Der volle Inhalt der QuelleThe historic of contamination by metals in the Valley of Ribeira de Iguape River is well known, and there are evidences that these chemicals are carried to the Estuarine Complex of Cananeia-Iguape (ECCI), adsorbed on the suspended particles. Metals can thus reach the south of the ECCI, where they may accumulate in the sediments and filter feeding organisms. This study aimed to evaluate the ecological risks associated to the metals originated from the former mining areas of the upstream Ribeira de Iguape River, by using different lines of evidence, which included geochemical and biological analysis (acute toxicity tests, bioaccumulation and biochemical biomarkers in bivalves\' gills). The sediments presented moderate to high levels of metals and acute toxicity, which was correlated with the concentrations of metals (Cu, Pb and Zn). These elements were found in high concentrations in the bivalves soft tissues. The biochemical biomarkers (MT, GSH, GST, GPx, GR, AChE, LPO, and DNA damage) indicated the occurrence of significant effects in the bivalves. Therefore, the results showed that metals are carried into the South portion of the estuary, accumulating in the biota and causing negative effects to the aquatic organisms, producing thus ecological risks to the ECCI.

Der volle Inhalt der QuelleThe neural crest is a multipotent, migratory cell population of vertebrate embryos that give rise to a variety of cell derivatives. An evolutionary-conserved epistatic network, which involves secreted factor BMP, FGF and WNT; has been proposed to control neural crest formation. This gene network suggests secreted factors induce the expression of neural border specifiers that will in turn induce neural crest specifiers genes. During my doctoral training I have investigated early steps in the induction of neural crest cells, focusing on the role of transcription factors AP2 and Hairy2. AP2 and Hairy2 are both expressed in the presumptive neural crest domain and are required for neural crest induction. My work demostrates that AP2 activity controls sequential steps in the neural crest network. First, AP2 acts as a neural border specifier to integrate FGF and WNT signals and allow neural border formation. Second, AP2 acts as a neural crest specifier in neural crest formation independent from its earlier role in neural border establishment. Additionally, I showed that Hairy2 activity is required in parallel with AP2 and downstream of FGF signaling to allow neural border formation. Hairy2 both blocks neural crest specification and maintains neural border genes expression. Hairy2 also induces Delta1 expression leading to activation of Notch pathway in the adjacent cells and suggest an additional role in specification of this tissue. Taking together my results allowed us to place AP2 and Hairy2 at specific steps in the regulatory network, which controls neural crest formation. This serves to highlight a new role of AP2 in neural crest development. Furthermore, I propose a model to explain how the embryo sets up a balance between neural crest cell specification and maintenance of a progenitor pool

Der volle Inhalt der QuelleIn this work, we investigated the effects of AIP deficiency in vivo on somatotroph cells, both at the secretory and proliferative levels and explored in vitro the signaling pathways potentially involved in AIP-dependent pituitary tumorigenesis. Phenotype analyzes of Aip+/- mice, especially of GH pulsatility, show that, unlike humans, mutant mice do not develop gigantism nor GH hypersecretion and present with a much lower penetrance of pituitary adenomas than initially described. In vitro studies in fibroblasts of AIP-mutation positive patients with pituitary adenomas and in somatolactotroph GH3 cells demonstrate that AIP mutations alter AhR (Aryl hydrocarbon Receptor) transcriptional activity and modify the cAMP pathway. Finally, GPR101 mutations, recently reported in patients with sporadic acromegaly, have also been identified in a small portion of patients with sporadic non-somatotroph pituitary adenomas, without any association with AIP mutations. This research work defines the consequences of AIP mutations on somatotroph cell function and AhR transcriptional activity. The role of cAMP signaling in AIP-related pituitary tumorigenesis will be further evaluated in a new transgenic mouse model

Admixtures can be used to provide the necessary fluidity, so the material can be efficiently pumped down as well as penetrate porous rocks with controlled filter loss. Construction admixtures can be used to modify the properties of oil well cements provided they can withstand and hold their properties at the higher than ambient temperatures usually encountered in oil fields. In civil construction, superplasticizer play the role of dispersants that reduce the facto r of water cement improve mechanical properties and fluidity of the cement, whereas anti-segregation agents improve the workability of the slurry. In the present study, oil well cement slurries were produced adding both a dispersant and an anti-segregation agent conventionally used in Portland CPII-Z-32 RS cement aiming at materials for primary cementing and squeeze operations. Three basic aspects were evaluated: fluidity, filter loss and the synergetic effect of the admixtures at two temperatures, i.e., 27?C and 56?C, following API RP 10B practical recommendations. The slurries were prepared using admixture concentrations varying from 2.60 Kgf/m3 (0.02 gallft3) to 5.82 Kgf/m3 (0.045 galJft3) BWOC. The density of the slurries was set to 1.89 g/cm3 (15.8 Ib/gal). 0.30 to 0.60% BWOC of a CMC-based anti-segregation agent was added to the cement to control the filter loss. The results showed that the addition of anti-segregation at concentrations above 0.55% by weight of cement resulted in the increased viscosity of the folders in temperatures evaluated. The increasing the temperature of the tests led to a reduction in the performance of anti-segregation. At concentrations of 5.20 kgf/m3 (0,040 gallft3) and 5.82 Kgf/m3 (0,045 gal/ft 3) observed a better performance of the properties evaluated in the proposed system. At low temperature was observed instability in the readings of rheology for all concentrations of anti-segregation. Contents that increasing the concentration of anti?-segregation is limited concentrations greater than 0.55 % BWOC of the CMC in temperature analyzed. The use of the system with CMC promoted a good performance against the properties evaluated. The principal function of anti?-segregation was optimized with increasing concentration of superplasticizer, at temperatures above the 2rC. The study of the behaviour of systemic additives, resulting in slurries of cement, which can be optimized face studies of other intrinsic properties in oil fields

Para o controle da ?gua perdida por filtra??o foi utilizado um agente anti-segregante a base de CMC com concentra??es variando de 0,30 % a 0,60 % em peso de cimento. Os resultados mostraram que a adi??o do anti-segregante em concentra??es acima de 0,55 % em peso de cimento resultou no aumento da viscosidade das pastas nas temperaturas avaliadas. Foi verificado que o aumento da temperatura dos testes provocou uma redu??o no desempenho do anti-segregante. Nas concentra??es de 5,20 kgf/m3 (0,040 gal/p?3) e 5,82 Kgf/m3 (0,045 gal/p?3) observou-se um melhor desempenho das propriedades avaliadas no sistema proposto. Na temperatura ambiente foi verificado instabilidade nas leituras de reologia para todas as concentra??es do anti-segregante. Foi verificado que o aumento da concentra??o de anti-segregante ? limitada as concentra??es superior a 0,55 % em peso de cimento nas temperaturas analisadas. O uso do sistema contendo superplastificante, aliado ao anti-segregante promoveu um bom desempenho face ?s propriedades reol?gicas e de controle de filtrado das pastas. A fun??o principal do anti-segregante foi otimizada com o aumento da concentra??o do superplastificante, a temperaturas acima da ambiente. Por?m o aumento da concentra??o do agente dispersante nas pastas de cimento pode provocar o retardamento das pastas em temperaturas pr?ximas da ambiente. Der volle Inhalt der QuelleHIV-1 manipulates antigen-presenting cells (APC) such as dendritic cells (DC), witch orchestrate innate and adaptive immune responses, in order to propagate in the host and to establish viral reservoirs. We are studying the role of autophagic processes in DC/HIV-1 interactions with a focus on antigen presentation. We have previously shown that macroautophagy in DC participates in the degradation of incoming HIV-1 particles leading to activation of HIV-1-specific (HS) CD4 T cells. HIV-1 can also productively infect DC. I thus first asked whether neo-synthetized viral proteins might represent an additional source of HIV-1 antigens. Remarkably, I have shown using infected monocyte derived DC that de novo expression of Gag leads to the activation of HS CD4 T cells, highlighting that this antigen is endogenously processed in order to be presented into MHC-II molecules. Since macroautophagy and chaperon-mediated autophagy (CMA) are known to be involved in this process for other viral antigens and model antigens, I then dissected the role of these two pathways. Using several tools including inhibitors and shRNA, I demonstrated that in HIV-1-infected DC neither macroautophagy nor CMA contribute significantly to the processing of HIV-1-Gag epitopes into MHC-II molecules. I also used a Gag-LC3 fusion protein to specifically channel Gag into LC3+ autophagic vesicles in DC. In this context, inhibiting autophagy dramatically reduced the presentation of HIV-1-Gag epitopes to CD4+ T cells. Strikingly, channelling Gag into autophagosomes generated epitopes that were not processed endogenously in the context of HIV-1 infection. Thus specifically directing HIV-1 proteins toward autophagosomes might influence the repertoire of MHC II-restricted HIV-1 antigens. I further analyzed whether autophagy could affect HIV-1 replication in infected DC. In these cells, in contrast to what has been described in macrophages, Gag did not colocalize with LC3 or with the autophagic adaptor p62, suggesting that newly-produced HIV-1 particles are not sequestrated into autophagosomes. The Gag-LC3 fusion protein was used here as a positive control of colocalization. To determine whether my findings might reveal a DC-specific escape mechanism developed by HIV-1, I used various HIV-1 mutants, enhanced autophagic flux using drugs or TLR ligands, and expressed Gag in the absence of other HIV-1 proteins. Overall, my work suggests that HIV-1 does not manipulate autophagy in productively-infected DC. Moreover, modulating autophagy in DC (using shRNA) does not impact HIV-1 replication and propagation. Finally, my work highlights the complexity of the interactions between the autophagic process and HIV-1 replication in DC. Unlike during viral entry, HIV-1 does not seem to be targeted into autophagosomes after viral replication in infected DC, and autophagy does not contribute significantly to the processing of endogenous viral antigens. Nonetheless HIV-1-infected DC efficiently activates HS CD4 T cells, and targeting HIV antigens into autophagosomes greatly enhances this activation and might broaden the repertoire of MHC-II-restricted antigen. Further dissection of the various routes of endogenous HIV antigen processing would aid in the development of innovative vaccines

Eurycoma longifolia (Tongkat Ali) is a plant with folk medicine in Indonesia, Malaysia, Thailand and Vietnam. The residences in Malaysia usually treat it as “ginseng of Malaysia”, and think it possesses multiple medical functions such as anti-hypertension and appetite stimulation. The purpose of this project was to examine the effect of E. longifolia extract on plasma NO and cGMP concentration in mice. Viagra was also used for comparison. Mice were treated by E. longifolia, Viagra, L-Arginine, Tongkat Ali plus ginseng, Dioscorea for 2 and 4 weeks. Plasma NO, cGMP level and the activity of type 5 phophodiesterase and cytochrome P450 were analyzed. Results showed that 5 treatments increased the NO and cGMP levels, and inhibited the activity of type 5 phophodiesterase. Tongkat Ali and Tongkat Ali plus ginseng treated mice at 4 weeks has significantly higher cytochrome P450 mRNA expression. These treatments did not affect the anti-oxidative defense in liver and kidney. These findings implied that Tongkat Ali could provide protective or medical functions via elevating plasma NO and cGMP concentration and decreasing PDE5 enzyme activity.
