ABSTRACT Cell-bound C-signal guides the building of a fruiting body and triggers the differentiation of myxospores. Earlier work has shown that transcription of the csgA gene, which encodes the C-signal, is directed by four genes of the act operon. To see how expression of the genes encoding components of the aggregation and sporulation processes depends on C-signaling, mutants with loss-of-function mutations in each of the act genes were investigated. These mutations were found to have no effect on genes that are normally expressed up to 3 h into development and are C-signal independent. Neither the time of first expression nor the rate of expression increase was changed in actA, actB, actC, or actD mutant strains. Also, there was no effect on A-signal production, which normally starts before 3 h. By contrast, the null act mutants have striking defects in C-signal production. These mutations changed the expression of four gene reporters that are related to aggregation and sporulation and are expressed at 6 h or later in development. The actA and actB null mutations substantially decreased the expression of all these reporters. The other act null mutations caused either premature expression to wild-type levels (actC) or delayed expression (actD), which ultimately rose to wild-type levels. The pattern of effects on these reporters shows how the C-signal differentially regulates the steps that together build a fruiting body and differentiate spores within it.

Qu’ont en commun l’art spiritualiste de Hector de Saint-Denys Garneau et le monisme de Paul-Émile Borduas ? Premier poète québécois à faire de la peinture un objet de réflexion esthétique, Saint-Denys Garneau tient pourtant en suspicion la modernité artistique, refusant « l’injure à l’art » que représentent pour lui le cubisme et l’art abstrait. Si l’ekphrasis, chez Saint-Denys Garneau, s’éloigne du « pittoresque » pour s’approcher du « pictural », l’idéalisation du sujet et l’objectivation de la vision qu’il cultive diffèrent cependant fortement de la conception du tableau de Borduas. Aux notions d’ordre, de centre, d’harmonie, d’unité, de transparence et d’élévation autour desquelles le poète et artiste ordonne encore le visible, Borduas oppose celles d’automatisme psychique, d’écriture automatique, de liberté du geste, de « désir-passion », de « grâce surrationnelle », appelant une exploration du chaos du monde intérieur et une plongée dans l’inconscient menant inéluctablement à un contact brutal avec la matière sensible. Prenant acte de l’apparition de Garneau et Borduas au même moment sur la scène publique montréalaise, vers 1937-1938, et cherchant à mesurer le pouvoir de séduction particulier qu’exercent sur eux durant ces années décisives la pensée et l’oeuvre de John Lyman, l’auteur interroge la conception antagoniste que chacun se forge du dessin et de la couleur et examine certains des enjeux essentiels, après la publication du Journal de Saint-Denys Garneau, qui déterminent la posture double et ambiguë de Borduas à l’endroit de l’héritage de l’écrivain.

ABSTRACT Mutations within the −12 and −24 elements provide evidence that the act promoter is recognized by sigma-54 RNA polymerase. Deletion of the −20 base pair, which lies between the two conserved elements of sigma-54 promoters, decreased expression by 90%. In addition, mutation of a potential enhancer sequence, around −120, led to an 80% reduction in act gene expression. actB, the second gene in the act operon, encodes a sigma-54 activator protein that is proposed to be an enhancer-binding protein for the act operon. All act genes, actA to actE, are expressed together and constitute an operon, because an in-frame deletion of actB decreased expression of actA and actE to the same extent. After an initially slow phase of act operon expression, which depends on FruA, there is a rapid phase. The rapid phase is shown to be due to the activation of the operon expression by ActB, which completes a positive feedback loop. That loop appears to be nested within a larger positive loop in which ActB is activated by the C signal via ActA, and the act operon activates transcription of the csgA gene. We propose that, as cells engage in more C signaling, positive feedback raises the number of C-signal molecules per cell and drives the process of fruiting body development forward.

Abstract This article studies the discourse in the late eighteenth- and early nineteenth-century French periodical press on the topic of translations of travel writing. It reveals that travel reviews were arenas for discussing the political and ideological value of translating travelogues into French, notably from English. In the context of the Franco-British conflicts at the turn of the century, the French press perceived translations of British travel writing as potential patriotic tools that allowed different ways of countering or subverting British global influence. Paratextual elements of translations, the translator’s prefaces and notes, appeared to be particularly important in this respect. By analysing the periodical discourse on travel book translations, the article shows how travel writing was constructed as a politically invested genre.

Abstract Alternative Complex III (ACIII) is an example of the robustness and flexibility of prokaryotic respiratory chains. It performs quinol:cytochrome c oxidoreductase activity, being functionally equivalent to the bc1 complex but structurally unrelated. In this work we further explored ACIII investigating the role of its monoheme cytochrome c subunit (ActE). We expressed and characterized the individually isolated ActE, which allowed us to suggest that ActE is a lipoprotein and to show its function as a direct electron donor to the caa3 oxygen reductase.

<p class="abstract"><strong>Background:</strong> <span lang="EN-IN">Acne is a chronic inflammatory disease known to occur in adolescent age group. There are many myths and misconceptions in patients as well as health physicians regarding the causes and treatment of acne. Objective of the study was to assess the knowledge, attitude and practice towards acne vulgaris among acne patients attending skin outpatient department in a private medical college.</span></p><p class="abstract"><strong>Methods:</strong> This is a cross sectional, questionnaire based study conducted between January - April 2016. 100 patients having acne lesions were included in the study.<strong></strong></p><p class="abstract"><strong>Results:</strong> <span lang="EN-IN">Majority of the study subjects belonged to the age group 15-24. The study showed that 72% had good knowledge. More than half of the study subjects had wrong belief that eating oily foods, chocolates, spicy food caused acne but more than 40% of the study subjects had good knowledge about the causes and aggravating factors like it worsens by squeezing/picking/rubbing (83%), commonly found in oily skin (67%), has seasonal occurrence (54%), associated with premenstrual flare (42%), aggravated with use of cosmetics (41%). </span></p><p class="abstract"><strong>Conclusions:</strong> <span lang="EN-IN">The acne patients had poor practice and unfavorable attitude in spite of good knowledge. Many myths exist among patients. Despite being so common and very well responsive to treatment, it is a major cause of depression among patients.</span></p>

Abstract Nephroblastomas or Wilms tumors are the most common pediatric renal cancer. Across all cases, the 5-year overall survival is over 90%. This is largely in part due to brute-force, chemotherapy regimens such as dactinomycin (actD) that have been routinely used to treat Wilms tumors. However, those that do respond to treatment still face a 15% chance of relapse. Patients with anaplastic tumor histology require more toxic therapies, and yet they still face a worse prognosis of only 66% 4-year survival and relapse rate of 43%. Furthermore, recurrence cuts survival rate down to 50%. With the widespread use of actD in Wilms tumors and other childhood malignancies, understanding the mechanism to recalcitrance against this chemotherapy can potentially reveal important vulnerabilities that can be exploited in the clinic. While transcriptomic and genomic analyses have been done in Wilms tumors, no targetable pathways have been identified in actD chemotherapy-resistant and anaplastic tumor cases. To that end, we performed ribosome sequencing of actD-treated anaplastic Wilms tumor cell line WiT49 and uncovered that the proteasome pathway is preferentially translated. Moreover, loss of proteasome subunits in a CRISPR library dropout screen sensitized the cells to actD. Consistent with these findings we found upregulation of proteasome subunit levels and proteasome enzymatic activity in Wilms tumor lines following actD treatment. To interrogate the effect of cotreating bortezomib with dactinomycin in vivo, we subcutaneously implanted anaplastic Wilms tumor patient-derived xenograft (PDX) lines in NOD/SCID Gamma (NSG) mice. Combinatorial treatment was found to be retard tumor growth compared to vehicle and single drug conditions. With further investigation, our findings can provide a foundation for exploring proteasome inhibitors with dactinomycin in clinical trials. Our study has important implications to improving the outcomes for not just for recurrent and anaplastic Wilms tumors, but to other pediatric cancers that use actD as well. Abstract Actinomycin D (ActD) was the first anti-cancer antibiotic approved for the management of human cancers. However, the notorious toxicity profile limits its widespread application in cancers including cancers of the aerodigestive tract. Recent studies show that combining low-dose ActD with existing chemotherapies could potentially reduce the toxicity of chemotherapy drugs through p53 mediated cyclotherapy (protection of normal cells while eliminating cancer cells), a recently coined concept. An understanding of ActD’s effect on p53 signaling is critical for meaningful application of ActD in cyclotherapy-based combinations. This study evaluated the antitumor efficacy and mechanism of action of ActD in aerodigestive tract cancers. We found that ActD strongly inhibited the growth of a panel of aerodigestive tract cancer cell lines and induced efficient apoptosis although the sensitivity varies among cell lines. The IC50 values of ActD spanned between 0.021-2.96nM. Mechanistic studies revealed that ActD time- and dose-dependently increased the expression of total and phosphorylated p53 (ser15). Furthermore, ActD-induced apoptosis is dependent on p53 in cells expressing wild-type p53. Further studies demonstrated that ActD induced context-dependent differential expression of downstream targets p21 and PUMA without significant effects on p27. Interestingly, our study revealed that p53-p21 is the predominant pathway activated by low-dose ActD in cells with wild type p53. Therefore, our findings further rationalize the premise for future studies to explore the application of low-dose ActD as a chemoprotectant for cyclotherapy combinations in aerodigestive tract cancers. <p class="abstract"><strong>Background:</strong> Acne vulgaris is a chronic inflammatory dermatosis of pilosebaceous unit. Adult acne is defined as presence of acne after the age of 25 years. Though acne vulgaris has been studied extensively, there is paucity of literature on adult acne.</p><p class="abstract"><strong>Methods:</strong> A total of 150 patients with adult acne were enrolled in the study. A detailed history was taken followed by detailed dermatological examination. Ultrasonography abdomen and pelvis, hormonal evaluation was done in case of women with symptoms and signs of hyperandrogenism.<strong></strong></p><p class="abstract"><strong>Results:</strong> Out of 150 patients studied, majority (61.3%) were in the age group of 26-30 years. The mean age of patients with adult acne was 30.1 years. Females (86%) outnumbered males. Majority of the patients (53.3%) had persistent adult acne and 46.7% had late onset acne. Exposure to sunlight (24%) was the most common aggravating factor followed by use of cosmetics in 19.3%. Menstruation was causing flare up of acne in 72 (55.8%) female patients. Face was involved in all the patients. Cheek was the most common site involved followed by forehead. Post acne scarring was present in 46.6% of the patients. Majority of the patients (67.3%) had grade 2 acne. Symptoms and signs of hyperandrogenism were present in 14 female patients.</p><p class="abstract"><strong>Conclusions:</strong> Adult acne is more common in females. Persistent acne is more common than late onset acne. Patients with persistent adult acne are more prone to develop nodulocystic acne and acne scars.</p>

In its 2021 ruling in Consorzio, the European Court of Justice adopted a more pragmatic approach to its acte clair doctrine. This article analyses this reform and shows that in important respects the Court has eased the requirements for finding a question of EU law to be acte clair. Simultaneously, the Court has introduced a duty for national courts of last instance to give reasons where they are faced with questions of EU law and decide not to make a preliminary reference. The article also considers the Court’s discreet change of approach regarding the application (rather than the interpretation) of EU law by national courts of last instance as reflected in the Consorzio ruling, and argues that there is acte clair where the abstract interpretation of EU law is clear and leaves no scope for any reasonable doubt, even if the national court of last instance entertains doubts as to the correct application of said EU law to the case at hand.

Deux définitions du terrorisme coexistent au Canada, l’une en matière pénale et l’autre dans le domaine de l’immigration. Le droit suisse n’incrimine que le financement du terrorisme. Contrairement au droit canadien, la Suisse a opté pour une norme générale et abstraite inspirée de la Convention internationale pour la répression du financement du terrorisme. La loi suisse n’exige pas que l’acte terroriste soit commis au nom d’un but, d’un objectif ou d’une cause de nature politique, religieuse ou idéologique. Cette solution est préférable. Par contre, le législateur suisse a exclu expressément l’incrimination du dol éventuel. Le droit canadien offre, dans ce cas, une possibilité d’incrimination plus large, davantage favorable à la mise en œuvre effective du droit pénal. En outre, le droit suisse ne sanctionne que le soutien financier à un acte terroriste précis, le droit canadien ayant dans ce cas aussi un champ d’application plus étendu et plus adapté. En droit canadien, l’incrimination du financement du terrorisme pourrait s’inspirer de la formulation suivante : « Est coupable d’un acte criminel passible d’un emprisonnement maximal de dix ans quiconque, directement ou non, fournit ou réunit des fonds, dans l’intention de les voir utiliser ou en sachant qu’ils seront utilisés pour financer un acte — ou une entité dont l’un des objets ou l’une des activités est de se livrer à ce type d’actes — destiné à tuer ou blesser grièvement un civil, ou toute autre personne qui ne participe pas directement aux hostilités dans une situation de conflit armé, lorsque, par sa nature ou son contexte, cet acte vise à intimider une population ou contraindre un gouvernement ou une organisation internationale à accomplir ou à s’abstenir d’accomplir un acte quelconque ».

<p class="abstract"><strong>Background:</strong> Acne vulgaris is a chronic inflammatory disease of pilosebaceous units which has great psychological impact and causes depression, anxiety, and low self-esteem.</p><p class="abstract"><strong>Methods:</strong> It was a hospital-based, cross-sectional, prestructured, questionnaire-based study done in 120 individuals in the age group 12-30 years using two specific HRQoL indices-Cardiff Acne Disability Index (CADI) and Acne-specific Quality of Life (Acne-QOL) to understand the impact of acne on the quality of life.<strong></strong></p><p class="abstract"><strong>Results:</strong> Male to female ratio was 2:3. The mean age was 19.23±1.7. The mean CADI score was 5.3. Majority of the patients had grade 2 acne. Based on the CADI questionnaire, the number of males (61.7%) and females (60.2%) was maximum in the medium score. Grade 1 acne had maximum number of patients showing low score followed by grade 2 acne. Based on the acne-QoL questionnaire, maximum patients were in the 11-20 score range, followed by the 21-30 score range.</p><p class="abstract"><strong>Conclusions:</strong> Acne is not only a cosmetic problem, but also has significant psychological impact. Hence, impact of acne on QoL must be considered in the management of facial acne.</p>

<p class="abstract"><strong>Background:</strong> Acne vulgaris is a seemingly harmless disease affecting majority of adolescents. It is actually a psycho-dermatological disorder associated with psychological trauma. The impact of acne on the quality of life of Indian patients remains undocumented. A study is undertaken to assess the impact of acne on health, family, emotional and social wellbeing of acne patients.</p><p class="abstract"><strong>Methods:</strong> 100 patients between age group of 13 to 19 years with acne vulgaris, attending St. Antony’s Skin Clinic, Pala, Kerala, between April 2010 and March 2012 were selected for study. The control group consisted of 100 adolescents without acne. Both groups had no other dermatological or systemic disease. Acne cases were graded using global acne grading system. Quality of life was assessed using WHO QOL BREF.Four domains were derived from this, physical and psychological health, social relationship and environment. Adjustment levels were assessed using adapted version of bell adjustment inventory.<strong></strong></p><p class="abstract"><strong>Results:</strong> The parameters assessed were quality of life, family, social, emotional, health related and overall adjustments in those with and without acne and severity of acne in relation to quality of life and all values were significantly in favour of those without acne.</p><p class="abstract"><strong>Conclusions:</strong> Acne affects personality, self-esteem and self-image of patients. It affects social interaction. Adolescents with acne report low quality of life and resultant poor psychosocial adjustment. There is an inverse relation between severity of acne and quality of life. The severity levels and adjustment also has an inverse relationship.</p>

<div class="Section1"><p class="abstract"><strong>Background:</strong> Acne vulgaris is a multifactorial skin disease. A potential role for insulin-like growth factor-1 (IGF-1) has been suggested in the pathogenesis of acne. Several studies have shown that elevated levels of serum IGF-1 correlate with overproduction of sebum and acne. Objective: Measurement of the serum level of IGF-1 in acne patients in comparison to normal controls and evaluating the relationship of these levels to severity of acne and body mass index (BMI), in order to investigate the role of this factor in the pathogenesis of acne.</p><p class="abstract"><strong>Methods:</strong> Fifty-four patients with acne vulagaris and 42 healthy controls were included. History taking, dermatological examination, clinical assessments of acne severity, calculation of BMI were performed for patients. Blood samples were collected from all participants for estimation of serum IGF-1 level using enzyme linked immunosorbant assay.<strong></strong></p><p class="abstract"><strong>Results:</strong> There was a significantly higher serum IGF-1 level in acne patients (p&lt;0.05) than controls. Authors didn’t find a relation of significance between elevated serum IGF-1 level and degree of acne severity and BMI (p&gt;0.05). There was a significant positive correlation between serum IGF-1 level and age of the patients.</p><p class="abstract"><strong>Conclusions:</strong> There is a significantly higher serum IGF-1 in acne patients than controls not related to acne severity and BMI. That is adding to the scientific evidence of IGF-1 role in pathogenesis of acne vulgaris.</p></div>

It is usually prevalent in adolescent may persist in adulthood. This study was conducted to evaluate the quality of life (QoL) in patients suffering from acne vulgaris.</p><p class="abstract"><strong>Methods:</strong> Hospital based cross sectional study was conducted in 60 patients. Dermatology life quality index (DLQI) questionnaire was used to observe the effect of acne among patients.</p><p class="abstract"><strong>Results: </strong>Total numbers of patients enrolled were 60 (43 females,17 males). The mean age was 24.85±5.63 years. Maximum patients were of the age group 26-30 years (22 patients; 36%) and least were of 31-36 age group. Out of 60 patients, 29 patients had grade II acne followed by 20 with grade I acne, grade III acne was present in 11 patients. Most of the patients 31 (52%) were having “moderate impact” on quality of life followed by 23 (38%) with “small impact” on QOL. There was a significant positive correlation between grade of acne and DLQI (Pearson correlation=0.753, p=0.00).</p><p class="abstract"><strong>Conclusions: </strong>Our study showed significant impairment of QoL in acne patients. DLQI increases with increase in severity of acne. Treatment of acne should not be guided only by the clinical grade of acne, but take into consideration the psychosocial impact the condition has on the patient. Adequate counselling along with early treatment of acne vulgaris is essential in order to reduce the disease related psychosocial sequelae and increase treatment efficacy.</p>

Though not life threatening it can cause physical and psychological scars which can produce social problems in house, school and office. It is a multifactorial disease. Many treatment options are available, but not optimal. Zinc was found to be effective in pustular acne in a patient with acrodermatitis enteropathica. Hence a study on the level serum zinc in acne patients and the effect of zinc on acne is worthwhile. The aim was estimation of serum zinc level in acne patients and correlation of serum zinc level and improvement of acne.</p><p class="abstract"><strong>Methods:</strong> Pre-treatment serum zinc level was estimated in acne and non-acne patients. Acne patients were given oral zinc 220 mg two times a day and control group was given placebo. Post treatment serum zinc levels were estimated and the serum zinc level and improvement of acne was correlated.<strong></strong></p><p class="abstract"><strong>Results:</strong> Study subjects attained average serum zinc level of 5.829 µg/ml from 1.33 µg/ml. 59.5% patients on zinc therapy showed more than 50% improvement in acne severity. 10.6% showed &gt;75%, 48.9% showed 50-75%, 36.1% showed 25-50% and 4.25% patients showed less than 25% improvement.</p><p class="abstract"><strong>Conclusions:</strong> Statistically significant low levels of zinc were not observed in acne. Oral zinc sulphate 220 mg two times a day raised serum level to 5.82 µg/ml Exponential response in acne was noticed as serum zinc level was raised. Inflammatory lesions showed better response than comedo. Oral zinc has definite role in treatment of acne.</p>

Adult acne is more common in women and may be a marker of hyperandrogenism.The present study was carried out to study the clinical profile of acne vulgaris in semiurban patients.</span></p><p class="abstract"><strong>Methods:</strong> <span lang="EN-IN">The study was conducted for 1 year on all eligible patients of acne vulgaris attending dermatology OPD of a hospital catering to semiurban patients. Data including grade of acne, markers of androgenicity and post-acne scarring was collected. Statistical analysis was done using SPSS 15 software</span>.<strong></strong></p><p class="abstract"><strong>Results:</strong> <span lang="EN-IN">Frequency of acne vulgaris patients in the dermatology OPD was 2.8% with 429 acne patients out of 15,322 new patients. Female to male ratio was 1.44:1. The mean age of onset in males was 16.24 and in females, 14.84 years. Grade 2 acne (66%) was commonest followed by grade 3 (5.1%) and grade 4 (3.7%). Associated conditions seen were seborrhea (60.8%), alopecia (18.9%), acanthosis (4.9%) and PCOD (2.3%). Markers of androgenicity were more frequently seen in grade 2 acne which was statistically significant. Atrophic scars were commonest including icepick scars (80.2%), rolling scars (67.4%) and box scars (57.8%). </span></p><p class="abstract"><strong>Conclusions:</strong> <span lang="EN-IN">In our study females had an earlier onset and closed comedones were the most common acne lesion. Grade 2 acne vulgaris formed majority of patients. Grade 4 acne vulgaris was more common in males.</span></p>

This study was done<strong> </strong>to evaluate the dermatology life quality index in patients with acne vulgaris.</p><p class="abstract"><strong>Methods:</strong> A cross sectional study was done with pretested Dermatology life quality index (DLQI) questionnaire on 100 acne patients 15 years and above, in a tertiary care hospital. Interpretation of score and impact on quality of life (QoL) was done and graded as no effect, small effect, moderate effect, and very large effect.<strong></strong></p><p class="abstract"><strong>Results:</strong> In our study, females outnumbered males (M: F ratio=1: 1.22). The age group most affected was within 15 and 20 years of age. The mean DLQI score was 5.59±4.07. Most of them had a ‘small impact’ on the QoL. QoL was worse in females. The most common grade of acne overall was grade 2 acne. Grade 3 acne was more common in men. Most of the patients (84%) experienced some degree of embarrassment or increased self-consciousness due to their acne.</p><p class="abstract"><strong>Conclusions:</strong> Our study showed significant impairment of QoL in acne patients. Treatment of acne should not be guided only by the clinical grade of acne, but take into consideration the psychosocial impact the condition has on the patient. Adequate counselling along with early treatment of acne vulgaris is essential in order to reduce the disease-related psychosocial sequelae and increase treatment efficacy.</p>

Abstract Purpose: There is an inverse relationship between diagnosis to initiation of treatment interval and outcome of cancer treatment. A significant number of diagnosed cervical cancer patients don’t receive timely treatment in low resource settings. The Cervical Cancer Chemotherapy Access Program (C3AP) is one of End Cervical Cancer Nigeria Initiative’s interventions to IMProve Access to Cervical Cancer Treatment (IMPACT) in low resource settings. This report provides documentary evidence of the evolution of the implementation of the C3AP and highlights best practices that can be scaled up. Methods: The C3AP was conceptualized to increase the proportion of diagnosed cervical cancer patients that receive timely treatment by exploring adjuvant and neoadjuvant role of chemotherapy in the management of cervical cancer patients in settings where access to radiotherapy services can not be guaranteed. We instituted Plan Do Study and Act (PDSA) quality improvement cycle for the C3AP and used RE-AIM framework to study the implementation processes to be acted upon as part of the PDSA quality improvement process towards improving reach of the program. Results: The C3AP supported treatment of only 4 patients over the reporting period of 5months. Poor referral to the program is a major hurdle the program is battling. Only 2(17%) of the 12 confirmed cases of cervical cancer diagnosed in the facility within the 5month study period were able to access chemoradiation, suggesting a missed opportunity for 6/10(60%) that couldn’t afford chemoradiation. The program evolved over a PDSA cycle to eliminate social eligibility in the criteria to access the support thereby able to shorten application -support interval to same day with potential positive impact on prognosis. Conclusion: Providing equitable support for indigent cancer patients should be beyond the emotion to help humanity. We recommend the use of structured quality improvement for all programs providing support to indigent cancer patients to ensure implementation fidelity. Abstract A series of 2,5-disubstituted-benzoxazole derivatives (1–13) were evaluated as possible inhibitors of acetylcholinesterase (AChE) and butyrylcholinesterase (BChE). The results demonstrated that the compounds exhibited a broad spectrum of AChE and BChE inhibitory activity ranging between 6.80% and 90.21% except one compound which showed no activity against AChE at the specified molar concentration. Another derivative displayed a similar activity to that of reference drug (galanthamine) for inhibition of AChE and BChE. In addition, molecular docking of the compounds into active site of AChE was performed using recombinant human AChE (PDB ID: 4ey6) in order to understand ligand–protein interactions.

<p class="abstract"><strong>Background:</strong> Acne is one of the most common inflammatory chronic skin diseases that affect teenagers and continues into adulthood. Self-medication is common among medical students. Hence this present study was undertaken to study the knowledge, attitude and practice of self-medication for acne among medical students.</p><p class="abstract"><strong>Methods:</strong> This cross-sectional study included medical students of the second phase and final phase (Part I and Part II). A pretested questionnaire was given to them which included questions on knowledge, attitude and practice about self-medication for acne.<strong></strong></p><p class="abstract"><strong>Results:</strong> Self-medication for acne was seen in 240 (77.4%) students out of 310 students. Mild nature of illness was the most common reason for self-medication for acne followed by privacy. Most common source of information was seniors/friends/family members. Most of the students felt that self-medication is part of self-care (48%). Clindamycin was the most common drug self-medicated for acne.</p><p class="abstract"><strong>Conclusions:</strong> Self-medication for acne is quite high among medical students. Self-medication is part of self-care so it should be encouraged for minor illness. But should be based on thorough knowledge and restricted to over the counter drugs.</p>

<p class="abstract"><strong>Background: </strong>To study clinical and epidemiological characteristics and hormonal profile of adult females with acne</p><p class="abstract"><strong>Methods: </strong>Adult female patients of &gt;25 years of age with acne were enrolled. Demographic profile, habits, location of acne lesions, associated disease and acne flare association with menstrual cycle were noted. Prevalence of abnormalities in testosterone, dihydroepiandrosterone-sulfate (DHEAS), prolactin, luteinizing hormone (LH), follicle stimulating hormone (FSH), LH:FSH ratio, anti-mullerian hormone (AMH), and serum insulin were noted. Ultrasound (abdomen and pelvis) was performed for screening the presence of any abnormality.</p><p class="abstract"><strong>Results:</strong> Fifty one patients [mean (SD) age 30.96 (4.93) years] were included in the study. Thirty six (70.59%) patients had persistent type of acne. Grade 2 acne was present in 40 (78.43%) patients. Oily skin was seen in 27 (52.94%) patients. Hirsuitism and menstrual irregularity was present in 24 (47.06%) and 13 (25.49%) patients respectively. Pigmentation and scarring was present in 27 (52.94%) and 35 (68.63%) patients respectively. Acne lesions were seen on cheeks in 36 (70.59%) patients. Eight (15.69%) patients had elevated testosterone. DHEAS and prolactin levels were normal in all patients. Elevation of LH and decreased FSH levels were seen in five (9.80%) and three (5.88%) patients respectively. Five (9.80%) patients had elevated LH:FSH level. Raised insulin level was seen in 8 (15.69%) patients.</p><p class="abstract"><strong>Conclusions: </strong>Derangement of hormonal profile is not very common among adult female patients with acne. However, in some patients laboratory markers of hyperandrogenism are seen. </p>

<p class="abstract"><strong>Background:</strong> Acne is one of the most common disease affecting adolescence. Although prior studies have evaluated epidemiologic patterns of acne in different ethnicities and regions, literature is lacking from this part of the country. The objectives of the study were to delineate the clinical and epidemiological profile of acne vulgaris in a population from northern Kerala in South India.</p><p class="abstract"><strong>Methods:</strong> The study was carried out for 6 months and it included 100 patients. Epidemiology including pattern, aggravating factors, seasonal variation, diet, smoking, markers of androgenecity and associations were evaluated in all newly diagnosed cases of acne vulgaris who attended the outpatient department in Kannur Medical College, Kannur, Kerala, India from January 2018 to June 2018.<strong></strong></p><p class="abstract"><strong>Results:</strong> Out of 100 patients females (74%) outnumbered males (26%) and although the majority affected belonged to the age group of 21-25 years (38%), adult acne was observed in 28%. Grade 3 acne was the commonest (44%), followed by grade 2 (30%) and grade 4 (18%). Face was affected in all patients with cheeks (91%), followed by forehead (72%), mandible (36%) and chin (28%) and trunk (23%). Scarring was found in 34% and pigmentation in 40%. Family history of acne was observed in 33%.</p><p class="abstract"><strong>Conclusions:</strong> This study brings out the clinical profile of acne in a tertiary care hospital in South India.</p>

<p class="abstract"><strong>Background:</strong> Adolescent facial acne vulgaris impairs their quality of life (QOL). This impairment of QOL in has been documented to improve with treatment. In Nigerian adolescents who have facial acne vulgaris, it is not known if QOL improves with treatment. The aim was to assess QOL improvement with in adolescents who have facial acne vulgaris. To identify the socio-demographic, CADI QOL items and clinical characteristics related to this QOL improvement.</p><p class="abstract"><strong>Methods:</strong> This was a cross-sectional comparative study. One hundred and nine adolescents aged 15-20 years clinically examined for facial acne vulgaris. Quality of life before and after 6 weeks of treatment with 10% benzoyl peroxide was assessed using the Cardiff acne disability index. Data was analyzed using SPSS 16. The differences in quality of life scores were tested using non-parametric tests. Level of significance of all tests was p&lt;0.05.<strong></strong></p><p class="abstract"><strong>Results:</strong> Post-treatment, the median CADI score improved to 2 from a pre-interventional score of 4, p&lt;0.001. Improvement was observed in all the components of the CADI especially with the question, “how bad do you think your acne is now” with a percentage reduction from 81.4% to 54.6% and this improvement was in more males with improvement from 92% to 65%. Also, post-treatment, moderate to severe clinical severity of acne improved from 49.1% to 10.3%, p&lt;0.001.</p><p class="abstract"><strong>Conclusions:</strong> Treatment of adolescent facial acne leads to improvement of QOL especially in the perception of facial skin appearance and the severity of acne.</p>

Acne severity was graded using the global acne grading system(GAGS). Fasting glucose, fasting insulin levels were done and insulin resistance was assessed using homeostasis model assessment of insulin resistance (HOMA-IR)</span>.<strong></strong></p><p class="abstract"><strong>Results:</strong> <span lang="EN-IN">We did not find any statistically significant difference in fasting insulin levels between cases and controls. However, a weak positive correlation between acne severity and fasting insulin levels (r =0.3, p=0.04) were observed. Fasting glucose levels and HOMA-IR values observed between cases and controls were not statistically significant (p=0.05, p=0.59 respectively). </span></p><p class="abstract"><strong>Conclusions:</strong> <span lang="EN-IN">Our study did not suggest a major role of insulin resistance in acne.</span></p>

<abstract> <p>Achieving carbon neutrality requires high efficiency in agricultural carbon emissions. This study employs a super efficiency Slack Based Measure-Data Envelopment Analysis (SBM-DEA) model to measure the Agricultural Carbon Emission Efficiency (ACEE) of 31 provinces, cities, and autonomous regions within the Chinese Mainland from 2001 to 2021. Additionally, it utilizes the modified gravity model and a social network analysis to establish the spatial correlation relationship of ACEE, and extensively investigates the characteristics and transmission mechanism of China's spatial correlation network structure regarding ACEE. The findings reveal the following: 1) The spatial correlation relationship of China's ACEE from 2001 to 2021 exhibits a complex network structure; 2) in terms of the overall network structure characteristics of the spatial correlation, the ACEE network demonstrates a high degree of correlation and displays a stable temporal evolution trend; 3) concerning the centrality network structure characteristics of the spatial correlation, most provinces in China experience a continuous decline in point centrality and near centrality, while the interdependence of the ACEE between provinces increases; and 4) regarding the clustering characteristics of the spatial correlation, variations exist in the correlation among the four plates of the ACEE. However, they mostly assume a mediating role, and in 2021, the ACEE network sectors witnessed a robust interoperability.</p> </abstract>

Abstract Acne keloidalis nuchae (AKN) / folliculitis keloidalis nuchae (FKN) is a chronic inflammatory condition which involves hair follicles localized predominantly in occipital scalp and posterior neck area leading to hypertrophic scarring alopecia. We present a 59-year-old factory worker, Caucasian male with a whitish alopecic oval plaque about 10 cm in diameter in the occipital region. The peripheral part of plaque was mildly inflammated, with groups of tufted terminal hairs, while the central part showed cicatricial alopecia and discrete non-adherent dry scales. Skin changes firstly occurred 6 years earlier, as itchy papules and pustules that sometimes healed with scarring. The applied relevant diagnostic and therapeutical measures are discussed in this report.

Abstract Acne vulgaris is a common skin disease, which affects individuals of all races and ages. In Caucasians, almost 85% of individuals between 12 and 25 years, as well as 25% of adults, are affected with some forms of acne. The pathophysiology of acne is multifactorial, and thus, the treatment must cover all the possible causes of acne. For this reason, acne therapy is mostly a combination therapy, with the main goal to achieve clinical improvement, without scarring and residuals, as much as possible. The treatment should be planned individually, depending on the clinical appearance, severity and psychological profile of the patient. The treatment usually takes time and requires dedication and patience of both the patient and the physician.

Abstract Meristem is the most functionally dynamic part in a plant body. The shaping of the meristem requires constant cell division and cell elongation, which are influenced by hormones and cell cytoskeletal component, actin. Although the roles of hormones in modulating meristem development have been extensively studied, the role of actin in this process is still elusive. Using the single and double mutants of the vegetative class actin, we demonstrate that actin isovariant ACT7 plays an important role in root meristem development. In absence of ACT7, but not ACT8 and ACT2, depolymerization of actin was observed. Consistently, act7 mutant showed reduced cell division, cell elongation, and meristem length. Intracellular distribution and trafficking of auxin transport proteins in the actin mutants revealed that ACT7 specifically functions in root meristem to facilitate the trafficking of auxin efflux carriers PIN1 and PIN2, and consequently the transport of auxin. Compared with act7, act7act8 double mutant shows slightly enhanced phenotypic response and altered intracellular trafficking. The altered distribution of auxin in act7 and act7act8 affects the response of the roots to ethylene but not to cytokinin. Collectively, our results suggest that ACT7 dependent auxin-ethylene response plays a key role in controlling Arabidopsis root meristem development.

Abstract Background Parasite persistence, exacerbated and sustained immune response, and continuous oxidative stress have been described to contribute to the development of the cardiac manifestations in Chronic Chagas Disease. Nevertheless, there are no efficient therapies to resolve the Trypanosoma cruzi infection and prevent the disease progression. Interestingly, trypanocide, antioxidant, and immunodulatory properties have been reported separately for some major terpenes, as citral (neral plus geranial), limonene, and caryophyllene oxide, presents in essential oils (EO) extracted from two chemotypes (Citral and Carvone) of Lippia alba. The aim of this study was to obtain L. alba essential oil fractions enriched with the aforementioned bioactive terpenes and to evaluate the impact of these therapies on trypanocide, oxidative stress, mitochondrial bioenergetics, genotoxicity, and inflammatory markers on T. cruzi-infected macrophages. Methods T. cruzi-infected J774A.1 macrophage were treated with limonene-enriched (ACT1) and citral/caryophyllene oxide-enriched (ACT2) essential oils fractions derived from Carvone and Citral-L. alba chemotypes, respectively. Results ACT1 (IC50 = 45 ± 1.7 μg/mL) and ACT2 (IC50 = 80 ± 1.9 μg/mL) exhibit similar trypanocidal effects to Benznidazole (BZN) (IC50 = 48 ± 2.5 μg/mL), against amastigotes. Synergistic antiparasitic activity was observed when ACT1 was combined with BZN (∑FIC = 0.52 ± 0.13 μg/mL) or ACT2 (∑FIC = 0.46 ± 1.7 μg/mL). ACT1 also decreased the oxidative stress, mitochondrial metabolism, and genotoxicity of the therapies. The ACT1 + ACT2 and ACT1 + BZN experimental treatments reduced the pro-inflammatory cytokines (IFN-γ, IL-2, and TNF-α) and increased the anti-inflammatory cytokines (IL-4 and IL-10). Conclusion Due to its highly trypanocidal and immunomodulatory properties, ACT1 (whether alone or in combination with BZN or ACT2) represents a promising L. alba essential oil fraction for further studies in drug development towards the Chagas disease control.

Rationale: Imbalance in TGF-β signaling molecules SMAD2/3 and SMAD1/5/9 is a key driver of vascular dysfunction and pulmonary arterial hypertension (PAH) pathophysiology. KER-012, a novel ActRIIB ligand trap, designed to bind activin (Act) A and ActB to inhibit SMAD2/3: thereby permitting bone morphogenic protein (BMP) ligand signaling via SMAD1/5/9 to potentially rebalance TGF-β signaling and restore vascular homeostasis in PAH and associated disorders. Methods: Using hypoxia (1% O2) to mimic the pulmonary cell environment of PAH, we evaluated the effect of KER-012 or RKER -012 (a research form of KER-012) on TGF-β signaling in human pulmonary arterial endothelial cells (HPAECs) and smooth muscle cells (HPASMCs), two key cell types involved in the vascular remodeling in PAH. Gene expression of hypoxia/normoxia cultured cells was assessed by qPCR, phosphorylation of SMAD2 (pSMAD2) by alphaLISA, and ActA protein levels by ELISA. Results: HPAECs and HPASMCs both responded to TGF-β ligands implicated in PAH. ActA and Growth Differentiation Factor (GDF)11 increased SMAD2 phosphorylation and BMP9 increased SMAD1 phosphorylation (pSMAD1), confirming activation of the respective signaling pathways. KER-012 inhibited ActA and GDF11 induced pSMAD2, but not BMP9 induced pSMAD1. In hypoxic HPAECs, ActA gene expression increased >3-fold and BMPR2 expression decreased >2-fold relative to normoxia. ActA protein levels from hypoxic HPAEC supernatant also increased 10-fold, which was associated with an observed increase in pSMAD2 activation. While RKER-012 did not affect ActA gene expression, it did reduce ActA protein levels in supernatant to normoxic levels. This reduction in ActA may represent the fraction not bound to RKER-012, and potentially demonstrates a reduction in ActA availability for SMAD2/3 activation. Conclusions: ActA inhibition by RKER-012 restored homeostatic SMAD2 signaling in HPAECs and HPASMCs in an in vitr o PAH hypoxia model. RKER012 has also shown not to effect BMP signaling in these cells. These studies suggest that RKER-012 can potentially correct imbalanced SMAD signaling in PAH, partially via binding and inhibition of ActA protein.
