Endothelial cells (ECs) respond to concurrent stimulation by biochemical factors and wall shear stress (SS) exerted by blood flow. Disruptions in flow-induced responses can result in remodeling issues and cardiovascular diseases, but the detailed mechanisms linking flow-mechanical cues and biochemical signaling remain unclear. Activin receptor-like kinase 1 (ALK1) integrates SS and ALK1-ligand cues in ECs; ALK1 mutations cause hereditary hemorrhagic telangiectasia (HHT), marked by arteriovenous malformation (AVM) development. However, the mechanistic underpinnings of ALK1 signaling modulation by fluid flow and the link to AVMs remain uncertain. We recorded EC responses under varying SS magnitudes and ALK1 ligand concentrations by assaying pSMAD1/5/9 nuclear localization using a custom multi-SS microfluidic device and a custom image analysis pipeline. We extended the previously reported synergy between SS and BMP9 to include BMP10 and BMP9/10. Moreover, we demonstrated that this synergy is effective even at extremely low SS magnitudes (0.4 dyn/cm2) and ALK1 ligand range (femtogram/mL). The synergistic response to ALK1 ligands and SS requires the kinase activity of ALK1. Moreover, ALK1’s basal activity and response to minimal ligand levels depend on endocytosis, distinct from cell–cell junctions, cytoskeleton-mediated mechanosensing, or cholesterol-enriched microdomains. However, an in-depth analysis of ALK1 receptor trafficking’s molecular mechanisms requires further investigation.

Sewage sludge (SS) is derived from wastewater treatment plants and can be used as a biofertilizer when properly stabilized. This work aimed to evaluate SS application for agricultural production improvement. SS was tested on Porto Santo Island (Portugal). The experiment was randomly designed with three 25 m2 plots for each treatment (2 SS concentrations + control without SS) and performed in two consecutive cycles. For the first cycle, dehydrated sludge was mixed with soil, obtaining final concentrations of 0.8 kg/m2 (C1) and 1.6 kg/m2 (C2). Half of the concentration was used for the second cycle. Fifty-eight sweet potato plants were used in each plot. SS application boosted the agronomic parameters of biomass, productivity, and shoot biomass. Furthermore, improvements in soil properties were observed, mainly for pH, CEC, and NO3-N, with no significant increase in heavy metals. For the edible parts, heavy metal concentrations decreased, and Pb was the only one that still exceeded the maximum limits. The results demonstrated that SS application to low-fertility soil is effective in improving the agronomic parameters of sweet potato and enhancing soil features. Further studies considering other variables, i.e., SS origin, soil properties, and the crop, must be carried out to propose custom applications.

The goal of the present study was to quantify and correlate the contribution of the cytosolic p67 phox subunit of NADPH oxidase 2 to mitochondrial oxidative stress in the kidneys of the Dahl salt-sensitive (SS) hypertensive rat. Whole kidney redox states were uniquely assessed using a custom-designed optical fluorescence three-dimensional cryoimager to acquire multichannel signals of the intrinsic fluorophores NADH and FAD. SS rats were compared with SS rats in which the cytosolic subunit p67 phox was rendered functionally inactive by zinc finger nuclease mutation of the gene (SSp67 phox-null rats). Kidneys of SS rats fed a 0.4% NaCl diet exhibited significantly ( P = 0.023) lower tissue redox ratio (NADH/FAD; 1.42 ± 0.06, n = 5) than SSp67 phox-null rats (1.64 ± 0.07, n = 5), indicating reduced levels of mitochondrial electron transport chain metabolic activity and enhanced oxidative stress in SS rats. When fed a 4.0% salt diet for 21 days, both strains exhibited significantly lower tissue redox ratios ( P < 0.001; SS rats: 1.03 ± 0.05, n = 9, vs. SSp67 phox-null rats: 1.46 ± 0.04, n = 7) than when fed a 0.4% salt, but the ratio was still significantly higher in SSp67 phox rats at the same salt level as SS rats. These results are consistent with results from previous studies that found elevated medullary interstitial fluid concentrations of superoxide and H2O2 in the medulla of SS rats. We conclude that the p67 phox subunit of NADPH oxidase 2 plays an important role in the excess production of ROS from mitochondria in the renal medulla of the SS rat.

In this review, strategies for improving the antimicrobial properties of stainless steel (SS) are presented. The main focus given is to present current strategies for surface modification of SS, which alter surface characteristics in terms of surface chemistry, topography and wettability/surface charge, without influencing the bulk attributes of the material. As SS exhibits excellent mechanical properties and satisfactory biocompatibility, it is one of the most frequently used materials in medical applications. It is widely used as a material for fabricating orthopedic prosthesis, cardiovascular stents/valves and recently also for three dimensional (3D) printing of custom made implants. Despite its good mechanical properties, SS lacks desired biofunctionality, which makes it prone to bacterial adhesion and biofilm formation. Due to increased resistance of bacteria to antibiotics, it is imperative to achieve antibacterial properties of implants. Thus, many different approaches were proposed and are discussed herein. Emphasis is given on novel approaches based on treatment with highly reactive plasma, which may alter SS topography, chemistry and wettability under appropriate treatment conditions. This review aims to present and critically discuss different approaches and propose novel possibilities for surface modification of SS by using highly reactive gaseous plasma in order to obtain a desired biological response.

Hypertension is a classic example of a complex polygenic trait, impacted by quantitative trait loci (QTL) containing candidate genes thought to be responsible for blood pressure (BP) control in mammals. One such mapped locus is on rat chromosome 9, wherein the proof for a positional candidate gene, regulated endocrine-specific protein-18 ( Resp18) is currently inadequate. To ascertain the status of Resp18 as a BP QTL, a custom targeted gene disruption model of Resp18 was developed on the Dahl salt-sensitive (SS) background. As a result of this zinc-finger nuclease (ZFN)-mediated disruption, a 7 bp deletion occurred within exon 3 of the Resp18 locus. Targeted disruption of Resp18 gene locus in SS rats decreases its gene expression in both heart and kidney tissues regardless of their dietary salt level. Under a high-salt dietary regimen, both systolic and diastolic BP of Resp18mutant rats were significantly increased compared with SS rats. Resp18mutant rats demonstrated increased renal damage, as evidenced by higher proteinuria and increased renal fibrosis compared with SS rats. Furthermore, under a high-salt diet regimen, the mean survival time of Resp18mutant rats was significantly reduced compared with SS rats. These findings serve as evidence in support of Resp18 as a gene associated with the development of hypertension and renal disease.

Accurate identification of strand residues aids prediction and analysis of numerous structural and functional aspects of proteins. We propose a sequence-based predictor, BETArPRED, which improves prediction of strand residues and β-strand segments. BETArPRED uses a novel design that accepts strand residues predicted by SSpro and predicts the remaining positions utilizing a logistic regression classifier with nine custom-designed features. These are derived from the primary sequence, the secondary structure (SS) predicted by SSpro, PSIPRED and SPINE, and residue depth as predicted by RDpred. Our features utilize certain local (window-based) patterns in the predicted SS and combine information about the predicted SS and residue depth. BETArPRED is evaluated on 432 sequences that share low identity with the training chains, and on the CASP8 dataset. We compare BETArPRED with seven modern SS predictors, and the top-performing automated structure predictor in CASP8, the ZHANG-server. BETArPRED provides statistically significant improvements over each of the SS predictors; it improves prediction of strand residues and β-strands, and it finds β-strands that were missed by the other methods. When compared with the ZHANG-server, we improve predictions of strand segments and predict more actual strand residues, while the other predictor achieves higher rate of correct strand residue predictions when under-predicting them.

The purpose of this study is to quantitatively assess lens opacity, using a swept-source optical coherence tomography (SS-OCT) device for anterior segment assessment, and establish the correlation with Scheimpflug imaging, corrected distance visual acuity (CDVA) and cumulative dissipated energy (CDE). This prospective cross-sectional single-center study enrolled 51 patients (51 eyes) with crystalline lens opacity. Patients with previous ocular surgery, pathologies or general disorders affecting vision were excluded. Eyes were scanned with an SS-OCT device, and lens densitometry was automatically analyzed using a custom MATLAB script which examined lens density, nuclear density and linear density. The same analyses were performed on Scheimpflug images. Preoperative CDVA and CDE during phacoemulsification were recorded. Spearman’s (ρ) and Pearson’s (r) correlation coefficients were assessed according to data normality. Statistically significant correlations were established between SS-OCT and Scheimpflug imaging using lens analysis (ρ = 0.47, p < 0.001), nuclear analysis (ρ = 0.73, p < 0.001) and linear analysis (r = 0.44, p < 0.001). A significant correlation with CDE was found with all the SS-OCT methods (r = 0.57, p < 0.001). Only the nuclear analysis of the SS-OCT scans (Tb = −0.33, p < 0.01) and Pentacam Nucleus Staging (Tb = −0.26, p < 0.05) showed a statistically significant correlation with CDVA. Good inter-device agreement in lens densitometry was found. However, SS-OCT yielded improved lens imaging compared with the Scheimpflug device and a higher correlation with clinical parameters. Thus, high-resolution SS-OCT has the potential to become a preferable option for automatic cataract grading and preoperative planning.

This chapter refers to SS Sturmbannführer Hermann Höfle, the head of Operation Reinhard in Warsaw, who read out the order for displacement of Jews from the ghetto on July 22, 1942. It discusses how the order contains the task for the Jewish Order Service to serve as the executive organ of the Judenrat responsible for carrying out the deportations. It also mentions the few hundred prisoners from Gesia Street who joined the group of deportees from Dzika Street and were led into custody in front of despairing family members. The chapter notes how the Jewish Order Service did not give any explanation as to why they included children in the deportation, but it was speculated that the roundup of beggar children was intended for taking them to the Jewish prison. From the day of the deportation, the Jewish Order Service was then tasked to bring six thousand people to the Umschlagplatz every day.

Abstract This chapter deals with antisemitic murder cases in the winter and spring of 1933. Though records are fragmentary and incomplete, and contemporary authorities disputed their very existence, the surviving documentary evidence permits us to classify them into three categories: 1) the more-or-less spontaneous murder without prior planning; 2) the “shot while trying to escape” murder of people already in the custody of the SA and SS; and 3) the planned “revenge murder.” Many of these murder cases were well documented and, in several instances, the police apprehended the SA killers with sufficient evidence to convict them. Because of two amnesties for “Offenses Committed in Connection with the National Socialist Revolution” of March and July 1933, culprits evaded justice in 1933. After the war, however, several of these murder cases were reopened by friends and relatives of the victims, though severe sentences remained the exception. An American journalist estimated the number of anti-Jewish murder cases to be “at least 300” for the period between early March and mid-June 1933. The enormity of this figure becomes apparent in view of the fact that there were a mere 1,336 cases of murder and manslaughter in all of Germany in the crisis-ridden year 1931. This chapter analyzes a number of individual murder cases in all three categories and also examines different types of Nazi murderers, as well as the (often stillborn) effort to bring some of them to justice after the war. It becomes apparent that the often conjured up Rechtsstaat was already completely extinct by the spring of 1933.

Abstract This paper reports on the performance evaluation of stainless steel (SS) thermal spray coatings aimed at shielding lightweight aluminum (Al) brake rotor disks from excessive heat and providing an adequate tribological surface in contact with brake pads. Coating wear, corrosion and heat resistance performances were evaluated using pin-on-disk, cyclic corrosion tests and thermal cycling using a custom laser rig, respectively. Arc spray optimized coatings displayed lower or equivalent wear rates when compared with the baseline gray cast iron disks, with similar frictional behavior. However, arc spray coating exhibited low adhesion which limits the maximum coating thicknesses achievable and leads to early coating spalling after about 1000 thermal cycles. Arc sprayed coatings also corroded and delaminated under corrosion tests. Optimized cold spray coatings present high corrosion resistance and could resist above 10,000 thermal cycles without spalling. However, cold spray coatings exhibit wear rates at least 4 times those of the cast iron. Taking advantage of both types of coatings, it was found that the production of a duplex coating made of a cold spray bond coat and an arc spray top coat could meet the requirements for protecting Al disks, with near 50% weight reduction.
