Peer-to-peer interactions, either through discussion forums or the peer review process, provide students with essential articulation skills as they reflect and respond to the ideas of others. Unfortunately, many students lack confidence in the value of their thoughts and feedback, or students experience difficulty maintaining interest in a peer's thoughts/ideas, which results in a lack of motivation to participate or in comments that are overly superficial, flattering, or brief. The interactive application NORA emphasizes that no one reviews alone by allowing the users to write comments and then combine, like, and rearrange them on a large canvas as they analyze a piece of work, examine a topic, or provide feedback. Thus, NORA facilitates the peer review and discussion process, addressing the challenges faced by students. NORA's visual-oriented interface novel in how it presents content to the students in smaller pieces, allows several threads of comments to be seen at the same time, and provides for easy interaction between users as they write or combine comments, guiding them through specific learning goals chosen by the instructor. Targeted at college students, the application was tested in two different classes, Rhetoric and Communication and Spanish /, with different classroom activities that are typically done orally with extensive class discussion. In both classes, students analyzed the subject matter and reviewed the medium as they responded to the comments of each other and the guidelines provided by the professor. The peer-to-peer interaction allowed users to build upon each other's comments, and promoted accurate, thorough, and relevant feedback in an engaging manner. NORA was seen to encourage more interaction, draw out quieter and shyer students, and boost the number of thoughtful, analytical responses.

This research project investigates the difficulties students encounter when solving physics problems involving the integral and the area under the curve concepts and the strategies to facilitate students learning to solve those types of problems. The research contexts of this project are calculus-based physics courses covering mechanics and electromagnetism. In phase I of the project, individual teaching/learning interviews were conducted with 20 students in mechanics and 15 students from the same cohort in electromagnetism. The students were asked to solve problems on several topics of mechanics and electromagnetism. These problems involved calculating physical quantities (e.g. velocity, acceleration, work, electric field, electric resistance, electric current) by integrating or finding the area under the curve of functions of related quantities (e.g. position, velocity, force, charge density, resistivity, current density). Verbal hints were provided when students made an error or were unable to proceed. A total number of 140 one-hour interviews were conducted in this phase, which provided insights into students’ difficulties when solving the problems involving the integral and the area under the curve concepts and the hints to help students overcome those difficulties. In phase II of the project, tutorials were created to facilitate students’ learning to solve physics problems involving the integral and the area under the curve concepts. Each tutorial consisted of a set of exercises and a protocol that incorporated the helpful hints to target the difficulties that students expressed in phase I of the project. Focus group learning interviews were conducted to test the effectiveness of the tutorials in comparison with standard learning materials (i.e. textbook problems and solutions). Overall results indicated that students learning with our tutorials outperformed students learning with standard materials in applying the integral and the area under the curve concepts to physics problems. The results of this project provide broader and deeper insights into students’ problem solving with the integral and the area under the curve concepts and suggest strategies to facilitate students’ learning to apply these concepts to physics problems. This study also has significant implications for further research, curriculum development and instruction.

Dialoger med användarna genomfördes för att förstå deras prioriteringar och vanor. En Participatory Design metod användes för att främja användarnas känsla av delaktighet och ägandeskap av appen. Användarna föreslog idéer som att inkludera symboler för användare som inte kan läsa och kopiera familjära element från Mexiko mest använda kommunikationsapp, WhatsApp. Användarvänlighetstesten visade att designen var användbar. Testen gav även rum för en naturlig dialog och appropriering av appen. Vidare, diskuterar jag möjligheter samt begränsningar med applikationen.

Genom att delta kan de göra sitt perspektiv synligt. Under det senaste årtiondet har barns användning av interaktiv teknik ökat kraftigt. Applikationer och web-sidor används nuförtiden även flitigt inom barnsjukvården. Avhandlingens övergripande syfte var att utveckla och pröva, tillsammans med barn, ett interaktivt kommunikationsverktyg avsett att möjliggöra yngre barns delaktighet i vårdsituationer. Intervjuer, teckningar och vignetter användes för att, fenomenografiskt, förstå barnens uppfattningar av att vara i en vårdsituation (I). En iterativ deltagarbaserad design användes för att utveckla en prototyp av en applikation. En hermeneutisk tolkning av video-observationer fångade meningen i barnens sätt att visa sin delaktighet (hintar) vid användningen av applikationen i vårdsituationer (III). En deduktiv, kvantitativ ansats användes för att identifiera mönster i barnens sätt att visa sin delaktighet när de använde applikationen (IV). Resultatet visade att barnen uppfattade sig själva, föräldrarna och vårdpersonalen som aktörer i situationen, även om alla uppfattades agera på olika sätt (I). Barnen bidrog i den iterativa processen med viktig information gällande aspekter som åldersanpassning, användbarhet och hur väl den tilltalar dem, vilket slutligen ledde fram till den färdiga applikationen (II). Barnens sätt att visa sin delaktighet när de använde applikationen förstods ha en nyfiken, tankfull och självbekräftande mening (III). Nyfikenheten visades mest vid användningen av applikationen. Treåringarna samt barnen med minst vårderfarenhet använde applikationen i störst utsträckning (IV). Sammanfattning: när applikationen användes i vårdsituationen visade barnen en situerad delaktighet vilken byggde på deras perspektiv på den aktuella situationen samt på deras inter-inter-aktion med applikationen och vårdpersonalen. Genom detta erbjöds vårdpersonalen ytterligare ett sätt att guida barnet utifrån barnets eget perspektiv.

Der volle Inhalt der QuelleGenetic modification of dendritic cells with plasmid DNA is plagued with low transfection efficiencies because DNA taken up by non-dividing dendritic cells rarely reaches the nucleus. But this difficulty can be overcome by the use of messenger RNA (mRNA), which exerts its biological function in the cytoplasm and obviates the need to enter the nucleus. Since pioneering work of Boczkwoski et al, the ex-vivo application of mRNA-transfected dendritic cells as a vaccine has been evaluated in numerous phase I trials worldwide and is still currently being actively optimized in clinical trials.
However, a major disadvantage of using mRNA-transfected DCs as a vaccine is that it requires patients to undergo at least one 4-hour leukapheresis procedure, followed by separation of the peripheral blood mononuclear cells (PBMCs), from which monocytes are isolated and cultured for a week in a defined medium with cytokines. The resulting DCs are matured after being loaded with mRNA and frozen for storage. Aliquots are subsequently thawed prior to administration to patients. This process of harvesting, culturing and loading DCs is more time- and resource-intensive than Provenge, the first FDA approved cell based tumor vaccine in 2011.Recent evidence has confirmed a lack of broad translation of Provenge due to complexity and cost of treatment. This predicates a similar fate for mRNA-transfected dendritic cell vaccine going forward.
This thesis presents alternative delivery strategies for mRNA mediated tumor vaccination. Through the application of synthetic and natural biomaterials, this thesis demonstrates two viable approaches that reduce or eliminate the need for extensive manipulation and cell culture.
The first approach is the direct in vivo delivery of mRNA encapsulated in nanoparticles for tumor vaccination. A selected number of synthetic gene carriers that have been shown to be effective for other applications are formulated with mRNA into nanoparticles and evaluated for their ability to transfect primary DCs. The best performing formulation is observed to transfect primary murine and human dendritic cells with an efficiency of 60% and 50% (based on %GFP+ cells) respectively. The in vivo transfection efficiency and expression kinetics of this formulation is subsequently evaluated and compared with naked mRNA via various routes of delivery. Following this, a proof-of-concept study is presented for a non-invasive method of mRNA tumor vaccination using intranasally administered mRNA encapsulated in nanoparticles. Results show that intranasally administered mRNA induces tumor immunity only if it is encapsulated in nanoparticles. And anti-tumor immunity is observed in mice intranasally immunized under both prophylactic as well as therapeutic models.
The second approach evaluates whole blood cells as alternative cell based mRNA carriers. A method is developed to encapsulate intact and functional mRNA in murine whole blood cells. Whole blood cells loaded with mRNA not only include erythrocytes but also T cells (CD3+), monocytes (CD11b), antigen presenting cells (MHC class II) as well as plasmacytoid DCs (CD45R-B220). Mice immunized with mRNA-loaded whole blood cells (intravenously) develop both humoral and cellular antigen-specific immune responses, and demonstrate delayed tumor onset and progression in a melanoma therapeutic immunization model (using tyrosinase related protein -2, TRP-2, as an antigen). Importantly, the therapeutic efficacy of mRNA-loaded whole blood cell vaccine formulation is found to be comparable to mRNA-transfected dendritic cell vaccine.
In conclusion, this thesis presents new methods to the delivery of mRNA tumor vaccines that reduce or eliminates the need for extensive cell manipulation and culture. Results presented in this thesis reveal viable research directions towards the development and optimization of mRNA delivery technologies that will address the problem of broad translation of mRNA tumor vaccines in the clinics.

ABSTRACT The purpose of the thesis is to illustrate the application of advanced technologies in securing and facilitating international trade while improving public welfare. In addition, hopefully the thesis could help profile the rapidly modernized Taiwan Customs, actively enhance the international image of Taiwan Customs, as well as enable customs officers, who specialize in other customs related jobs, to understand how well Taiwan Customs applies the advanced law enforcement technologies in fighting against illicit trafficking. With the trend of technological development, more and more advanced technologies are applied in specialized fields, i.e. the application of RFID, X-ray, as well as radiation detectors, both in private and public sector, especially the applications by Taiwan Customs. Globalization is resulting in increasingly expanded flows of goods, information and technology. However, this is not only beneficial to legal trade; it benefits illegal trade too. These developments are resulting in the growth of the complexity of risks that have to be managed at the border. The lack of effective controls presents risks to the economy and society and can undermine gains that have been made. Therefore, Taiwan Customs has adopted and applied high-tech mobile X-ray Container Inspection Machines and conducted the non-intrusive inspection of the containers that pose high risk to enhance the security and efficiency for the shipments of container cargos. Taiwan Customs also applies radiation portal monitors to detect radiation, as well as handheld devices to identify the radioactive isotope. We call the important measure as Megaports Initiative, an initiative which aims at actively detecting and interdicting illicit trafficking in nuclear and radioactive materials across borders. Moreover, Taiwan Customs becomes the first public sector around the world after successfully applying RFID E-seal to escort the security for transshipment container. After 911, the issue on supply chain security has attracted worldwide attention. WCO develops SAFE Framework of standards to secure and facilitate global trade, and encourages governments to promote AEO system. Operators, who meet the security criteria for AEO and are certified by Taiwan customs, will be offered preferential measures to reduce goods inspection rate and expedite customs clearance. Furthermore, through international mutual recognition, the AEOs will enjoy more preferences. Therefore, Taiwan Customs is actively implementing AEO Certification and Management System, with the aim of bringing Taiwan Customs to come up with the standard. The World Customs Organization (WCO) is actively encouraging the use of the WCO Data Model in an attempt to guide countries throughout the world to build a standard customs clearance operation model, and a one-stop export/import service structure, so that cooperation among the Customs and exchange of Customs clearance data can be achieved. To meet with the standard, Taiwan Customs launches CMT Single Window Plan aiming at integrating the existing Customs Clearance System, Marine Port Information System and Facile Trade Net. In addition, Taiwan Customs also actively promotes the Advance Cargo Information service to provide public sectors with relevant electronic cargo information well ahead of the arrival of the goods to help profile and target the goods that may pose a security risk. Taiwan Customs will succeed to working on the Ubiquitous Economic and Trade Network Plan to achieve the goals of trade facilitation and security, and will try its best to conform to international norms and standards to positively enhance the international image of Taiwan Customs.

Physicians and nurses performing clinical application based on the evidence-based healthcare (EBHC) to improve quality of patient care is an important issue. In particular, the physicians and nurses of EBHC competitions are already competent with specific EBHC practices and aware of EBHC clinical application. Whether the clinical application of these EBHC practices reaches the ‘adhered to’ stage is a critical issue requires further investigation. This study was first to understand the distribution of the EBHC clinical application in seven action stages, then to explore factors influencing the behavior of EBHC clinical application by adopting the Decomposed Theory of Planned Behavior (DTPB), and finally to identify factors predicting the ‘adhered to’ stage of EBHC clinical application. Methods: The study employed a cross-sectional study design. The total population of physicians and nurses of EBHC competitions was 631. A survey was conducted using a structural questionnaire. Data was collected from 312 of the population; the returning rate 49.45%. The overall model of the study was analyzed using the structural equation modeling. Results: In the distribution of seven action stages (7As), 33.3% of the survey reported the ‘adhered to’ stage. In DTPB model testing revealed that all hypotheses, except for the association between ease of use and attitude and the association between technology facilitating condition and perceived behavioral control, were supported by the data with good overall model fit. The influences of behavioral intention and perceived behavioral control on the behavior of EBHC clinical application was positive with 37% variance of the behavior explained. As for predicting the ‘adhered to’ stage of EBHC clinical application, the odds for supervisory vs. nonsupervisory was 2.03 (OR=2.03, CI=1.10-3.77) resource facilitating condition, perceived behavioral control, and behavioral intention each incremental point the odds increased by 1.06 (OR=1.06, CI=1.01-1.11), 2.21 (OR=2.21, CI=1.47-3.32) and 1.96 (OR=1.96, CI=1.40-2.73) times respectively. Conclusion and recommendation: The study found that: perceived usefulness, compatibility, peer and superior influences, self-effcacy, resource facilitating condition, attitude, subjective norm, perceived behavioral control, and behavioral intention influencing EBHC clinical application; supervisory duties, resource facilitating condition, perceived behavioral control, and behavioral intention predicting the ‘adhered to’ stage of EBHC clinical application. Therefore, we recommend as follows: promoting continuing education for nonsupervisory physicians and nurses to improve knowledge and skill related to EBHC clinical application; installing databases and equipments necessary for EBHC clinical application as well as encouraging involvement of EBHC competitions. The study model based on the DTPB model revealed a good fit and the questionnaire was valid and reliable hence suitable for measuring the behavior of EBHC clinical application and exploring other relevant factors facilitating the behavior of EBHC clinical application in future studies.
