Bromodomain testis (BRDT), a member of the bromodomain and extraterminal (BET) subfamily that includes the cancer targets BRD2, BRD3, and BRD4, is a validated contraceptive target. All BET subfamily members have two tandem bromodomains (BD1 and BD2). Knockout mice lacking BRDT-BD1 or both bromodomains are infertile. Treatment of mice with JQ1, a BET BD1/BD2 nonselective inhibitor with the highest affinity for BRD4, disrupts spermatogenesis and reduces sperm number and motility. To assess the contribution of each BRDT bromodomain, we screened our collection of DNA-encoded chemical libraries for BRDT-BD1 and BRDT-BD2 binders. High-enrichment hits were identified and resynthesized off-DNA and examined for their ability to compete with JQ1 in BRDT and BRD4 bromodomain AlphaScreen assays. These studies identified CDD-1102 as a selective BRDT-BD2 inhibitor with low nanomolar potency and >1,000-fold selectivity over BRDT-BD1. Structure–activity relationship studies of CDD-1102 produced a series of additional BRDT-BD2/BRD4-BD2 selective inhibitors, including CDD-1302, a truncated analog of CDD-1102 with similar activity, and CDD-1349, an analog with sixfold selectivity for BRDT-BD2 versus BRD4-BD2. BROMOscan bromodomain profiling confirmed the great affinity and selectivity of CDD-1102 and CDD-1302 on all BET BD2 versus BD1 with the highest affinity for BRDT-BD2. Cocrystals of BRDT-BD2 with CDD-1102 and CDD-1302 were determined at 2.27 and 1.90 Å resolution, respectively, and revealed BRDT-BD2 specific contacts that explain the high affinity and selectivity of these compounds. These BD2-specific compounds and their binding to BRDT-BD2 are unique compared with recent reports and enable further evaluation of their nonhormonal contraceptive potential in vitro and in vivo.

Sudden sensorineural hearing loss is a hearing loss of >30 dB in at least three consecutive frequencies that occurs in 3 days. The aim of this study was to investigate anxiety and depression caused by sudden, idiopathic, one-sided hearing loss. The levels of anxiety and depression in patients with this type of hearing loss were determined using the Beck Anxiety Scale (BAS) and the Beck Depression Inventory (BDI) at the time of the patient's first visit. In total, 56 patients (32 men and 24 women) with a mean age of 32.8 ± 9.9 years (range: 20 to 58 years) were selected as the patient group and 45 individuals without symptoms of anxiety and depression were selected as the control group. The mean pretreatment air-conduction threshold and bone-conduction threshold were 61.1 ± 26.1 and 49.4 ± 13.8, respectively. In the patient group, the pretreatment mean anxiety, depression, and hopelessness scores were 19.5 ± 10.7, 11.6 ± 8.4, and 6.2 ± 4.7, respectively. The control group's mean anxiety, depression, and hopelessness scores were 4.1 ± 3.0, 3.8 ± 2.1, and 1.8 ± 1.0, respectively. For all the tests, the difference between the patient group and the control group was statistically significant (p < 0.001 for all). Hearing levels were not correlated with scores on the BAS, BDI, and Beck Hopelessness Scale (p = 0.1, p = 0.6, and p = 0.4, respectively). In conclusion, the results of this study show that sudden hearing loss can cause anxiety and depression. Questioning patients with sudden hearing loss about symptoms associated with anxiety and depression might be useful, and a psychiatric consultation should be requested if necessary.

ABSTRACT Objective: Aim of the present study was to evaluate the retreatability and reestablishment of apical patency of two calcium silicate-based sealers, TotalFill BC Sealer (BCS) and mineral trioxide aggregate Fillapex (MTA F), versus AH Plus, when used in combination with Gutta-percha (GP). Materials and Methods: The canals of 54 single-rooted anterior teeth were instrumented and filled with GP/AH Plus (Group A), GP/MTA F (Group B), or GP/BCS (Group C) using continuous wave obturation technique. The groups were subdivided into subgroups with the master-GP cone placed to the working length (WL) or intentionally 2 mm short. The retreatment procedures were performed using ultrasonics, chloroform, rotary, and hand files. The ability to establish the patency and reach WL was determined as well as the time taken to reach WL was calculated in minutes. Furthermore, the samples were observed under a dental, optical microscope, after vertically splitting them. Results: The WL and patency were reestablished in 100% of specimens in all groups. The Mann–Whitney U-test indicated that there was a significant difference in the amount of time required to reach WL between the groups (P < 0.05) with group GP/BCS short of the WL showing the most amount of time to be retreated. Conclusion: The novel calcium silicate-based sealers are negotiable under simple root canal anatomy. However, the conventional retreatment techniques are not able to fully remove them.

AbstractA sensitive, specific and cost-effective HPLC method was developed for quantitative determination of carbinoxamine in human plasma using liquid chromatography with ultraviolet detection. A simple liquid–liquid extraction by ethyl acetate was used to extract carbinoxamine from human plasma. Satisfactory separation was achieved by a mobile phase consisting of 20 mM ammonium dihydrogen orthophosphate containing 0.01% triethyl amine (pH adjusted to 3 by using phosphoric acid) and acetonitrile in ratio of (75:25, v/v) at a flow rate of 0.9 mL/min on Hypersil® BDS C18 column (250×4.6 mm, 5 μm) column. The UV detector was set at 260 nm. The developed method was validated in human plasma with a lower limit of quantitation of 5 ng/mL for carbinoxamine. Linearity was demonstrated over the concentration range 5–200 ng/mL. The observed within- and between-day assay precision ranged from 1.902 to 14.90%; accuracy varied between 98.87 and 108.0%. This method can be used suitably for pharmacokinetic studies and in therapeutic drug monitoring in patients treated with carbinoxamine.

Penelitian ini bertujuan untuk mengembangkan perangkat pembelajaran berbasis inkuiri terbimbing dengan menggunakan program simulasi PhET yang layak untuk mereduksi miskonsepsi siswa. Pengembangan perangkat pembelajaran menggunakan model 4-D yang direduksi menjadi 3-D dan diujicobakan di tiga kelas XI SMAN 4 Surabaya tahun pelajaran 2015/2016 dengan one group pretest posttest design. Pengumpulan data menggunakan observasi, tes, dan angket.Data dianalisis menggunakan deskriptif kualitatif dan kuantitatif. Temuan penelitian yaitu: (1) Validitas: (a) Validasi RPP kategori baik, BAS kategori cukup baik, LKS kategori baik, Tes Uji Miskonsepsi umumnya valid; (b) Rata-rata keterbacaan BAS sebesar 59,8% dan LKS sebesar 71,3%; (c) Rata-rata tingkat kesulitan BAS sebesar 32,5% dan LKS sebesar 34,3%; (2) Kepraktisan meliputi: (a) Perangkat pembelajaran ditinjau dari keterlaksanaan RPP berkategori baik, dan (b) Tingkat aktivitas siswa dikategorikan baik (3) Keefektifan yang meliputi: (a) hasil tes uji miskonsepsi dengan N(g) 0,73; dan hasil analisis uji statistik one-way ANOVA, yaitu : 1) tidak ada perbedaan yang signifikan penurunan miskonsepsi dari ketiga kelas, 2) N-Gain penurunan miskonsesi siswa dialami pada masing-masing kelas (b) Respon siswa terhadap KBM adalah positif. Berdasarkan hasil temuan dalam penelitian ini, disimulkan bahwa perangkat pembelajaran listrik dinamis dengan model inkuiri terbimbing layak digunakan oleh siswa SMA untuk menurunkan miskonsepsi siswa.

A great deal of interest currently surrounds the development of electrophosphorescent materials, as they may exhibit efficiencies greater than the theoretical limit of 25% for purely fluorescent materials. The most studied material for such purposes is jac-Ir(ppY)3, which exhibits efficient green phosphorescence in solution and in electroluminescent devices. Europium(III) complexes have also received attention for their narrow bandwidth red emission. A number of charge-neutral iridium(III) complexes have been prepared as potential electrophosphorescent materials. Bis-terdentate coordination has been investigated, in a quest to provide improved stability with respect to jac-Ir(ppY)3 derivatives, an important objective in commercial development. The complexes described herein are the first examples of iridium bound to NACAN coordinating 1,3- di(2-pyridyl)benzene derivatives. Methyl substitution at C4 and C6 of the phenyl ring (i.e. 1,5-di(pyridin-2-yl)-2,4-dimethylbenzene (dpydmbH)) is required to inhibit competitive cyc1ometalation at these positions. Efficient red emission was observed from Ir(dpydmb-N,C,N)(dppy-C,N,C) (dppyH2 = 2,6-diphenylpyridine) (Am ax = 585 nm, PL = 0.21 in degassed CH3CN). Fluorination of the dppy ligand at the 2 and 4 positions of the phenyl rings results in a simultaneous blue-shift and increase in efficiency (Amax = 547 nm, PL = 0.41 in degassed CH3CN). These complexes exhibit an unusual meridional geometry, difficult to achieve for tris-bidentate complexes. The complex Ir( dpydmb-N,C,N)(ppy-C,N)Cl is exceptionally emissive (Amax = 508 nm, PL = 0.76 in degassed CH3CN). Ir( dpydmb-N,C,N)(tppic-C,N,0) and Ir( dpydmb-N,C,N)(hbqcC, N,O) (tppicH2 = 4-p-tolyl-6-phenylpicolinic acid; hbqcH2 = 4-hydroxybenzo[h]quinoline- 2-carboxylic acid) were poorly emissive, attributed to a photo activated labilisation of the Ir-C bond in solution. The effect of substitution position of a senslt1s1ng aminobenzophenone chromophore was investigated in a series of europium(III) macrocyc1ic complexes. Maximum photoluminescence quantum yield was observed for the para-substituted isomer, attributed to the distance dependence of the efficiency of energy transfer. An unexpected rearrangement was observed for the ortho-substituted ligand, resulting in a quinoline derivative with a rare N-arylated substitution pattern.

Fixed bed reactors are widely applicable in a range of chemical process industries. Their ease of use and simplified operation make them an attractive and preferred option in reactor selection, however the geometric complexities within the bed as a result of the unstructured packing has made the design of such beds historically based on pseudo-homogenous models together with correlation-based transport parameters. Low tube-to-particle diameter ratio (N) beds, in particular, are selected for highly exothermic or endothermic reactions, such as in methane steam reforming or alkane dehydrogenation. Due to the large fraction of tube to catalyst particle contact in these low N beds, wall effects induce a mass transfer boundary layer at the wall, and in the case of thermal beds, a simultaneous resistance to heat transfer. Computational Fluid Dynamics (CFD) has been shown to be an accurate tool for experimental validation and predictive analysis of packed beds, and may be used to derive more accurate design parameters for fixed bed reactors. More specifically, the elucidation of dispersion, or the transport of reactant and product within the bed due to molecular diffusion and convective flow is of fundamental interest to the design of fixed beds. Computational Fluid Dynamics was used in this research to study solute dispersion in eight beds of varying N at a range of particle Reynolds numbers in the laminar flow regime. In the first stage of research, flow development was simulated in three-dimensional packed beds of spheres. Then, the reactor wall was sectioned to include a boundary condition of pure methane, from which the solute could laterally disperse into the bed. In the second stage, a two-dimensional representation of the bed was created using the commercial Finite Element Analysis software COMSOL Multiphysics. In these models, axial velocity profiles and radial methane concentration profiles taken from the 3-D models were supplied, and a fitting procedure by use of the Levenberg-Marquardt Least-Squares optimization algorithm was completed to fit radial dispersion coefficients and near-wall mass transfer coefficients to the CFD data. These optimization runs were conducted for all N at a number of bed depths in each case. Two sub-studies were conducted in which a constant velocity profile and a local velocity profile were supplied to the 2-D model, and the optimization re-run. It was found that this two parameter model did not fully account for various mechanisms of dispersion in the bed, namely the increasing rate of dispersion from the tube wall boundary layer up to the bed center, but only accounted for a diffusive-dispersion at the wall and a constant-rate, convective-dispersion everywhere else in the bed. Length dependency of dispersion coefficients were also noted, particularly in the developing sections of the bed. Nevertheless, the combined CFD and optimization procedure proved to be an accurate and time-efficient procedure for the derivation of dispersion coefficients, which may then lend themselves to the standard design of packed bed reactors.

The current study examined the relationship between depression and uncertainty within the stress-diathesis model. Depression is a mental health disorder that is wide spread within our society. However, few causes of this disease have been able to be identified. Studies in uncertainty have shown that it is a major stressor in day-to-day life. Previous research has shown that individuals with high levels of uncertainty show higher levels of depression. The stress-diathesis model, a model originally developed to explain differences in development of schizophrenia, provides a theoretically meaningful way to combine these two concepts. The model states that a person who is likely to develop a disease has an internal mechanism, a diathesis, that will be triggered upon presentation of a stressor. This study tested the idea that uncertainty can be used as a stressor to activate diathesis within an individual, aiding in the prediction of depression. In total, 163 participants were randomly assigned one of three conditions, an uncertainty threat, an affirmation condition, and a control condition. Participants were given pre and post independent variable measures of depression and anxiety. Findings suggest that uncertainty activated negative emotions differentially within the participants, resulting in higher levels of negative affects after the uncertainty threat, especially for participants who already scored relatively higher on depression indicators. These results hint at a possible understanding of why depression rates and diagnosis rates of mental health issues rise during economic downturn and other times of strong uncertainty.

Den aktuellen Forschungsstand darzustellen und Grenzen und Möglichkeiten der Interpretation archäologischer Phänomene aufzuzeigen ist ein zentrales Anliegen des interdisziplinär arbeitenden Teams. Ab dem 13. Jh. v. Chr. begann ein klimatisch begünstigter Zeitabschnitt, der einen Anstieg der Bevölkerungszahl mit sich bringt. In Mitteleuropa bildete sich eine neue Kulturgruppe, in der die Leichenverbrennung vorherrschte und deshalb Urnenfelderkultur genannt wird. Die bäuerliche Wirtschaftsweise war weiterhin das prägende gesellschaftliche Element. Die Haushalte waren charakterisiert durch eine ausgeprägte Vielfalt an Keramikformen und -verzierungen. Es war ein Zeitabschnitt, in dem das Bronzehandwerk seinen Höhepunkt erfuhr. Weitreichende Handelsbeziehungen und befestigte Zentralsiedlungen sind Beispiele für den Wohlstand am Ende der Bronzezeit. Nicht zuletzt wurden in dieser Zeit die ersten Eisengegenstände geschaffen, die die Wende zur Eisenzeit einleiteten.

Juden im Sport vor und während des Nationalsozialismus: 130 Lokalstudien in Nordrhein-Westfalen zeigen die Vielfalt des jüdischen Sportlebens. In der Geschichtswissenschaft bildet das Thema Sport im Kontext der jüdischen Geschichte nach wie vor weitgehend eine Leerstelle. Ungleich stärker hat sich die Forschung auf die geistesgeschichtlichen Verdienste konzentriert und dabei den Beitrag der Jüdinnen und Juden bei der Verbreitung einer Massenkultur - dem Sport - vernachlässigt. Dabei gab es mit dem Verband jüdisch neutraler Turn- und Sportvereine Westdeutschlands (Vintus) seit 1925 einen jüdischen Sportverband, der, einzigartig in Deutschland, eigene Meisterschaften im Fußball, in der Leichtathletik und im Turnen organisierte, da der bürgerliche Westdeutsche Spielverband jüdischen Sportvereinen die Teilnahme an seinen Rundenspielen und Meisterschaften verweigert hatte. Zahlreiche jüdische Sportlerinnen und Sportler blieben jedoch auch in den paritätischen Turn- und Sportvereinen aktiv, bis sie nach 1933 ausgeschlossen wurden. Die Autoren untersuchen das Engagement von jüdischen Sportlerinnen und Sportlern sowohl in den großen Städten Köln, Düsseldorf oder Dortmund als auch in kleineren Gemeinden. Es zeigt sich, dass in der Zeit der Diskriminierung und Verfolgung der Sport im jüdischen Alltagsleben eine herausragende Rolle spielte.

Introdução: O cenário brasileiro se destaca pela liderança no setor do agronegócio, o qual vem impactando na crescente utilização de insumos importados, como agroquímicos sintéticos. Visto que, o fungo Fusarium solani é capaz de infectar diversas culturas de grande interesse econômico como por exemplo: soja, milho, feijão e trigo, em diferentes estágios de desenvolvimento, sendo que a complexidade do solo e a variabilidade genética existentes no F. solani tornam as doenças causadas por ele de difícil controle, surgiu-se a necessidade de cultivos de maior qualidade e baixa nocividade, tanto a saúde humana, como ao meio ambiente, gerando assim uma procura por novos agentes de controle biológico, como fungos do gênero Trichoderma, que veio a contribuir diretamente na construção desse cenário mais sustentável e produtivo. Objetivos: Avaliar a eficiência de isolados do gênero Trichoderma, contra o fitopatógeno F. solani, utilizando-se do teste de avaliação de efeito dos metabólitos voláteis. Material e Métodos: Foi realizado o teste de avaliação de efeito de metabólitos voláteis, que consistiu em posicionar fundos de placas de petri umas sobre as outras, após ter vertido em cada uma delas, em meio de cultura Batata Dextrose Ágar (BDA). Foram posicionados os discos de 3 mm dos isolados de Trichoderma, na parte inferior das placas e na parte superior discos de 3 mm do patógeno F. solani, mantidos a 25ºC, por 7 dias, utilizando-se do controle como referência para o crescimento. O teste foi submetido à análise de variância e de médias, comparadas pelo teste de Scott-Knott, a 5% de probabilidade, empregando-se ao software estatístico Sisvar®. Resultados: Após o período de 7 dias do início do teste, foi possível notar que os isolados de Trichoderma, foram capazes de produzir metabólitos voláteis suficientes, inibindo assim o crescimento do fitopatógeno. Conclusão: Com base nos resultados obtidos, foi possível concluir que, os isolados do fungo Trichoderma, possuem potencial para atuar na inibição do crescimento do fitopatógeno F. solani, visto que, além de ser sustentável e de grande interesse ao mercado de biocontrole, contribui também com o crescimento e maior produtividade no cultivo ao qual foi aplicado.

The relative performance of (i) a body-fitted unstructured grid Navier-Stokes solver [Moin and Apte, AIAA J. 2006], and (ii) a fictitious domain based finite-volume approach [Apte et al. JCP 2009] is examined for simulating flow through packed beds of spheres at moderate flow rates, 50 ≲ Re ≲ 1300. The latter employs non-body conforming Cartesian grids and enforces the no-slip conditions on the pore boundaries implicitly through a rigidity constraint force. At these flow rates, fluid inertia can result in complex steady and unsteady pore scale flow features that influence macro-scale properties. We examine the requirements on both methods to properly capture these features in both simple and complex arrangements of spheres. First, two prototypical test cases of flow through packed beds are studied thoroughly at a range of Reynolds numbers in the inertial flow regime. Next flow through a random packing of 51 spheres at Re = 1322 is simulated using both methods. The suitability of both approaches to the complex configurations observed in large randomly packed beds is discussed.

Abstract In the present study, the performance and emissions characteristics of three low-temperature plasma (LTP) ignition systems were compared to a more conventional strategy that utilized a high-energy coil (93 mJ) inductive spark igniter. All experiments were performed in a single-cylinder, optically accessible, research engine. In total, three different ignition systems were evaluated: (1) an Advanced Corona Ignition System (ACIS) that used radiofrequency (RF) discharges (0.5–2.0 ms) to create corona streamer emission into the bulk gas via four-prong electrodes, (2) a Barrier Discharge Igniter (BDI) that used the same RF discharge waveform to produce surface LTP along an electrode encapsulated completely by the insulator, and (3) a Nanosecond Repetitive Pulse Discharge (NRPD) ignition system that used a non-resistor spark plug and positive DC pulses (∼10 nanoseconds width) for a fixed frequency of 100 kHz, with the operating voltage-controlled to avoid LTP transition to breakdown. For the LTP ignition systems, pulse energy and duration (or number) were varied to optimize efficiency. A single 1300 revolutions per minute (rpm), 3.5 bar indicated mean effective pressure (IMEP) homogeneous operating point was evaluated. Equivalence ratio (ϕ) sweeps were performed that started at stoichiometric conditions and progressed toward the lean limit. Both the ACIS and NRPD ignition systems extended the lean limit (where the variation of IMEP &lt; 3%) limit (ϕ = 0.65) compared to the inductive spark (ϕ = 0.73). The improvement was attributed to two related factors. For the ACIS, less spark retard was required as compared to spark ignition due to larger initial kernel volumes produced by four distinct plasma streamers that emanate into the bulk gas. For the NRPD ignition system, additional pulses were thought to add expansion energy to the initial kernel. As a result, initial flame propagation was accelerated, which accordingly shortens early burn rates.

The rise-times, from just above the base to the top of the bed, of single particles of dried sewage sludge and of two types of petroleum coke were measured in a slugging bed of sand fluidized by air. The sewage sludge and the petroleum cokes contained, on an as received basis, 49.5, 9.0 and 0.5 wt.% volatile matter, respectively. The bed (i.d. 25.5 mm; height 1300 mm) was electrically heated to a fixed temperature between 700 and 900°C. Two sizes of sand were fluidized: +212, −300 μm and +710, −850 μm. The expansion of the bed was held constant by keeping the excess gas velocity at (U − Umf) = 190 mm/s. In a bed of the coarser sand, it was found that the time of rise of single particles of sewage sludge (< 4 mm diam.) was large enough to allow devolatilization and char burn-out to be complete before the particle reached the upper surface of the bed. The measured times of rise are significantly longer than those found in bubbling beds [12, 13]. Thus, a slugging bed, of say 2 m in depth, would have significantly better heat retention and contact of the volatile material with the particulate phase than a bubbling bed of the same depth. With the finer sand, particles of sewage sludge and of petroleum coke containing volatile material rose rapidly to the top of the bed, well before combustion was complete. Published theory [6] was used to predict the rise-times of inert particles in a hot slugging bed. The predictions compare favourably with measurements using petroleum coke containing negligible amounts of volatile material and moisture. The use of a modified form of Stokes’s law to predict the velocity of rise of inert particles is shown to compare well with experiments. The experimental results suggest that once a minimum rate of gas evolution (from volatiles or moisture) is achieved a bubble forms which is large enough to lift a fuel particle to the surface. For particles meeting this criterion, the rise-time appears to be independent of the size of the fuel particle.

This paper presents a feasibility study for a floating medical support system, and describes its basic concept with regards to the installation location, its primary function and patient capacity. In 2011, Japan experienced a major catastrophe: the Great East Japan Earthquake and subsequent tsunami. After the tsunami struck, overland transport routes were cut off, and consequently emergency supplies and people requiring medical attention could not be transported very efficiently. Many hospitals and other emergency facilities were destroyed or were operating with reduced capabilities during this period, meaning victims could not receive treatment within a reasonable timeframe. Therefore, we need to consider emergency facilities, particularly for medical support, which are resilient and versatile for use in such situations. Floating bases for disaster relief and support ships are already available for deployment along coastal regions or on rivers near affected areas. A medical support floating base has also been proposed, which is called the Medi-float hereafter, in preparation for the anticipated Tokyo Inland Earthquake. The original proposal was deemed inadequate in terms of its choice of deployment area, and its maximum capacity for patients. The objectives of this paper are to decide the Medi-float’s primary function, the location where it should be installed, and to determine its capacity and a floor plan which adheres to the relevant regulations. It is determined that the primary function of the Medi-float should be as a dialysis treatment center during normal operation and primarily for the treatment of crush syndrome patients during a disaster. A location in Sumida ward is chosen for the installation area. In the floor plan, the Medi-float has 55 beds in dialysis treatment rooms. Also, there are areas of open space in the Medi-float to be used for coordinating the distribution of emergency supplies. The maximum capacity for patients is approximately 5,300 people, and the Medi-float can treat up to 130 patients with crush syndrome every 2 days. There is estimated to be around 106 patients with crush syndrome around the installation area in the event of a major earthquake. Therefore, this study clearly demonstrates the usefulness of the Medi-float.
