Thus, an antifungal compound isolated fromCatharanthus roseuswas screened for its effectiveness in controlling the growth of these fungi. The antifungal compound isolated fromC. roseusextract was determined through thin layer chromatography (TLC) and nuclear magnetic resonance (NMR) analysis. EachC. roseusof the DCM extracts was marked as CRD1, CRD2, CRD3, CRD4, CRD5, CRD6, and CRD7, respectively. TLC results showed that all of theC. roseusextracts peaked with red colour at Rf = 0.61 at 366 nm wavelength, except for CRD7. The CRD4 extract was found to be the most effective againstR. microporusandG. philippiiwith inhibition zones of 3.5 and 1.9 mm, respectively, compared to that of other extracts. These extracts, however, were not effective againstP. noxius.The CRD4 extract contained ursolic acid that was detected by NMR analysis and the compound could be developed as a biocontrol agent for controllingR. microporusandG. philippii.Moreover, little or no research has been done to study the effectiveness ofC. roseusin controlling these fungi.

ABSTRACT Here we describe that the Helicobacter pylori sensor kinase produced by HP1364 and the response regulator produced by HP1365 and designated CrdS and CrdR, respectively, are both required for transcriptional induction of the H. pylori copper resistance determinant CrdA by copper ions. CrdRS-deficient mutants lacked copper induction of crdA expression and were copper sensitive. A direct role of CrdR in transcriptional regulation of crdA was confirmed by in vitro binding of CrdR to the crdA upstream region. A 21-nucleotide sequence located near the crdA promoter was shown to be required for CrdR binding.

ABSTRACT Binding of herpes simplex virus (HSV) envelope glycoprotein D (gD) to a cell surface receptor is an essential step of virus entry. We recently determined the crystal structure of gD bound to one receptor, HveA. HveA is a member of the tumor necrosis factor receptor family and contains four characteristic cysteine-rich domains (CRDs). The first two CRDs of HveA are necessary and sufficient for gD binding. The structure of the gD-HveA complex reveals that 17 amino acids in HveA CRD1 and 4 amino acids in HveA CRD2 directly contact gD. To determine the contribution of these 21 HveA residues to virus entry, we constructed forms of HveA mutated in each of these contact residues. We determined the ability of the mutant proteins to bind gD, facilitate virus entry, and form HveA oligomers. Our results point to a binding hot spot centered around HveA-Y23, a residue that protrudes into a crevice on the surface of gD. Both the hydroxyl group and phenyl group of HveA-Y23 contribute to HSV entry. Our results also suggest that an intermolecular β-sheet formed between gD and HveA residues 35 to 37 contributes to binding and that a C37-C19 disulfide bond in CRD1 is a critical component of HveA structure necessary for gD binding. The results argue that CRD2 is required for gD binding mainly to provide structural support for a gD binding site in CRD1. Only one mutant, HveA-R75A, exhibited enhanced gD binding. While some mutations influenced complex formation, the majority did not affect HSV entry, suggesting that most contact residues contribute to HveA receptor function collectively rather than individually. This structure-based dissection of the gD-HveA binding site highlights the contribution of key residues within HveA to gD binding and HSV entry and defines a target region for the design of small-molecule inhibitors.

Complex repetitive discharges (CRDs) are poorly understood phenomena in needle electromyography (EMG) recordings. The data presented here suggest that CRDs may mainly be a sign of motor unit reinnervation. EMG “video” data of 108 CRDs from neurogenic (ND, n = 39) and myogenic (MD, n = 14) disorders were retrospectively analyzed for cycle duration, potential-free time intervals, spike components (SC), maximum amplitudes, blockade, and increased jitter. CRD-SC in ND disorders (9.3 ± 7.8) outnumbered those in MD disorders (6.3 ± 6.2). The CRD cycle duration was correlated with SC and silent periods (p each < 0.000001). Blockade was observed in 36% and increased jitter in 27% of the CRDs. A higher number of CRD-SC in ND vs. MD fits the known differences in motor unit dimensions. Blockade and increased jitter are known features of diseased neuromuscular junctions, such as during reinnervation. The SC patterns of single CRD cycles resemble reinnervation potentials. Thus, CRDs may result from myo-axonal re-excitation in sprouting motor units. The purpose of this investigation was to better understand the circumstances under which CRDs may occur and eventually to contribute to the understanding of their pathogenesis.

Purpose The purpose of this paper is to investigate a new and practical decision support model of the coordinated replenishment and delivery (CRD) problem with multi-warehouse (M-CRD) to improve the performance of a supply chain. Two algorithms, tabu search-RAND (TS-RAND) and adaptive hybrid different evolution (AHDE) algorithm, are developed and compared as to the performance of each in solving the M-CRD problem. Design/methodology/approach The proposed M-CRD is more complex and practical than classical CRDs, which are non-deterministic polynomial-time hard problems. According to the structure of the M-CRD, a hybrid algorithm, TS-RAND, and AHDE are designed to solve the M-CRD. Findings Results of M-CRDs with different scales show that TS-RAND and AHDE are good candidates for handling small-scale M-CRD. TS-RAND can also find satisfactory solutions for large-scale M-CRDs. The total cost (TC) of M-CRD is apparently lower than that of a CRD with a single warehouse. Moreover, the TC is lower for the M-CRD with a larger number of optional warehouses. Practical implications The proposed M-CRD is helpful for managers to select the suitable warehouse and to decide the delivery scheduling with a coordinated replenishment policy under complex operations management situations. TS-RAND can be easily used by practitioners because of its robustness, easy implementation, and quick convergence. Originality/value Compared with the traditional CRDs with one warehouse, a better policy with lower TC can be obtained by the new M-CRD. Moreover, the proposed TS-RAND is a good candidate for solving the M-CRD.

Polycystin 2-type cation channels PKD2 and PKD2L1 interact with polycystin 1-type proteins PKD1 and PKD1L3 respectively, to form receptor–cation-channel complexes. The PKD2L1–PKD1L3 complex perceives sour taste, whereas disruption of the PKD2–PKD1 complex, responsible for mechanosensation, leads to development of ADPKD (autosomal-dominant polycystic kidney disease). Besides modulating channel activity and related signalling events, the CRDs (C-terminal regulatory domains) of PKD2 and PKD2L1 play a central role in channel oligomerization. The present study investigates the aggregation state of purified full-length PKD2L1-CRD as well as truncations of CRDs from PKD2 channels. Far- and near-UV CD spectroscopy show that the full-length PKD2L1 CRD (PKD2L1-198) and the truncated PKD2 CRD (PKD2-244) are α-helical with no β-sheet, the α-helix content agrees with sequence-based predictions, and some of its aromatic residues are in an asymmetric environment created at least by partially structured regions. Additionally, the CRD truncations exhibit an expected biochemical function by binding Ca2+ in a physiologically relevant range with Kd values of 2.8 μM for PKD2-244 and 0.51 μM for PKD2L1-198. Complimentary biophysical and biochemical techniques establish that truncations of the PKD2 and PKD2L1 CRDs are elongated molecules that assemble as trimers, and the trimeric aggregation state is independent of Ca2+ binding. Finally, we show that a common coiled-coil motif is sufficient and necessary to drive oligomerization of the PKD2 and PKD2L1 CRD truncations under study. Despite the moderate sequence identity (39%) between CRDs of PKD2 and PKD2L1, they both form trimers, implying that trimeric organization of CRDs may be true of all polycystin channels.

Several proteins other than the frizzled receptors (Fzd) and the secreted Frizzled-related proteins (sFRP) contain Fzd-type cysteine-rich domains (CRD). We have termed these domains “putative Fzd-type CRDs”, as the relevance of Wnt signalling in the majority of these is unknown; the RORs, an exception to this, are well known for mediating non-canonical Wnt signalling. In this study, we have predicted the likely binding affinity of all Wnts for all putative Fzd-type CRDs. We applied both our previously determined Wnt‒Fzd CRD binding affinity prediction model, as well as a newly devised model wherein the lipid term was forced to contribute favourably to the predicted binding energy. The results obtained from our new model indicate that certain putative Fzd CRDs are much more likely to bind Wnts, in some cases exhibiting selectivity for specific Wnts. The results of this study inform the investigation of Wnt signalling modulation beyond Fzds and sFRPs.

Gal (galectin)-8 is a tandem-repeat Gal containing N-CRDs (Nterminal carbohydrate-recognition domains) and C-CRDs (C-terminal carbohydrate-recognition domains) with differential glycan-binding specificity fused by a linker peptide. Gal-8 has two distinct effects on CD4 T-cells: at high concentrations it induces antigen-independent proliferation, whereas at low concentrations it co-stimulates antigen-specific responses. Associated Gal-8 structural requirements were dissected in the present study. Recombinant homodimers N–N (two N-terminal CRD chimaera) and C–C (two C-terminal CRD chimaera), but not single C-CRDs or N-CRDs, induced proliferation; however, single domains induced co-stimulation. These results indicate that the tandem-repeat structure was essential only for the proliferative effect, suggesting the involvement of lattice formation, whereas co-stimulation could be mediated by agonistic interactions. In both cases, C–C chimaeras displayed higher activity than Gal-8, indicating that the C-CRD was mainly involved, as was further supported by the strong inhibition of proliferation and co-stimulation in the presence of blood group B antigen, specifically recognized by this domain. Classic Gal inhibitors (lactose and thiodigalactoside) prevented proliferation but not co-stimulatory activity, which was inhibited by 3-O-β-D-galactopyranosyl-D-arabinose. Interestingly, Gal-8 induced proliferation of naïve human CD4 T-cells, varying from non- to high-responder individuals, whereas it promoted cell death of phytohaemagglutinin or CD3/CD28 pre-activated cells. The findings of the present study delineate the differential molecular requirements for Gal-8 activities on T-cells, and suggest a dual activity relying on activation state.

The Internet has opened opportunities to create world scale services. These systems require highavailability and fault tolerance, while preserving low latency. Replication is a widely adopted technique to provide these properties. Different replication techniques have been proposed through the years, but to support these properties for world scale services it is necessary to trade consistency for availability, fault-tolerance and low latency. In weak consistency models, it is necessary to deal with possible conflicts arising from concurrent updates. We propose the use of conflict free replicated data types (CRDTs) to address this issue. Cloud computing systems support world scale services, often relying on Key-Value stores for storing data. These systems partition and replicate data over multiple nodes, that can be geographically disperse over the network. For handling conflict, these systems either rely on solutions that lose updates (e.g. last-write-wins) or require application to handle concurrent updates. Additionally, these systems provide little support for transactions, a widely used abstraction for data access. In this dissertation, we present the design and implementation of SwiftCloud, a Key-CRDT store that extends a Key-Value store by incorporating CRDTs in the system’s data-model. The system provides automatic conflict resolution relying on properties of CRDTs. We also present a version of SwiftCloud that supports transactions. Unlike traditional transactional systems, transactions never abort due to write/write conflicts, as the system leverages CRDT properties to merge concurrent transactions. For implementing SwiftCloud, we have introduced a set of new techniques, including versioned CRDTs, composition of CRDTs and alternative serialization methods. The evaluation of the system, with both micro-benchmarks and the TPC-W benchmark, shows that SwiftCloud imposes little overhead over a key-value store. Allowing clients to access a datacenter close to them with SwiftCloud, can reduce latency without requiring any complex reconciliation mechanism. The experience of using SwiftCloud has shown that adapting an existing application to use SwiftCloud requires low effort.

Neste trabalho, é apresentada uma variação da técnica de espectroscopia por cavidade ressonante tipo ring-down CRDS (do acrônimo em inglês Cavity Ring-Down Spectroscopy) para a obtenção simultânea do espectro de absorção de multicomponentes numa faixa espectral larga do visível. Esta nova técnica se resume no uso do espectro supercontinuum (resultante da irradiação de meios não lineares através de lasers de femtossegundo, ou simplesmente gerada por fontes compactas) como fonte de luz para iluminar a cavidade. Neste contexto são descritas as características dos módulos para a montagem de um MC-SC-CRDS (Multicomponent Supercontinuum Cavity Ring-Down Spectroscopy): os pares de espelhos altamente refletivos, a cavidade ressonante e o sistema de detecção. Alguns problemas relacionados à excitação de multimodos, à luz difusa, ao uso efetivo do intervalo dinâmico de detecção, à baixa resolução do instrumento em resolver linhas estreitas de absorção são situados. Apresentamos os espectros de absorção de H2O (políades 4nu, 4nu + delta ) e O2 (transições proibidas de spin b-X) simultaneamente medidos por essa técnica na faixa do visível, e uma comparação com as linhas de absorção baseadas do banco de dados HITRAN é feita para demonstrar a funcionalidade deste método.
In this work, we present a variation of the technique CRDS (Cavity Ring-Down Spectroscopy) to obtain simultaneously a multicomponent absorption spectrum in a broad visible range. This new approach uses the Supercontinuum (SC) spectrum (resulting from irradiation of nonlinear media by femtosecond lasers, or simply generated by compact sources) as a light source to illuminate the cavity. In this context it is described the features of the modules assembling a MC-SC-CRDS (Multicomponent Supercontinuum Cavity Ring-Down Spectroscopy): a set of high refletivity mirrors, the resonant cavity and the detection system. Some problems related to the multimode excitation, stray light, effective use of the dynamic range of the detector, the poor resolution of the instrument to resolve narrow absorption lines are issued. We present the absorption spectra of H2O (polyads 4nu, 4nu + delta ) and O2 (spin-forbidden b-X branch) measured simultaneously by this technique in the visible range and a comparison with the absorption lines based on HITRAN database is made to demonstrate the functionality of this method.

The thrust of this research is the "MARKET-ORIENTATION OF TANZANIAN BANKING INSTITUTIONS: A CASE STUDY OF CRDB BANK". Being the first of its kind within the context of the study, it is largely an exploratory study on the marketorientation (MO) with twin purposes of: (a) examining the extent of MO in CRDB Bank as perceived by employees and (b) exploring the development and implementation of MO culture in the bank, with a special focus on the facilitating and hindering factors. Emanating from these purposes are four main objectives of the research that underpinned the research namely (1) to explore the existing marketing philosophies in the bank, (2) to measure the employees perceived level of MO and its constituent dimensions, including the development of the MO scale that is suitable to a banking institution, (3) to explore whether the perception of the level of MO differs according to the following attributes (a) hierarchical levels of the organisation (top, middle and lower management and between head office and branches offices of the bank), (b) employee-specific(personal) attributes(c) size of branches ( large, medium and small), (d) location of branches ( between those in competitive areas and in least competitive areas) and (e) profitability performance ( between above-median and below-median performing branches), and (4) to explore the facilitating and hindering factors for the development and implementation of MO in CRDB Bank and Tanzanian financial sector in general. The main methodology used in this research is a case study. This enabled us to use method triangulation, whereby both qualitative and quantitative methods were used. Principally, we applied the following research tools/techniques in data gathering: interviews, documentary analysis, survey questionnaire and personal observation. The field research involved two phases. The first was a pilot study that entailed conducting discovery-oriented interviews with 9 bankers in six banks. The second phase was conducting the substantive research in CRDB Bank. The findings of the study are fascinating and may have far- reaching implications both in terms of theory and practice. From a themantic dimension, we successfully reconceptualised MO, by developing a scale for measuring the extended MO. We clarified this as the Simultaneous Market orientation, SMO, which composes five key dimensions: external customer-orientation, competitor-orientation, interfunctional orientation, internal 111 customer-orientation and profit-orientation. The scale was tested and validated for its psychometric properties. In other words, the SMO scale was found, reliable and valid, implying that the five components fully represented the SMO. External customerorientation and profit-orientation were ranked as first and second most important dimension of SMO in the bank. Competitor-orientation, interfunctional co-ordination and internal customer-orientation were ranked third, fourth and fifth, respectively. From the practice perspective, our findings indicates that marketing-orientation and salesorientation are the dominant marketing philosophies, while production-orientation and societal marketing-orientation are the less marketing philosophies. This in practice indicates a co-existence of different marketing philosophies in the organisation contrary to the main stream literature exhortations. Also, the research revealed an integration perspective of employees on SMO culture. Essentially, this has established the importance of internal customer-orientation in influencing or leading to employees' participation, morale, training, job satisfaction and retention as a critical determinants of successful SMO implementation. Furthermore, we have identified the main facilitative and hindering factors for the development and implementation of MO in the specific context of CRDB bank and the Tanzanian financial sector in general. These antecedent factors have a profound effect on MO implementation. In general, our findings have set a solid base and raised issues that are likely to chart the future direction of MO situation not only in Tanzania, but also in other African countries that are undergoing market-driven transformation of their economies. The main public policy implication of this study is that there is a need to strengthen the macroenvironment and to mount public education in order to foster MO behaviour and practice. Similarly, at the management level, there is a need for effective and adequate management leadership and support for adoption and implementation of MO culture and the need for sustained or continuous changes given the emergent internal and external environment. The essential role of informed "change agents" in fostering MO could not be discounted.

Cracking and occasional leaks have been reported in some Boiling Water Reactor (BWR) control rod drive (CRD) stub tubes. Roll expansion of the housing against the Reactor Pressure Vessel (RPV) bottom head penetration has been used successfully to provide a leak barrier. The recently approved ASME Code Case N-730 “Roll expansion of Class 1 Control Rod Drive (CRD) Bottom Head Penetrations in BWRs, Section XI, Division 1” provides the specific criteria for the application of roll expansion. The minimum roll band length in the Code Case was based on the requirement that the roll joint capability exceed the scram forces on the CRD. The roll joint capability was based on a simplified analytical model with assumed friction factors. The predictive model was then compared with the results of extensive testing on mockups. This paper describes the results of the testing that has been performed to determine the load capability of roll repairs for different roll band lengths, material combinations (stainless steel and Alloy 600), percent wall thinning, thermal cycling and surface condition. The mock-ups were rolled using procedures and rolling equipment similar to those used in actual plant application. The mock-ups were tested in a testing machine by applying a ‘push force’ on the housing. In addition to measuring the force using a load cell, strain gages were also used to measure the strains on the housings. LVDTs were used to monitor the displacement during the test. The results showed that the resistance of the rolled joint (i.e. the load capability) is proportional to the roll length. The load capability was not a strong function of wall thinning or thermal cycling. It was strongly affected by the surface condition (e.g. oxidation) and the housing material yield strength. The predictive model was consistent with the test results and confirmed that the roll expansion joint has substantial load capability. Thus, the roll joint is not only a leak barrier, but also a structural load-carrying joint that is sufficient to resist the upward scram loads on CRDs.

The control rod drive mechanism (CRDM) is an important safety-related component in the nuclear power plant (NPP). When CRDM steps upward or downward, the pressure-containing housing of CRDM is shocked axially by an impact force from the engagement of the magnetic pole and the armature. To ensure the structural integrity of the primary coolant loop and the functionality of CRDM, dynamic response of CRDM under the impact force should be studied. In this manuscript, the commercial finite element software ANSYS is chosen to analyze the nonlinear impact problem. A nonlinear model is setup in ANSYS, including main CRDM parts such as the control rod, poles and armatures, as well as nonlinear gaps. The transient analysis method is adopted to calculate CRDM dynamic response when it steps upward. The impact loads and displacements at typical CRDM locations are successfully obtained, which are essential for design and stress analysis of CRDM.

The motion of CRDM (Control Rod Drive Mechanism) is affected by the electro-magnetic force and the fluid resistance, also the movement process affected the control system. In this paper, we considered all the coupled factor and try to decouple the problem to several simple problems. We supposed 3 decoupling methods, and try to verification each assumption, and finally we found that an assumption is valid — the electromagnetic force is mainly affected by the displacement of CRDM in the attract process of kinetic jaw armature and kinetic jaw magnetic pole. We studied the fluid resistance force by CFD simulation, and exported the force to the electromagnetic model which is built by Infolytica/Magnet, the control system model is built by Matlab/Simulink coupling interface. In order to simulate the continuous movement process of CRDM, we studied the boundary data at each time period, and linked each period by in-house code. Then we tested the coupling simulation, listed the duration time of electric current and the movement duration time of mechanism. Compare to the test data, the maximum error is 17%. On the whole, this solution coupled CFD, electro-magnetic and control system, the simulation result can help engineers to optimize the control algorithm and shape design in a certain extent.

The article highlights 5 axes of client-server applications that are relevant for the implementation of CRDT. On the selected axes, 6 main application architectures are selected. For each dedicated architecture, the optimal logical location of the CRDT in the application is analyzed and a graphical diagram of the CRDT deployment is constructed. To increase the practical applicability of the study, the latest technologies and trends in the deployment of client-server applications were taken into account, and recommendations were developed for the use of CRDT in different application architectures and for popular Enterprise application development platforms. In addition, the most suitable CRDT implementations for 5 broad classes of applications are identified.

We built three successive versions of a thermal decomposition cavity ring-down spectrometer and tested their response to explosives. These explosive compound analyzers successfully detected nitroglycerine, 2,4,6-trinitrotoluene (TNT), pentaerythryl tetranitrate, hexahydro-1,3,5-trinitro-s-triazine and triacetone triperoxide (TATP). We determined the pathlength and limits of detection for each, with the best limit of detection being 13 parts per trillion (ppt) of TNT. For most of the explosive tests, the peak height was higher than the expected value, meaning that peroxy radical chain propagation was occurring with each of the explosives and not just the peroxide TATP.
