagnetic phenomena in the body are then presented. The difficulties of the modelling are explained and linked to the unusual electrical and geometrical properties of the human body, and to the diversity of the sources of electromagnetic fields. Formulations and boundary conditions especially developed for the problem are described. Using the finite element method, and assuming the electrical properties of the body are known, various models (mesh, formulation, boundary conditions) are studied. Their efficiency is evaluated by simulating 2D and 3D representative canonical problems. The results allow estimating the accuracy of the computation and choosing the more efficient modelling for each frequency between 10Hz and 1GHz. The work gives arguments which can be used to prove the consistency of the results when studying the interaction between the electromagnetic fields and the human body. It has also direct applications in the accurate characterisation of biological media and in the establishment of new norms on human exposure to electromagnetic fields
