Die quenching of an AA2024 aluminum alloy billet was carried out using a servo press with ram motion control of WC-20mass%Co dies directly after solution heat treatment (SHT). The cylindrical billets were heated in an electric furnace and transferred to the press, then subjected to uniaxial compression with a reduction in height (Δh/h0) of 2%, 5% or 10% after SHT at 773 K or 823 K. The die quenching was successfully carried out without the precipitation hardening only on the billet after SHT at 823 K by sandwiching for 8 s. The subsequent artificial aging behavior at 463 K in an oil bath was investigated. It is found that the precipitation kinetics is accelerated in the die-quenched billets after SHT at 823 K. The peak hardness of the billet processed with 5% after SHT at 823 K is as high as that of the water-quenched billet, HV 149.0.

e15556 Background: Chemotherapy based on DNA-damaging anti-cancer drugs remains the most predominant treatment for advanced gastric cancer. The predictors for their sensitivity are under development. Our data showed that sperm-associated antigen 5 (Spag5) contributed to cancer cell proliferation and decreased DNA-damaging drug sensitivity in vivo and vitro. Methods: Spag5 mRNA expression were compared in The Cancer Genome Atlas (TCGA) and GEO datasets (GSE56807, GSE63089, GSE65801), while protein level were detected in 52 patients diagnosed with gastric cancer in Nanfang Hospital by immunohistochemistry. BGC-823 and MGC-803 were transfected with spag5 and control siRNA and then subjected to MTT and EDU assay to verify proliferation rate. Cell viability and apoptosis were tested in spag5-silenced BGC-823 and MGC-803 treated with oxaliplatin, epirubicin and 5-fluorouracil independently. DNA-damaged were detected by comet assay and yH2ax level with western blot. Treatment of oxaliplatin was given intraperitoneally to nude mice carrying xerographs of Shspag5 or control BGC-823 cells every three days. Results: Spag5 mRNA level was significantly upregulated in cancer tissues compared with paired normal tissues in all of the datasets. Immunohistochemistry shows higher protein level of spag5 in paired cancer tissue. Knockdown of Spag5 BGC-823 and MGC-803 showed a significantly decreased proliferation in both MTT and EDU assay. Spag5 silencing led to increase vulnerability to oxaliplatin, epirubicin and 5-fluorouracil, thus less cell viability and more apoptosis. Spag5 knockdown BGC-823 and MGC-803 subjected to DNA-damaging drugs showed upregulated yH2ax level and increased DNA damage in comet assay. Tumor growth rate were significantly reduced in shspag5 group and treatment of oxaliplatin led to relatively higher inhibition of tumor growth. Conclusions: Spag5 serves as a candidate to predict proliferation and chemosensitivity to DNA-damaging chemotherapy.

Five new meroterpenoids, zizhines P-S and U (1−4,7), together with two known meroterpenoids (5 and 6) were isolated from Ganoderma sinensis. Their structures including absolute configurations were assigned by using spectroscopic, computational, and chemical methods. Racemics zizhines P and Q were purified by HPLC on chiral phase. Biological evaluation found that 4, 5 and 6 are cytotoxic toward human cancer cells (A549, BGC-823, Kyse30) with IC50 values in the range of 63.43–80.83 μM towards A549, 59.2 ± 2.73 μM and 64.25 ± 0.37 μM towards BGC-823, 76.28 ± 1.93 μM and 85.42 ± 2.82 μM towards Kyse30.

The chlorination reaction of Y2O3 with ZrCl4 in LiCl-KCl eutectic melt was investigated by X-ray absorption fine structure (XAFS) technique. The chlorination reaction was observed between 773 K and 823 K as the 1st peak shift of the Fourier transform magnitude function |FT(k3χ(k))|. The peak corresponding to the nearest Y3+-Cl− correlation was observed in the XAFS analysis at 823 K as the result of the chlorination. It was confirmed that the mixture melts after the reaction is almost equivalent to a molten 5% YCl3-(LiCl-KCl eutectic) mixture.

Gastric carcinoma is a frequently detected malignancy worldwide, while its mainstream drugs usually result in some adverse reactions, including immunosuppression. λ-carrageenan oligosaccharides (COS) have attracted increasing attention as potential anticancer agents due to their ability to enhance immune function. Our current work assessed the antitumor mechanism of λ-COS using BGC-823 cells. Our findings indicated that λ-COS alone did not have a significant impact on BGC-823 cells in vitro; however, it was effective in inhibiting tumor growth in vivo. When THP-1 cells were pre-incubated with λ-COS and used to condition the medium, BGC-823 cells in vitro displayed a concentration-dependent induction of cell apoptosis, nuclear damage, and the collapse of mitochondrial transmembrane potential. These findings suggested that the antineoplastic effect of λ-COS was primarily due to its immunoenhancement property. Treatment with λ-COS was found to significantly enhance the phagocytic capability of macrophages, increase the secretion of TNF-α and IFN-γ, and improve the indexes of spleen and thymus in BALB/c mice. In addition, λ-COS was found to inhibit the growth of BGC-823-derived tumors in vitro by activating the Par-4 signaling pathway, which may be stimulated by the combination of TNF-α and IFN-γ. When used in combination with 5-FU, λ-COS demonstrated enhanced anti-gastric carcinoma activity and improved the immunosuppression induced by 5-FU alone. These findings suggested that λ-COS could be used as an immune-modulating agent for chemotherapy.

Their structures were mainly determined on the basis of MS, IR, 1D and 2D NMR spectral evidence. Compound 13-epi-toruolsol (17) showed cytotoxicities against BGC-823 and Hela cancer cell lines with IC50 values of 23.0 and 49:9 μM, and compound 3-epitriptobenzene B (19) showed cytotoxicities against BGC-823, Hela and A549 cancer cell lines with IC50 values of 19.1, 30.3 and 24:5 μM, respectively

In the present paper, on the basis of molecular hybridization, a series of 4,6-dihydrazone pyrimidine derivatives containing the pyridine moiety were synthesized, structurally characterized, and evaluated in vitro for their antitumor activity. According to the results, all the tested compounds demonstrated broad-spectrum antitumor activity against selected tumor cell lines (MCF-7, BGC-823, A549, and BEL-7402) and no obvious toxicity toward normal cells HL-7702. In particular, compounds 10a and 10f were found to be the most promising antitumor agents among the tested compounds against BGC-823 cells (IC50 = 9.00 μM and 7.89 μM) and BEL-7402 cells (IC50 = 6.70 μM and 7.66 μM), respectively. Compounds 10a and 10f exhibited higher potency against BGC-823 and BEL-7402 than the positive control 5-FU (IC50 = 15.18 μM and 15.81 μM). Further mechanism investigations demonstrated that compounds 10a and 10f could significantly increase the level of cellular ROS and induce early apoptosis of BGC-823 cells in a dose-dependent manner. Moreover, the DNA binding results from UV/Vis, CD spectroscopy, and molecular docking studies indicated that 10a and 10f bind with DNA via groove binding and partial intercalation. These results demonstrated that 10a and 10f may serve as novel lead compounds for the discovery of more dihydrazone pyrimidine derivatives with improved antitumor potency and selectivity.

Uppsatsens huvudsyfte är att undersöka om amerikanska och brittiska stridserfarenheter av kalibern 5,56 i Afghanistan kan bidra till att utveckla den svenska Försvarsmaktens taktiska förmåga. Anledningen till varför jag valde att utgå från dessa länder är därför att de har fler öppna källor samt har mer erfarenhet än många andra. Studien genomfördes genom en kvalitativ textanalys av bland annat reglementen, rapporter och artiklar. Uppsatsen visar att Försvarsmaktens kaliber till sin automatkarbin, 5,56, var potent för stridstaktikanpassad för invasionsförsvar. Detta delvis på grund av kaliberns egna logistiska fördelar men även därför att de prioriterade vapensystemen för infanteriförbanden, lätta pansarvärnsvapnen, innebar relativt korta stridsavstånd. Studien visar därefter att idag, för amerikanska och brittiska styrkor i dagens konflikt i Afghanistan, är 5,56 otillräcklig. Stridsavstånden är generellt för långa för att kalibern skall avge sin optimala verkan i målet. Studien belyser vad dessa nationer vidtagit för åtgärder, samt vilka åtgärder som planeras eller finns som förslag för att förbättra sin stridstaktiska förmåga. Den mest optimala förbättringen anser många är att byta ut kalibern 5,56 mot en grövre mellankaliber i spannet 5,56-7,62. Förslag som förändrad skjututbildning, optikutveckling och nya vapeninköp nämns också. I diskussionen diskuteras om eller hur den svenska Försvarsmakten kan dra nytta av de andra nationernas erfarenheter. Resonemanget ger det generella slutsatsen att Försvarsmakten kan utveckla sin stridstaktiska förmåga genom att studera andra nationer. Ett snabbt konkret exempel vore att implementera skarpskyttevapnet ak4 med förstorande optiskt hjälpmedel i svenska Försvarsmaktens skytteförband.


The main purpose with this study is to investigate whether U.S. and British combat experience of the caliber 5.56 in Afghanistan is helping to develop the Armed Forces' tactical ability. The reason why I chose to rely on these countries is because they have more unclassified sources and has more combat experience than many others. This study was conducted through a qualitative text analysis of for example regulations, reports and articles. This essay shows that the Swedish Armed Forces standard caliber that is being used in today’s assault rifle (ak5c), were potent in battle tactics designed for the defence of Sweden. This is partly becauseof 5,56 own logistical advantages and also because light anti-tank weapons had priority over small arms weapons. This resulted in relatively short shooting distances. The study shows thereafter that the U.S. and British troops in today's conflict in Afghanistan, thinks that 5.56 is insufficient. Combat distances are generally too long for the caliber to deliver its optimum effect in the target. The study highlights what kind of actions these nations have taken, and what measures are planned to improve its combat tactical ability. The study reveals that the most optimal improvement, are to replace the 5.56 with a bigger medium sized caliber. Proposals to change the marksmanship training, optics development and to purchase new weapons are also mentioned. In the end of this essay there is a discussion on, or how the Swedish Armed Forces can implement U.S and British experiences. The reasoning in the final chapter gives the general answer that the Swedish Armed Forces indeed can develop their tactical combat skills by studying other nations. An example would be to implement the older ak4 with magnifying optics that could improve the longer range capabilities of the infantry units.

Terpene hydrocarbons are high energy capacity hydrocarbons. The most known terpenoid biomass is Euphorbiaceae family. Euphorbia rigida, a member of Euphorbiaceae, was used as the biomass feedstock and natural zeolite was used as the catalyst in this study. In the experimental studies, firstly the raw material was analysed for its moisture, ash, volatile matter and fixed carbon. Then experiments were carried out in steam atmosphere in a fixed-bed reactor with a heating rate of 7 K/min, pyrolysis temperature of 823 K and mean particular size of 0.55 mm by mixing the catalyst to feedstock in different percentages. Experiments were performed with the catalyst ratios of 5, 10, 20 and 25 (weight-%) under steam atmosphere with the velocities of 12, 25 and 52 cm3/min to determine the effect of catalyst and steam on the product yields and bio-oil composition. Steam velocities were considered as the average steam velocities in the inlet tube of the reactor. The maximum bio-oil was reached to a value of 39.7% when using catalyst ratio of 20% and steam flow rate of 25 cm3/min. Pyrolysis oils were examined by using elemental analysis, IR and 1H-NMR spectroscopy. The liquid products were also fractionated by column chromatography and the gas chromatographic analysis of n-pentane eluate was performed.

We report new observations of a fast fluorescence process on 6p—6s transitions in xenon (0.01-5 Torr). A 600-ps pulse (λ~ 250 nm, Δν~ 3 GHz) from a frequency-doubled dye laser populates a single 6p state by two-photon coupling. The ensuing fluorescence (823 nm < λ < 993 nm) is collected by f/1 optics, detected by a photomultiplier or microchannel plate phototube, and displayed on an oscilloscope (1-GHz bandwidth) or digitized and recorded for statistical analysis. We previously characterized the emission process as amplified spontaneous emission (ASE) based on emission rate (> 10× spontaneous), high directionality, existence of threshold, and other time-independent characteristics.1 New time-resolved observations with the microchannel plate detector (500-ps overall rise time) showed that far above threshold the fluorescence pulses temporally reproduced the excitation pulses and occurred simultaneously. Pulses near threshold showed widths up to 3 ns and delays up to 3.5 ns. Ringing or multiple-pulse emission also occurred. When the emission was spontaneous, fluorescence energy yields showed the quadratic dependence expected of two-photon excitation and decreased monotonically when N2 was added to the xenon. ASE-dominated yields were linear in excitation energy and increased when N2 was introduced. ASE yields were smaller than predictions based on work of Raymond et al. 2 Mechanisms for these results are discussed.

Introdução: Dentre os marcadores de DNA baseados em PCR para estudos de diversidade, destaca-se o ISSR (Inter Simple Sequences Repeats), o qual apresenta vantagens como baixo custo relativo, alta reprodutibilidade e dispensa quanto aos conhecimentos prévios dos genomas. Objetivos: Estudar a otimização das condições de amplificação via PCR (gradientes de temperaturas de anelamento), envolvendo primers ISSR e os genomas de acessos de milhos crioulos (Zea mays L.) oriundos do Norte de Minas Gerais. Material e métodos: As extrações de DNAs seguiram um protocolo CTAB adaptado, utilizando-se folhas maduras de milhos crioulos. Para otimização das amplificações com primers ISSR (Coleção UCB, Canada), foram realizados testes de gradientes de temperaturas de anelamentos em termociclador configurado para o espectro de 45ºC a 65ºC, gerando 12 temperaturas dentro deste intervalo. As reações seguiram a seguinte programação: uma fase inicial de desnaturação a 94ºC por 5 min; seguida por 35 ciclos de [desnaturação (94ºC por 1 min), anelamento (45ºC a 65ºC por 1 min) e extensão (72ºC por 2 minutos)]; e uma fase de extensão final de 72ºC por 7 min / conservação a 4°C. Utilizou-se DNA's de dois indivíduos escolhidos em uma coleção de 44 acessos, variando-os a cada teste. Na placa de PCR (96 poços), testou-se quatro primers por reação, tendo as linhas como variantes de genótipos e primers; e colunas quanto às diferentes temperaturas. Resultados: Durante a execução do estudo, foram testados 56 primers ISSR, sendo que 34 deles caracterizam-se como sendo amplamente polimórficos, sob temperaturas de anelamento específicas identificadas nos gradientes: primers UCB 816, 820, 823, 828, 829, 836, 850, 880, 884 e 890 (46ºC); 808, 810, 812, 825, 827, 847, 849 e 855 (47ºC); 818 e 876 (48ºC); 848 (49ºC); 864, 865, 886, 887 e 889 (51ºC); 807, 830 e 888 (54ºC); 842, 856 e 866 (56ºC); 858 e 867 (59ºC). Quanto aos primers dispensados, 18 deles não amplificaram e quatro geraram apenas bandas monomórficas. Conclusão: A identificação prévia das temperaturas de anelamentos (PCR) que otimizem polimorfismos em uma população sob estudo, mostra-se como uma etapa imprescindível na otimização de tempo e insumos de Biologia Molecular.
