Abstract A group of homochiral imidazolium salts bearing hydrophobic camphor derived moiety and ester or amide functional group were synthesized and characterized. The novel imidazolium bromides were tested as antimicrobial and antifungal agents and their minimal inhibitory concentration (MIC) was evaluated and compared to clinically used benzalkonium bromide (BAB) and carbethopendecinium bromide. The MIC values of amide derivatives 2a and 2b were slightly smaller than those for BAB, indicating their good activity. None of the prepared salts was more effective than carbethopendecinium bromide. The biocidal efficacy of amide derivatives was much higher compared to the ester analogues.

Inclusion complexation of an antimicrobial quaternary ammonium salt, carbethopendecinium bromide (SBr, Septonex), with five cyclodextrins was investigated potentiometrically using the prepared membrane electrodes selective to the surface-active carbethopendecinium cations (S+). Relatively strong complexation of the S+ cations with native α-cyclodextrin (α-CD), β-cyclodextrin (β-CD), and their random-substituted derivatives, namely hydroxypropyl-α-cyclodextrin (HP-α-CD), methyl-β-cyclodextrin (M-β-CD), and hydroxypropyl-β-cyclodextrin (HP-β-CD), was evidenced in dilute aqueous solution. In the region where the potentiometrically determined concentration of the free S+ cations remained lower than the critical micelle concentration (cmc) of SBr by a factor of ca ten, formation of the 1:1 complexes with the complexation constants K11 ≈ 104-105 was evaluated by two respective methods, based on a modified linear and a non-linear regression. Deviations from the 1:1 complexation were observed when concentration of the free S+ cations approached cmc of SBr more closely and also in solutions with large excess of α-CD. Comparison of K11 values corroborated the inclusion of the n-C14H29 alkyl chain of the carbethopendecinium cation as the mechanism of its complexation with cyclodextrins. The well-soluble native α-CD with good complex-forming capability towards S+ cations may be especially suitable for possible blocking the undesirable residues of carbethopendecinium bromide.

The rheological properties of hydrogels prepared by physical interactions between oppositely charged polyelectrolyte and surfactant in micellar form were studied. Specifically, hyaluronan was employed as a negatively charged polyelectrolyte and Septonex (carbethopendecinium bromide) as a cationic surfactant. Amino-modified dextran was used as a positively charged polyelectrolyte interacting with sodium dodecylsulphate as an anionic surfactant. The effects of the preparation method, surfactant concentration, ionic strength (the concentration of NaCl background electrolyte), pH (buffers), multivalent cations, and elevated temperature on the properties were investigated. The formation of gels required an optimum ionic strength (set by the NaCl solution), ranging from 0.15–0.3 M regardless of the type of hydrogel system and surfactant concentration. The other compositional effects and the effect of temperature were dependent on the polyelectrolyte type or its molecular weight. General differences between the behaviour of hyaluronan-based and cationized dextran-based materials were attributed to differences in the chain conformations of the two biopolymers and in the accessibility of their charged groups.

The critical micellar concentrations (cmc) of the title cationic surfactants were measured in aqueous solutions at 25 °C in the presence of KBr, KF, KCl, KI, KSCN, KNO3, KAc and HCOOK. The conductometric and, in part, potentiometric methods employing ion selective electrodes were used. For 1-cetylpyridinium bromide in the absence of any of the electrolytes, the decreasing temperature dependence of cmc was measured conductometrically over the region of 12 - 40 °C and the association degree of the micelles with Br- anions was calculated. The decrease in cmc of the two surfactants due to the presence of electrolytes (in concentrations up to tenfold cmc) was expressed by means of a semiempirical equation. There are indications that I- and SCN- anions enter into specific interactions with micelles.

The diploma thesis deals with the preparation and characterization of catanionic complexes and their possible application to pharmaceutics and medicine. The catanionic complexes were prepared by mixing two oppositely charged surfactants with the concentration of 20 mmol·dm­3 in various volume ratios. Two systems were analyzed – CTAB + SDS and Septonex + SDS. Both systems were prepared in an aqueous and physiological environment. The turbidimetric analyses provided results of the intensity of turbidity of the prepared mixtures. The size and stability of the prepared particles were determined by DLS and ELS measurements. CTAB-rich samples showed significant viscosity changes and they were therefore characterized by rheological measurements. The results of this work show that the asymmetry of alkyl chains, surfactant selection, temperature, and ionic strength have a significant influence on the properties and self-assembly of surfactants in catanionic complexes.

This doctoral thesis studies the properties and behaviour of phase-separated hydrogels. These can be prepared by interaction of polyelectrolytes with oppositely charged surfactants. Negatively charged polysaccharide hyaluronan and cationic surfactant carbethopendecinium bromide (Septonex), whose properties, behaviour and utilization are described in the first two chapters of theoretical part, were selected for this role. Hyaluronan is naturally-occuring in living organisms and is known for his specific targeting to the tumour cells. Septonex is used as antiseptic and disinfectant. Experimental part of this work is focused on examination of the structure and behaviour of these types of hydrogels especially from rheological point of view. The crucial part of this study was to establish mechanical properties and their dependence on environment. This led to design further studies. Structural analysis was held using the methods of ATR-FTIR, ionic chromatography and ICP-OES helping to measure the inner content of entry materials in the gels and supernatants respectively. The last chapter deals with antimicrobial activity. The results of this study indicate a potential usage of these substances in the field of medical applications.

The thesis deals with the study of hydrophobic domains in phase separated hydrogels. Two types of polyelectrolytes (hyaluronan, dextran) and oppositely charged surfactants (carbethopendecinium bromide, sodium dodecylsulphate ) are used. Phase separated hydrogels are formed by electrostatic interactions. The target of the thesis is to study the polarity of environment in phase separated hydrogeles by fluorescence spectroscopy. Fluorescence measurements is performed by using pyrene, Nile red and laurdane as fluorescence probes. All probes are sensitive to the polarity of the environment. The results showed that probes can be expected in the polar environment of hydrogels.
